To assess the effect of branched-chain amino acids (BCAA) supplementation on endurance performance in the heat, six women and seven men participated in two trials of rest in the heat (Ta = 34.4 +/- 1.8 degrees C; rh = 39 +/- 14%), followed by 40% VO2peak exercise to exhaustion. Subjects ingested 5 mL x kg(-1) of a placebo (PLAC) or BCAA drink every 30 min. Cycle time to exhaustion increased during BCAA (153.1 +/- 13.3 vs 137.0 +/- 12.2 min, P < 0.05) for men and women. Plasma glucose was maintained at baseline values for both drinks; however, women had significantly higher concentrations (5.9 +/- 0.6 vs 4.0 +/- 0.2 mM, P < 0.05). Plasma free fatty acids and ammonia were not influenced by drink or gender but increased over time. BCAA resulted in a significant (P < 0.05) increase in plasma BCAA (1209 +/- 119 vs 496 +/- 44 microM), while F-TRP (9.6 +/- 0.9 vs 12.0 +/- 1.3 microM) and F-TRP:BCAA were decreased (0.009 +/- 0.001 vs 0.024 +/- 0.003 ND) in both men and women. Cardiovascular and thermoregulatory data were similar between treatments for all subjects. Psychological data were not influenced by BCAA. These results indicate BCAA supplementation prolongs moderate exercise performance in the heat.

Potential physiological, anthropometric, and training determinants of running economy (RE) were studied in a heterogeneous group of habitual distance runners (N = 188, 119 males, 69 females). RE was measured as VO2 (ml.kg-1.min-1) during level treadmill running at 161 m.min-1 (6 mph) (VO2-6). Examined as potential determinants of RE were heart rate and ventilation while running at 6 mph (HR6, VE6), VO2max (ml.kg-1 x min-1), % fat, age, gender, height, weight, estimated leg mass, typical training pace, training volume, and sit-and-reach test performance. RE was entered as the dependent variable and the potential determinants as independent variables in zero-order correlation and multiple regression analyses. Zero-order correlation analysis found VO2max, HR6, and VE6 to be significantly, positively correlated with VO2-6 (P < 0.001). Multiple regression analysis, in which the independent effect of each predictor variable was examined, revealed VO2-6 to be positively correlated with VO2max (P < 0.001), HR6 (P < 0.001), VE6 (P < 0.001), and age (P < 0.05) and negatively correlated with weight (P < 0.01). These findings indicate that, in a diverse group of runners, better RE (VO2-6) is associated with lower VO2max, lower submaximal exercise VE and HR, lower age, and greater weight.

ABSTRACT Despite agreement that methadone maintenance treatment (MMT) is an effective and safe option for treatment of heroin dependence, there have been controversies about its effect on heroin craving. A systematic literature review of the PubMed database was used to find studies eligible for inclusion in the study. The authors present the results of 16 articles that met all inclusion criteria. Overall, 7 studies reported that methadone could reduce heroin craving, 4 studies reported that patients in MMT are still at risk of having heroin craving, 1 study reported that methadone could increase heroin craving, and 4 studies reported that methadone has a neutral effect on heroin craving. One may speculate from these data that methadone may help with heroin craving, but patients in MMT may still be at risk of cue-induced heroin cravings. Methadone provides a helpful tool for reducing some components of craving and risk of relapse for patient receiving MMT.

