Effect of Buffer Components on the Determination of Hydrogen Peroxide using Fluorescein Chemiluminescence Reaction.

ペルオキシダーゼが触媒として働くフルオレセィン化学発光反応を利用した過酸化水素の定量におけるpH緩衝剤の効果を,クエン酸,リン酸,イミダゾールおよびmモルホリノ-1-プロパンスルポソ酸(MOPS)を用いて比較検討した。その結果,クエン酸リン酸およびイミダゾールを用いた場合がMOPSを用いた場合より発光量が大きかった。そのため,過酸化水素の検出感度はクエン酸を用いた場合,MOPSを用いた場合に比べて2.5倍向上した。また,クエン酸およびリン酸を用いた場合,そのマスキング効果のために遷移金属イオンの許容濃度が増大することを明らかにした。さらに,本化学発光反応の発光機構について検討を行い,フルオレセインが一重項酸素の生成に関与するとともに,一重項酸素からのエネルギー受容体としても働いて励起状態となり,その励起種が基底状態に戻るときに光を放出する機構を提案した。

Peroxyoxalate Chemiluminescent Assay of Ascorbic Acid Based on Autoxidation with Oxygen in Reversed Micelle

A highly sensitive chemiluminescence (CL) method was developed for the determination of ascorbic acid (AA) based on the conversion of AA to hydrogen peroxide (H2O2) in a cationic reverse micellar hexadecyltrimethylammonium chloride : chloroform medium. AA was indirectly determined by measuring the H2O2 generated by the peroxyoxalate CL method. The CL intensity was linearly correlated with AA concentration over the range from 5.0 × 10−8 to 1.0 × 10−6 M.
