A cross sectional study was conducted in and around Asella town from November 2007 to April 2008 on dairy cows to determine the prevalence of mastitis, impact of risk factors and isolate the dominant mastitis causing bacteria on total of 223 lactating cows, of which 92 were indigenous Arsi, and 131 Holstein Zebu cross by using clinical examination and California mastitis test (CMT). Of these 144 (65.6 %) were positive by clinical examination and CMT for clinical and sub clinical mastitis, with prevalence of 26.5 % and 38 %, respectively. There was a significant difference (P < 0.05) on the prevalence of mastitis between cows kept under different hygiene of milking process. Similarly a significant difference on the prevalence of mastitis between the two breeds (P < 0.05) was also observed. From 144 CMT and clinically positive milk samples analyzed microbiologically, 133 were culturally positive for known mastitis pathogens and while 11 were negative. The dominant bacterial isolates in the study animals were Staphylococcus species (41.4 %), Streptococcus species (24.8 %), and other Gram positive rods and Gram negative enteric bacteria (33.8 %). Good hygiene in milking process, milking clinically infected cows at last, culling chronic mastitis carriers, treating clinically infected cows and dry period therapy could reduce the prevalence of contagious mastitis in the study area.

A cross-sectional study of brucellosis in small ruminants was carried out from October 2008 to March 2009 in Jijiga District, Somali Regional State of Ethiopia. Seven hundred thirty sera samples (421 of sheep and 309 of goats) were randomly collected from purposively selected villages of the study area. Structured questionnaire format was developed, pre-tested and administered to assess the perception of the community pertaining to brucellosis in sheep and goats. Sera samples were screened by Rose Bengal Plate Test (RBPT), and all samples tested positive by the RBPT were subjected to Complement Fixation Test (CFT) for confirmation. Of 12 serum samples that were positive by RBPT, 11 were positive by CFT. Statistically significant differences were not observed between the species as well as the sex groups (P > 0.05); however, the variation between the age groups was statistically significant (P < 0.05). Analysis of the questionnaire survey suggests that improper handling of aborted materials, consumption of raw milk, and lack of awareness about the disease, among others, might greatly contribute to further spread of brucellosis in their livestock and exposes the community to a public health hazard. In general, the sero-prevalence in the study area was not so high; nevertheless, appropriate brucellosis control and prevention methods should be implemented to circumvent future potential for economic losses and the public health hazard of the disease.

Head and clothing lice from Jimma, Ethiopia were investigated for pathogenic bacteria. Genomic DNA from pools of lice was subjected to PCR analysis for Bartonella spp., Borrelia spp. Coxiella burnetii, Rickettsia spp. and Yersinia pestis. All 102 lice pools were negative for the afore mentioned pathogens, with the exception of Bartonella species found among 6 of 65 (9.2%) head lice pools and1 of 33 clothing lice pools. Identification was achieved by sequencing the ribosomal intragenic transcribed spacer region (ITS), revealing all to be Bartonella quintana. Although established as a clothing louse-borne infection, typically causing chronic bacteraemia, trench fever, bacillary angiomatosis and endocarditis, this has only been rarely reported among head lice. The higher numbers of infected head lice pools compared with clothing lice suggests their competence for maintaining this infection within Ethiopia.

Two regions (Jimma and Dire Dawa) in Ethiopia were investigated for the presence of soft ticks. Although no Ornithodoros spp. ticks were collected during this survey, published records of their existence in Ethiopia were found. An overwhelming infestation of Argas persicus was revealed in a village located adjacent to Dire Dawa. These ticks primarily were feeding on poultry, but were also biting humans. Furthermore, hard ticks were collected from livestock and companion animals in these regions. Collected ticks were assessed for Borrelia by real-time PCR followed by conventional PCR and sequencing to identify species present. A. persicus ticks were found to carry B. anserina in 3 of 40 (7.5%) A. persicus tick pools, whilst hard tick pools yielded 2 of 16 (12.5%) positive for B. theileri. Collectively, these borrelial species and their tick vectors are likely to have an important economic impact of particular relevance to subsistence farmers in Ethiopia.

Ethiopian soft ticks Argas persicus, hard ticks including both Amblyomma variegatum and Rhipicephalus (Boophilus) spp., and fleas were collected from livestock, traditional human dwellings, and cracks and crevices of trees. They were assessed in pools for the presence of Rickettsia using PCR-based methods. The extracted tick DNA was subjected to molecular screening for Rickettsia, which revealed 50.5% of the pooled samples to be positive for Rickettsia spp. These were then subjected to multi-gene analysis using both outer surface proteins and housekeeping genes with proven discriminatory potential. Sequencing of the citrate synthase and outer membrane genes clearly led to the identification of three distinct rickettsial species, Candidatus Rickettsia hoogstraalii in Argas persicus ticks; R. africae in hard tick pools, and R. felis in fleas. Furthermore, we demonstrated the presence of the plasmid-borne small heat-shock protein gene hsp2 in DNA from A. persicus ticks suggesting that Candidatus R. hoogstraalii carried by these ticks possess a plasmid. Unlike chromosomal gene sequences, the hsp2 gene failed to cluster with Candidatus R. hoogstraalii, instead falling into an isolated separate clade, suggesting a different origin for the plasmid.

