Excess use of chemical fertilizer causes soil acidification and accumulation of salt, and thus might bring to desertification of soil. To overcome this problem, it needs limited usage of chemical fertilizer and increased usage of natural fertilizer as an alternative. In this study, dry soil-borne Bacillus sp. SHL-3, which was isolated from arid and barren soil, with plant growth promoting activity was isolated for identification and to determine optimal culture condition. A bacterial strain SHL-3 had the IAA productivity (5.16 ± 0.10 mg L -1 ), ACC deaminase activity (0.36±0.09 at 51 hours) and siderophore synthesis. It was identified as genus Bacillus sp.. Also, optimal culture condition of SHL-3 were 20°C and pH 7 in LB medium. Bacillus sp. SHL-3 had up to 4% salt tolerance in the medium. We evaluated the plant growth promotion ability of SHL-3 using yam (Dioscorea japonica Thunb.). As a result, Bacillus sp. SHL-3 was effective on the increase of the shoot length (202.4% increase for 91 days). These results indicate that Bacillus sp. SHL-3 can serve as a promising microbial resource for the biofertilizers of soil.

These studies were attempted to investigate the change of microbial community of anode of microbial fuel cell using swine wastewater in the enrichment step with the lapse of time. Microbial fuel cells enriched by a 1 : 1 mixture of anaerobic digestive juices of the sewage treatment plant and livestock wastewater. Enrichment culture step was divided into three stages to indentify the microorganisms. It was separated by each lag phase, exponential phase, and stationary phase. These steps were determined by the change of the current value. The current after enrichment was generated about 0.84 ± 0.06 mA. We were cut out the different 17 bands in the DGGE fingerprint gel to do sequencing. The bands which the concentration was increasing or newly appearing with the lapse of time were included for this study. In the lag and exponential phase, Clostridium, Rhodocyclaceae, Bacteriodetes, and Uncultured bacterium etc. were detected. There were in the stationary phase Geobacter sp., Rhodocyclaceae, Candidatus, Nitrospira, Flavobactriaceae and uncultured bacterium etc. Geobactor among microorganisms detected in this study is known as the Electrochemically active microorganisms. It may include electrochemically active microorganisms to be considered as electrical activity microorganisms.

Coastal sand dunes have been seriously damaged caused by the development thoughtless for the environment and coastal erosion and destruction due to artificial structures like coast roads and breakwater. Hereupon, in this study we made a library of rhizobacteria that have the plant growth-promoting ability for plant rhizosphere of indigenous plants inhabiting in a coastal sand dune as well as the strong tolerance to salt, and evaluated the plant growth-promoting ability of these strains. Furthermore, we evaluated the effect of rhizobacteria on the growth rate of saline tolerant plants in sandy soil; selected out the most useful microorganism for the restoration of a damaged sand dune. The effect of inoculation of strains selected from the first experiment on the growth of Peucedanum japonicum and Arundo donaxes planted in a coastal sand dune was evaluated. As a result, Bacillus aerius MH1RS1 had plant growth promoting activities: indole acetic acid (IAA) production, siderophores and 1-aminocyclopropane-1carboxylate deaminase (ACC deaminase) activity, and also had a salinity tolerance. Also, in case of Peucedanum japonicum, the length of stems and weights of roots were enhanced by the inoculation of B. aerius MH1RS1. Fresh weights of stems and weights of roots in experimental group were, in particular, increased by 25% comparing with the control group. For an Arundo donax in experimental group, plant length increased by 18%, and weight of roots by 20% which is significant.
