SummaryThe expression of three heat shock proteins (HSPs)-HSP90α, HSP70, HSP27 in cells obtained from 22 patients with leukemia, K562 erythroleukemia cell line, and normal blood cells was observed by means of RNA dot blot analysis. The results showed that the expression of the HSP27 gene was enhanced in 4 cases of acute lymphoid leukemia (ALL), 7 cases of acute nonlymphoid leukemia (ANLL) and 2 cases of myelodysplastic syndrome. (MDS) as compared with that of the normal blood Cells, yet there was no significant difference in the HSP27 expression between the ALL and ANLL cells. The expression of HSP70 in all the 5 ALL and ANLL patients was much lower than that of the normal subjects, except 1 case of ALL and 1 case of MDS, in which the expression was obviously enhanced. All the cases including 11 ANLL, 5 ALL and 1 MDS had higher HSP90α expression than the normal subjects. The enhanced expression pf HSP90α in leukemia cells may be associated with the active and indefinite proliferation of leukemia cells. Our results also suggest that the high expression of the HSP27 gene may not be confined to a specific type of acute leukemia.

ZusammenfassungDie Untersuchung von 28 Hämophilie-Kranken mittels Zweitgeneration-Anti HCV-Reagens (Kehua Biotechnische Gesellschaft. Shanghai) ergab eine Infektionsrate von 78 %, und bei Patienten mit mehrmaliger Transfusionsbehandlung war sie noch höher. Bei 15 Patienten aus dieser Gruppe wurden auch die zugehörigen 10 Familien untersucht. 12 Patienten erwiesen sich als Anti-HCV positiv, 53 krankheitsfreie Familienangehörige als Anti-HCV negativ. Bei 8 Kindern und 9 Ehefrauen wurde kein positiver Anti-HCV Befund festgestellt. Das Ergebnis weist darauf hin, daß bei Hämophilie-Patienten mit mehrmaliger Therapie durch Bluttransfusionen und Präparate von Gerinnungsfaktoren die Gefahr einer HCV-Infektion sehr groß ist, die Gefahr einer HCV-Infektion infolge von Kontakten mit Familienangehörigen jedoch sehr gering.Abstract28 cases of hemophilia were examined for HCV infection status by using the Kehua anti-HCV ELISA kit of second generation. In this study peripheral blood mononuclear cells were tested for T lymphocyte subpopulations, interleukin-2 receptor (IL-2R) and HLA-DR antigen by monoclonal antibodies in 14 LN patients. In comparison with normal controls, 14 LN patients showed a decrease of T4 lymphocyte (0.01 < P < 0.05) and increase of T8 cells (P < 0.01) with reduction of T4/T8 ratio (P < 0.01). The expression of HLA-DR cells increased (P < 0.01), the expression of IL-2R activated by PHA significantly decreased (P < 0.01), while nonactivated IL-2R expression significantly increased (P < 0.01). T4/T8 ratio was significantly correlated with activated IL-2R expression (P < 0.01, r = 0.6599). The results indicated that there was a immunological defect in LN patients. The pathogenesis of LN might be heterogeneous.ZusammenfassungBei Lupusnephritis (LN) handelt es sichum eine Dysfunktion des Immunoregulationssystems. Die Pathogenese ist heute noch nicht geklärt. In unserer Studie wurden bei 14 Patienten mit LN in mononukleären Zellen des peripheren Blutes mittels monoklonaler Antikörper T Lymphozytensubpopulationen, lnterleukin-2 Rezeptor (IL-2R) und HLA-DR Antigen untersucht. Beim Vergleich mit normalen Probanden ließ sicn bei den Patienten mit LN eine signifikante Verminderung sowohl des prozentualen Anteils von T4-Lymphozyten (0,01<P<0,05) als auch der Ratio von T4/T8-Lymphozyten (P<0,01) feststellen. Die prozentualen Anteile von T8-Lymphozyten und die HLA-DR Expression zeigten bei den Patienten mit LN die Tendenz zur Erhöhung (P<0, 01). Die IL-2R Expression der mit PHA stimulierten T Zellen war bei Patienten mit LN geringer als bei Gesunden (P< 0, 01), und die IL-2R Expression der T Zellen ohne Stimulation mit PHA war gegenüber den Normalwerten erhöht (P<0, 01). Es zeigte sich eine signifikante Korrelation zwischen der T4/T8-Rate und der IL-2R Expression nach PHA-Stimulation (P<0, 01, r=0,6599). SummaryWe used A chain extracted from urine of patient with myeloma (IgD λ) as antigen for immunizing BALB/C mice, and 86 hybridoma cell clones secreting monoclonal antibodies (McAb) were obtained after fusing twice and cloning 3–4 times. 15 of these clones secreted monoclonal anti-idiotypic antibodies (anti-Id McAb). The results showed that 12 of 15 anti-Id antibodies reacted only with homologous λ chain and IgD, not with A chain, κ chain, IgG, IgA, IgM, IgD, IgE, albumin and paraglobulin from normal subjects. Indirect immunofluorescent assay demonstrated that positive reaction rate between anti-Id McAbs and peripheral blood lymphocytes or bone marrow cells of the patient with myeloma was up to 23 %. No reaction was observed between anti-Id McAb and peripheral blood lymphocytes or bone marrow cells from normal subjects. Some of these McAbs presented positive reaction with plasmacytoma cell lines cultured in our laboratory.