Lumpy skin disease (LSD) is one of the major livestock disease problems in most areas of Ethiopia. A questionnaire-based cross-sectional study was conducted from October 2011 to February 2012 in four selected districts of Afar and Tigray regions to estimate the herd-level prevalence of LSD, and to assess its associated risk factors. Herd-owners were selected based on the willingness to provide information to complete the questionnaire. A total of 393 questionnaires were collected. Out of 393 herd-owners, 173 reported having LSD in their herds, giving an estimated herd- and animal-level prevalence of (44%, 95% CI: 37-50%) and (7.4%, 95% CI: 6-8%), respectively. Herd prevalence between regions and among the districts were significantly different (χ(2)=8, P<0.01 and χ(2)=9.9, P<0.01), respectively. The risk factors of LSD occurrence were introduction of a new animal to the herd, herd size, and utilization of communal grazing and watering points. These management characteristics cannot be readily changed in the studied area, hence, disease control should rely on a greater use of effective LSD vaccines.

The prevalence of subclinical mastitis and associated risk factors at the herd, cow and quarter level were studied using multilevel modelling in smallholder dairy farms in Jimma, Ethiopia. Forty-two herds, out of the 55 dairy farms located in Jimma (76%), were visited, a questionnaire was performed, and 635 quarters belonging to 176 lactating cows were screened to detect the presence of subclinical mastitis using the California Mastitis Test (CMT). Sixty-two percent of the cows and 51% of the quarters were subclinically infected. Quarters from cows in later stage of lactation (>180 DIM) [opposed to early lactation (<90 DIM)] were more likely to be subclinically infected (OR=2.40, 95% CI=1.44-3.99). Overall, quarters from cows milked by squeezing (as opposed to stripping) were less likely to be subclinically infected (OR=0.45, 95% CI=0.29-0.71), although quarters from cows with tick-infested udders were more likely to be subclinically infected when milked by squeezing (as opposed to stripping). The milking technique did not influence the likelihood of infection in cows without ticks on the udder. This study stresses the high prevalence of subclinical mastitis in smallholder dairy farms in Jimma and a lack of awareness of the existence of the disease among dairy farmers. Implementation of a mastitis prevention program adapted to the local needs, including a focus on milking technique, application of appropriate tick control measurements as well as fertility management, allowing cows to be dried-off at a more appropriate moment, are needed. To conclude, milking by squeezing instead of stripping, but not of cows with tick-infested udders, as well as fertility management could reduce the subclinical mastitis prevalence (and incidence) on the short term.

The amount of trace elements present in edible bovine tissues is of importance for both animal health and human nutrition. This study presents data on trace element concentrations in semitendinosus and cardiac muscles, livers and kidneys of 60 zebu (Bos indicus) bulls, sampled at Jimma, Ethiopia. From 28 of these bulls, blood samples were also obtained. Deficient levels of copper were found in plasma, livers, kidneys and semitendinosus muscles. Suboptimal selenium concentrations were found in plasma and semitendinosus muscles. Semitendinosus muscles contained high iron concentrations. Trace elements were mainly stored in the liver, except for iron and selenium. Cardiac muscles generally contained higher concentrations of trace elements than semitendinous muscles except for zinc. A strong association was found between liver and kidney concentrations of copper, iron, cobalt and molybdenum. Liver storage was well correlated with storage in semitendinosus muscle for selenium and with cardiac muscle for cobalt and selenium. Plasma concentrations of copper, selenium, cobalt were well related with their respective liver concentrations and for cobalt and selenium, also with cardiac muscle concentrations. The data suggest multiple trace element deficiencies in zebu cattle in South-West Ethiopia, with lowered tissue concentrations as a consequence. Based on the comparison of our data with other literature, trace element concentrations in selected edible tissues of Bos indicus seem quite similar to those in Bos taurus. However, tissue threshold values for deficiency in Bos taurus cattle need to be refined and their applicability for Bos indicus cattle needs to be evaluated.

