Abstract We have performed restriction endonuclease digestion of kinetoplast DNA mini-circles to characterize different stocks of two South American trypanosomes, Trypanosoma cruzi and Trypanosoma rangeli . The use of two fine cutter restriction enzymes (BspRI and MspI) that split the mini-circles in their variable region makes differentiation possible between all the parasites studied. Another restriction enzyme (TaqI) split the mini-circles to give a band with one-quarter of their molecular weight, which was the same in all stocks and clones from the two species studied, thus confirming the close relationship between T. cruzi and T. rangeli .