Summary100%, 75%, 50%, 25% and 12.5% oxidized dextran T10 (Dex T1o) were used as intermediate carriers for conjugating drug daunorubicin (DNR) and antibody anti-human thymocitic globulin (AHTG), to form different immunoconjugates, AHTG:Dex:DNR. It was demonstrated that the conjugate with 25% oxidized Dex T10 as intermediate carrier linked more DNR molecules than the others. The degree of its substitution was 10–11 moles of DNR per mole of AHTG. Moreover, because the amount to reducing agent sodium borohydride (NaBH4), required for the reduction reaction, was relatively small, its damaging effect on AHTG and DNR was lessened accordingly.The antitumor effect of AHTG: Dex: DNR in vitro was tested by using 24-h cytotoxicity assay, with CEM as target cell. Cytotoxic effect of the conjugate was proven and the LD56 was 10.68 μg/ml. However, it showed only slight cytotoxic effect on non-target cell K562. When 10 min cytotoxicity assay was performed to show the specific tumor-killing effect of the conjugate, it revealed an obvious cytotoxic activity toward CEM, with the LD50 being 14.79 μg/ml, but hardly toward K562. These results suggest that AHTG:Dex:DNR possesses specific cytotoxic effect.

SummarySublethally irradiated mice were immediately treated with 250 mg/kg Ligustrazine Phosphiatis intraperitoneally twice a day for seven days, and the bone marrow sections of ulna were observed. On the 5th day, the number of bone marrow microvessels of the Ligustrazine group was much greater than that of the control group. On the 7th day, the amount of the control group decreased to normal, while the ligustrazine group was still increasing, and the microvessel area was enlarged obviously. The percentage of the hematopoietic tissue volume in bone marrow between the two groups had no significant difference in the first 7 days. On the 7th day after irradiation, the peripheral neutrophilic granulocytes increased in the Ligustrazine group. The results suggested that early use of Ligustrazine after acute radiation injury might improve the blood supply of bone marrow, and be helpful for recovery of hematopoiesis.

SummaryHemorrheological studies carried out on 14 recipients of cadaveric kidney transplant regularly before and after transplantation showed very significant differences in plasma viscosity in the period of stable function after successfully transplanted kidney, dialysis before transplantation, and renal rejection (P<0.00l). Significant difference was observed in fibrinogen level in cases with stable normal function and rejection (P<0.05,). Significant positive correlation was found between plasma viscosity and fibrinogen level in patients on dialysis as well as after transplantation. The results in this series showed that abnormal changes in hemorrheology respond to the development of rejection episode and deterioration of renal function in patients after receiving cadaveric renal allografts.

ZusammenfassungNach 5 Jahren intensiver experimenteller Vorarbeit wurden von Dezember 1977 bis Januar 1981 9 orthotope Lebertransplantationen durchgeführt. Die Hauptindikation stellt das nicht resezierbare primäre Hepatom dar. Zur Gewinnung einer vitalen Spenderleber wird die einfache hypotherme Konservierung mit Sacks’ Lösung als vorzüglich bewertet; die warme Ischämiezeit schwankte zwischen 0–8 Min., und die kalte Aufbewahrung darf nicht länger als 4 1/2 Std. dauern. Die wesentlichen Punkte der Operationstechnik bei der Empfänger-Hepatektomie und Einpflanzung der Spenderleber werden erörtert. Alle Gefäße werden mit fortlaufender Naht einschichtig End-zu-End anastomosiert; die direkte End-zu-End-Choledocho-Choledochostomie ist jetzt bevorzugt zu verwenden. Es gibt keinen sicheren Parameter für die Beurteilung der Abstoßung, deren Diagnose hauptsächlich auf der klinischen Beobachtung von einer Reihe von Symptomen beruht. Eine Kombination von Azathioprin und Prednison, eventuell mit Zusatz von ALG hat sich als immunosuppressiv wirkend erwiesen. Das Prinzip ist, niedrigere Steroiddosen zu verwenden, und die Dosis rascher zu reduzieren. Bis jetzt hatten wir keinen Exitus in tabula, aber die Perioperationsletalität war ziemlich hoch. Von 9 Fällen lebt noch einer, jetzt über 5 Monate, in gutem Allgemeinzustand und mit genügender Leberfunktion und mäßigem Ikterus. Unser am längsten Überlebender, bis jetzt auch der nach Lebertransplantation am längsten Überlebende in ganz China, lebte nahezu 9 Monate (264 Tage) und starb dann an allgemeiner Metastasierung

After irradiation with 8 Gy 60Co gamma-ray, mice were immediately given intraperitoneal injection of 200 mg 100% compound blood-activating soup twice a day. On the 3rd and 7th day, the P53 gene expression of bone marrow hematopoietic cells in Chinese drug group was found to be higher than that in normal group, and it was also significantly higher than that in control group. The expression level of GADD153 gene which was not expressed in normal group was much lower in Chinese drug group than that in control group. On the 7th day after irradiation, the P53 and GADD153 gene expression levels of splenic mononuclear cells were consistent with those of bone marrow hematopoietic cells both in Chinese drug group and control group. On the 3rd and 7th day, the bone marrow hematopoietic tissue volume in Chinese drug group was higher than that in control group, with no difference found between the two groups. While on the 14th day, the difference became significant (P < 0.01).