Background: Severe physical and environmental stress seems to have a suppressive effect on the hypothalamic–pituitary–gonadal (HPG) axis in men. Examining hormonal responses to an extreme 160-km competition across frozen Alaska provides a unique opportunity to study this intense stress. Objective: To examine hormonal responses to an ultra-endurance race. Methods: Blood samples were obtained from 16 men before and after racing and analyzed for testosterone, interleukin-6 (IL-6), growth hormone (GH) and cortisol. Six subjects (mean (SD) age 42 (7) years; body mass 78.9 (7.1) kg; height 1.78 (0.05) m raced by bicycle (cyclists) and 10 subjects (age 35 (9) years; body mass 77.9 (10.6) kg; height, 1.82 (0.05) m) raced by foot (runners). Mean (SD) finish times were 21.83 (6.27) and 33.98 (6.12) h, respectively. Results: In cyclists there were significant (p⩽0.05) mean (SD) pre-race to post-race increases in cortisol (254.83 (135.26) to 535.99 (232.22) nmol/l), GH (0.12 (0.23) to 3.21 (3.33) µg/ml) and IL-6 (2.36 (0.42) to 10.15 (3.28) pg/ml), and a significant decrease in testosterone (13.81 (3.19) to 5.59 (3.74) nmol/l). Similarly, in runners there were significant pre-race to post-race increases in cortisol (142.09 (50.74) to 452.21 (163.40) ng/ml), GH (0.12 (0.23) to 3.21 (3.33) µg/ml) and IL-6 (2.42 (0.68) to 12.25 (1.78) pg/ml), and a significant decrease in testosterone (12.32 (4.47) to 6.96 (3.19) nmol/l). There were no significant differences in the hormonal levels between cyclists and runners (p>0.05). Conclusions: These data suggest a suppression of the hypopituitary–gonadal axis potentially mediated by amplification of adrenal stress responses to such an ultra-endurance race in environmentally stressful conditions.

Protease supplementation has been shown to attenuate soft tissue injury resulting from intense exercise. The aim of this study was to evaluate the effects of protease supplementation on muscle soreness and contractile performance after downhill running. Ten matched pairs of male participants ran at a −10% grade for 30 min at 80% of their predicted maximal heart rate. The participants consumed two protease tablets (325 mg pancreatic enzymes, 75 mg trypsin, 50 mg papain, 50 mg bromelain, 10 mg amylase, 10 mg lipase, 10 mg lysozyme, 2 mg chymotrypisn) or a placebo four times a day beginning 1 day before exercise and lasting a total of 4 days. The participants were evaluated for perceived muscle soreness of the front and back of the dominant leg, pressure pain threshold by dolorimetry of the anterior medial, anterior lateral, posterior medial and posterior lateral quadrants of the thigh, and knee extension/flexion torque and power. The experimental group demonstrated superior recovery of contractile function and diminished effects of delayed-onset muscle soreness after downhill running when compared with the placebo group. Our results indicate that protease supplementation may attenuate muscle soreness after downhill running. Protease supplementation may also facilitate muscle healing and allow for faster restoration of contractile function after intense exercise.

Exercise is known to have beneficial effects on cognitive function. This effect is greatly favored by an exercise-induced increase in neurotrophic factors, such as brain-derived neurotrophic factor (BDNF) and insulin-like growth factor-1 (IGF-1), especially with high-intensity exercises (HIE). As a complication of type 1 diabetes (T1D), a cognitive decline may occur, mostly ascribed to hypoglycaemia and chronic hyperglycaemia. Therefore, the purpose of this study was to examine the effects of acute HIE on cognitive function and neurotrophins in T1D and matched controls. Ten trained T1D (8 males, 2 females) participants and their matched (by age, sex, fitness level) controls were evaluated on 2 occasions after familiarization: a maximal test to exhaustion and an HIE bout (10 intervals of 60 s at 90% of their maximal wattage followed by 60 s at 50 W). Cognitive tests and analyses of serum BDNF, IGF-1, and free insulin were performed before and after HIE and following 30 min of recovery. At baseline, cognitive performance was better in the controls compared with the T1D participants (p < 0.05). After exercise, no significant differences in cognitive performance were detected. BDNF levels were significantly higher and IGF-1 levels were significantly lower in T1D compared with the control group (p < 0.05) at all time points. Exercise increased BDNF and IGF-1 levels in a comparable percentage in both groups (p < 0.05). In conclusion, although resting levels of serum BDNF and IGF-1 were altered by T1D, comparable increasing effects on BDNF and IGF-1 in T1D and healthy participants were found. Therefore, regularly repeating acute HIE could be a promising strategy for brain health in T1D.