A cross-sectional study on clinical mastitis, intramammary infection (IMI) and blind quarters was conducted on 50 smallholder dairy farms in Jimma, Ethiopia. A questionnaire was performed, and quarters of 211 cows were sampled and bacteriologically cultured. Risk factors at the herd, cow, and quarter level for clinical mastitis and (pathogen-specific) intramammary infection were studied using multilevel modeling. As well, factors associated with quarters being blind were studied. Eleven percent of the cows and 4% of the quarters had clinical mastitis whereas 85% of the cows and 51% of the quarters were infected. Eighteen percent of the cows had one or more blind quarter(s), whereas 6% of the quarters was blind. Non-aureus staphylococci were the most frequently isolated pathogens in both clinical mastitis cases and IMI. The odds of clinical mastitis was lower in herds where heifers were purchased in the last year [odds ratio (OR) with 95% confidence interval: 0.11 (0.01-0.90)], old cows (>4 years) [OR: 0.45 (0.18-1.14)], and quarters not showing teat injury [OR: 0.23 (0.07-0.77)]. The odds of IMI caused by any pathogen was higher in herds not practicing teat drying before milking (opposed to drying teats with 1 towel per cow) [OR: 1.68 (1.05-2.69)], cows in later lactation (>180 DIM opposed to ≤90 DIM) [OR: 1.81 (1.14-2.88)], cows with a high (>3) body condition score (BCS) [OR: 1.57 (1.06-2.31)], right quarters (opposed to a left quarter position) [OR: 1.47 (1.10-1.98)], and quarters showing teat injury [OR: 2.30 (0.97-5.43)]. Quarters of cows in herds practicing bucket-fed calf feeding (opposed to suckling) had higher odds of IMI caused by Staphylococcus aureus [OR: 6.05 (1.31-27.90)]. Except for BCS, IMI caused by non-aureus staphylococci was associated with the same risk factors as IMI caused by any pathogen. No access to feed and water immediately after milking [OR: 2.41 (1.26-4.60)], higher parity [OR: 3.60 (1.20-10.82)] and tick infestation [OR: 2.42 (1.02-5.71)] were risk factors for quarters being blind. In conclusion, replacement of old cows, prevention of teat injuries/lesions, drying teats with 1 towel per cow before milking, improving fertility in order to shorten the lactation period, allowing (restricted) suckling, access to feed and water immediately after milking, and improving tick control could improve udder health in Jimma.

Four studies were performed to quantify milk production, quality and consumption in the town Jimma, Ethiopia. First, 47 dairy farmers and 44 milk retailers were interviewed to gain more insights in dairy farming and marketing, and associated constraints. Second, bulk milk samples (n=188) were collected for 4 consecutive weeks to investigate milk quality [Total Bacterial Counts (TBC), Coliform Counts (CC), Somatic Cell Counts (SCC), and antimicrobial residues]. Third, (bulk) milk samples from 32 farms, 46 milk retailers and the 3 local milk collection centers were collected to determine the presence of oxacillin susceptible-and oxacillin resistant Staphylococcus aureus. Fourth, 208 adult inhabitants were interviewed to gain more insight in milk consumption and associated concerns of consumers. The average dairy farm included in the studies consisted of 5 lactating cows, produced 43 liters of milk per day and was owned by male, literate adults. Milk was sold to retailers (71% of the production) and directly to customers (25%) without any quality control, whereas 4% was self-consumed. Shortage of animal nutrition and adulteration of the milk were the main constraints for farmers and retailers, respectively. The median TBC, CC and SCC were 122,500CFU/mL, 1,005CFU/mL and 609,500cells/mL, respectively. Antimicrobial residues were detected in 20% of all samples. In general, the milk quality was considered to be poor (TBC>10,000CFU/mL, and/or CC>100CFU/mL, and/or SCC>400,000cells/mL and/or presence of antimicrobial residues) in 97% of all samples. S. aureus was isolated from 12 (38%), 13 (33%), and 2 out of 3 of the milk samples originating from the dairy farms, the milk retailers, and the milk collection centers, respectively. Seven (26%) of the isolates were resistant to oxacillin suggesting the presence of MRSA (Lee, 2003). Local milk is occasionally consumed by adults but more frequently by children. Adults mainly drink spontaneously fermented milk (57% of 105 interviewees consuming local milk) whereas most milk for children is boiled (86% of 110 households with children consuming local milk). Most consumers are concerned about adulteration and milk borne diseases but not about antimicrobial residues. Educated consumers (secondary school or higher) were more likely to boil milk for own consumption, to be concerned about antimicrobial residues in milk, to be concerned about milk borne diseases and to be willing to pay more for milk with proven good quality compared to poorly educated consumers. We conclude that milk quality incentives should be introduced in Jimma, and investments should be made in knowledge transfer, training, milk collection systems and a central milk quality lab.
