Abstract Research covered six variants: control, unfertilized carrots and carrots fertilized with: KIO3, Na2SeO4, Na2SeO3, KIO3 and simultaneously with Na2SeO4, and fertilized with KIO3 and simultaneously Na2SeO3. Carrots enriched with iodate or selenite, or both iodate and selenite, were characterized by higher amount of these minerals. Changes to the content of micro- and macroelements, during the cooking time of the carrots, both in peeled and unpeeled carrots, did not head in the same direction (increase, decrease and no change). However, cooking an unpeeled carrot generally resulted in the increased content of polyphenol and carotenoids. On the other hand, cooking peeled carrots led to a decrease in the content of polyphenol and a general lack of change in carotenoid content in relation to the unpeeled cooked carrot. During cooking, the antioxidant activity of the carrot being assessed changed together with the direction of changes in polyphenol content but not in line with the direction of changes in carotenoids.

The aim of this study has been to measure the level of lead, cadmium, nitrates, and nitrites in the daily diets of children and adolescents from orphanages located in Krakow (Poland). Diets were collected over four seasons of 2009. The content of cadmium and lead was measured with flameless atomic absorption spectrometry. Nitrates and nitrites in diets were measured using the Griess colorimetric method. In all orphanages, the average intake of lead with daily diets, regardless of the season, ranged from 1.11xa0±xa00.15 to 22.59xa0±xa00.07xa0μg/kgxa0bw/week. The average cadmium intake by children and adolescents ranged between 3.09xa0±xa00.21 and 20.36xa0±xa02.21xa0μg/kgxa0bw/week and, for all orphanages, exceeded the tolerable weekly intake (TWI) level. Daily intake of nitrates and nitrites ranged respectively from 27 to 289xa0% and from 9 to 99xa0% of the acceptable daily intake (ADI). The youngest children, with lower body mass, were particularly sensitive to the excessive intakes of cadmium and nitrates.

The aim of this study was to analytically evaluate the daily intake of total polyphenols, β-carotene, and vitamin C by 10- to 12-year-old children. Twenty four–hour dietary recalls were collected in spring and autumn 2006 from students in Krakow and Skawina (Malopolska region, Poland). The content of vitamin C, polyphenols, and β-carotene was determined in reconstructed daily diets. Statistical analysis did not show any significant differences between the mean intake of measured compounds according to school location, gender, or season. Diets of children contained adequate amounts of polyphenols in relation to the recommended values. Intake of β-carotene usually exceeded the recommended retinol equivalent; in contrast, vitamin C did not meet requirements. Increased consumption of fruits and vegetables rich in vitamin C is recommended.

BackgroundOur previous study showed that fatty acids extract obtained from CLA-enriched egg yolks (EFA-CLA) suppressed the viability of MCF-7 cancer cell line more effectively than extract from non-enriched egg yolks (EFA). In this study, we analysed the effect of EFA-CLA and EFA on transcriptome profile of MCF-7 cells by applying the whole Human Genome Microarray technology.ResultsWe found that EFA-CLA and EFA treated cells differentially regulated genes involved in cancer development and progression. EFA-CLA, compared to EFA, positively increased the mRNA expression of TSC2 and PTEN tumor suppressors as well as decreased the expression of NOTCH1, AGPS, GNA12, STAT3, UCP2, HIGD2A, HIF1A, PPKAR1A oncogenes.ConclusionsWe show for the first time that EFA-CLA can regulate genes engaged in AKT/mTOR pathway and inhibiting cell cycle progression. The observed results are most likely achieved by the combined effect of both: incorporated CLA isomers and other fatty acids in eggs organically modified through hens’ diet. Our results suggest that CLA-enriched eggs could be easily available food products with a potential of a cancer chemopreventive agent.

Abstract The objective of this research were to investigate the effect of a conjugated linoleic acid (CLA)-enriched diet on Isa Brown laying hen health status and to provide a comprehensive analysis of changes in blood parameters, liver morphology and selected hepatic gene expression. Hens were allocated to the control and experimental group (diet enriched with 0.75% CLA) for a total period of 4 m. At the end of the experiment half of the hens from each group were slaughtered for analyses. The remaining hens were transferred to an organic farm for the next 5 m and fed on the diet without CLA supplementation. The CLA-enriched diet resulted in significant changes in blood and serum parameters; specifically, haematocrit (HCT), mean corpuscular volume (MCV) and white blood cells (WBC) count were decreased compared to the control. The total cholesterol (TC) was not significantly affected while the triacylglycerol’s (TG) concentration was elevated. The activity of alanine aminotransferase (ALT) was significantly increased in the CLA-supplemented group, while aspartate aminotransferase (AST) showed an increasing tendency. Liver biopsies showed pathological changes classified as non-alcoholic fatty liver disease (NAFLD). Additionally, the expression of hepatic genes involved in fatty acids synthesis (ME1, ACLY, ACC, FASN, SCD1), oxidation (CPT1α, PPARA), detoxification processes (Cytochrome P450, CYP, Flavin-containing monooxygenase, FMO3), oxidative stress (NOX4, XbP1) and inflammation (IL6, TNFα) were elevated. Cessation of CLA supplementation for 5 m of organic farming resulted in normalisation of blood and hepatic parameters to the levels observed in control hens. The results of this study indicate that dietary CLA triggers an integrated stress response in laying hens and activates mechanisms involved in liver detoxification.