J Strength Cond Res 29(3): 678–684, 2015—Many athletes avoid using mouthguards because they believe that they impair their ability to breath and negatively affect performance. Recently, some manufacturers have developed “vented” mouthguards (VentMGs) to address this concern. The purposes of this investigation were to describe the impact of a commercially available “vented” boil-and-bite mouthguard on the physiological responses to graded exercise and to determine whether the use of the same mouthguard influences performance during traditional physical agility tests. Recreationally trained males (n = 15) (age = 24 ± 1 year; = 43.5 ± 1.9 ml·kg−1·min−1; body mass index = 25.2 ± 0.9) completed 3 randomly assigned trials where they wore no mouthguard (control), a traditional mouthguard (TradMG), or a VentMG. During each trial, subjects completed a modified maximal exercise test on a cycle ergometer and a series of physical agility tests (40-m dash, vertical leap, broad jump, 3-cone drill, and shuttle run). No differences were seen between control and the TradMG in any cardiorespiratory measures at any time during the maximal exercise test. Ventilation and blood lactate were lower (p ⩽ 0.05) during VentMG at 200 W and at MAX; however, no differences in were observed. Although TradMG had no impact on physical agility, VentMG produced a higher (1.9 cm; p = 0.03) vertical leap than control. Both mouthguard conditions negatively affected perceptions of breathability, comfort, and ability to communicate, but no differences existed between the 2 conditions. These findings confirm that TradMG has no negative impact on physiological function during exercise and physical agility; however, VentMG may have a positive impact at higher workload and on vertical leap.

Purpose Cycling desks as a means to reduce sedentary time in the office has gained interest as excessive sitting has been associated with several health risks. However, the question rises if people will still be as efficient in performing their desk-based office work when combining this with stationary cycling. Therefore, the effect of cycling at 30% Wmax on typing, cognitive performance and brain activity was investigated. Methods After two familiarisation sessions, 23 participants performed a test battery [typing test, Rey auditory verbal learning test (RAVLT), Stroop test and Rosvold continuous performance test (RCPT)] with electroencephalography recording while cycling and sitting on a conventional chair. Results Typing performance, performance on the RAVLT and accuracy on the Stroop test and the RCPT did not differ between conditions. Reaction times on the Stroop test and the RCPT were shorter while cycling relative to sitting (p < 0.05). N200, P300, N450 and conflict SP latency and amplitude on the Stroop test and N200 and P300 on the RCPT did not differ between conditions. Conclusions This study showed that typing performance and short-term memory are not deteriorated when people cycle at 30% Wmax. Furthermore, cycling had a positive effect on response speed across tasks requiring variable amounts of attention and inhibition.

Type 1 diabetes (T1D) can have a significant impact on brain function, mostly ascribed to episodes of hypoglycemia and chronic hyperglycemia. Exercise has positive effects on acute and chronic glycemic control in T1D, and has beneficial effects on cognitive function by increasing neurotrophins such as BDNF and IGF-I in non-diabetic humans. The present study examines the effects of different types of exercise intensities on neurotrophins in T1D. 10 participants with type 1 diabetes were evaluated in 3 sessions: high-intensity exercise (10×[60 s 90%Wmax, 60 s 50 W]), continuous exercise (22 min, 70% VO2 max) and a control session. Blood glucose, serum free insulin, serum BDNF and IGF-I were assessed pre/post all the trials and after recovery. Blood glucose significantly decreased after both exercise intensities and BDNF levels increased, with a dose-response effect for exercise intensity on BDNF. IGF-I changed over time, but without a difference between the different exercise protocols. Both exercise intensities change neurotrophins in T1D, but also exhibit a dose response effect for BDNF. The intensity-dependent findings may aid in designing exercise prescriptions for maintaining or improving neurological health in T1D, but both types of exercise can be implemented.

Objective: The aim of this study was to examine the longitudinal effect of implementing bike desks in an office setting on physical health, cognition, and work parameters. Methods: Physical health, cognitive function, work engagement, and work performance measured before (T0) and after (T2) the intervention period were compared between office workers who used the bike desk (IG, n = 22) and those who did not (CG, n = 16). Results: The IG cycled approximately 98 minutes/week. The IG showed a significantly lower fat percentage and a trend toward a higher work engagement at T2 relative to T0, while this was not different for the CG. No effects on other parameters of health, cognition, or work performance were found. Conclusions: Providing bike desks in the office positively influences employees’ fat percentage and could positively influence work engagement without compromising work performance.
