Studies on the Significance of Oxidase-Reaction and Fat-Stain in Leukocytes*

I That the granules of leukocytes show positive stable oxidase reaction is commonly known. Furthermore it has been reported by SEHRT (1927),(’) GOLDMANN ( 1929) ,(*) ARAKI (1935) ,(3) HAYAMI (1936) ,(4) RHEINGOLD and WISLOCKI ( 1948),(5) BLOOM and WISLOCKI (1950) and others that these granules give positive results to fat-staining. Concerning the relation between these two reactions DIETRICH (1908) (’) and SEHRT (1931)@) discussed that oxidase reaction is not based on ferment action, but should be considered as a lipid reaction. In most of the studies the authors have the opinion that these two reactions are not connected with each other at all. In other words, oxidase reaction indicates the presence of indophenolase, while lipid reaction has nothing to do with this ferment. This opinion has been easily accepted by the majority of the authors presupposing that positive results of fat stain indicate the existence of fatty substances without regarding its staining mechanism. Therefore, we(931OJ1J2) attempted to criticize the relation of both reactions again without giving attention to their customary significance.