Objective: The objective of this study was analytical assessment of the concentration of selected antioxidants as well as total antioxidant capacity of daily diets of children and adolescents from orphanages in Krakow (Poland). Methods: Daily diets of children and adolescents were collected in 4 seasons of the year from 7 orphanages located in Krakow. The concentrations of vitamin C, β-carotene, polyphenolic compounds, and antioxidant capacity were measured. Results: The lowest content of vitamin C in diets of children and adolescents was measured in diets collected in autumn. The estimated average requirement (EAR) was in the range 49.0%–76.4%. In winter the content of this compound was significantly higher in daily diets (113.9%–192.0% coverage of EAR) compared to the level of this nutrient in daily diets from other seasons. The content of β-carotene, lower than 50% of EAR value, calculated as the vitamin A equivalent, has been demonstrated in 4 orphanages in summer and in one in spring and autumn. The highest mean content of this antioxidant in daily diets was found in winter, when β-carotene intake met recommendations in the range 131%–368%. The mean concentration of polyphenolic compounds per year was in the range 301.5–537.5 mg/person/day. It has been observed that diets during the winter and autumn generally provide higher amounts of polyphenols than in other seasons. The highest antioxidant activity was measured in diets collected during the winter season. Conclusions: The results indicate that the diets of children and adolescents should contain more fresh vegetables and fruits, which are good source of natural antioxidants.

Iodine is an essential trace element for humans. Foliar application of micronutrients is successfully used in order to increase the concentration of essential elements in vegetables. The aim of this study was to evaluate the iodine absorption in the rat organism fed foliar biofortified lettuce. The presented study was consisted of the vegetative and animal experiment. In the vegetative experiment with lettuce, two combinations of foliar application were used: (1) control—without iodine application and (2) iodine application in the potassium iodide (KI) form. In the animal experiment, Wistar rats were divided to four groups, which received one of four diets: (1) C—control diet containing iodine in the KI form, (2) D—diet deficient in iodine, (3) Dxa0+xa0BL—diet containing biofortified lettuce, and (4) Dxa0+xa0CL—diet containing control lettuce (as the only source of iodine in diet, respectively). The diets contained 0.260, 0.060, 0.254 and 0.075xa0mg I/kg, respectively. In order to determine the iodine absorption in the rat organisms, the content of this trace element was measured in urine, faeces and in selected organs with the use of the ICP-OES technique. Foliar application of the KI increased the content of iodine in lettuce. The rats from the Dxa0+xa0BL group excreted significantly less iodine in their urine and faeces and also accumulated more iodine in the organs than the rats from the C group. Iodine with biofortified lettuce was much bioavailable for rodents than iodine from control diet. Biofortified lettuce can be a source of iodine in a diet of human and can improve iodine nutrition.

Triclosan (TCS) is an antimicrobial agent that is used extensively in personal care and in sanitising products. A number of studies have shown the presence of TCS in different human tissues such as blood, adipose tissue, the liver, brain as well as in breast milk and urine. N-Methyl-D-aspartate receptors (NMDARs) are glutamate-gated ion channels that are widely expressed in the central nervous system and which play key roles in excitatory synaptic transmission. There is, however, no data on the involvement of NMDAR subunits in the apoptotic and neurotoxic effects of TCS. Our experiments are the first to show that TCS used at environmentally relevant concentrations evoked NMDA-dependent effects in neocortical neurons in primary cultures, as MK-801, an uncompetitive NMDA receptor antagonist, reduced the levels of TCS-induced ROS production as well as caspase-3 activity and LDH release. TCS caused a decrease in protein expression of all the studied NMDA receptor subunits (GluN1, GluN2A, GluN2B) that were measured at 3, 6 and 24xa0h post-treatment. However, at 48xa0h of the experiment, the level of the GluN1 subunit returned to the control level, and the levels of the other subunits showed a tendency to increase. In TCS-treated neocortical cells, protein profiles of NMDAR subunits measured up to 24xa0h were similar to mRNA expression of GluN1 and GluN2A, but not to GluN2B mRNA. In this study, cells transiently transfected with GluN1, GluN2A or GluN2B siRNA exhibited reduced levels of LDH release, which suggests the involvement of all of the studied NMDAR subunits in the neurotoxic action of TCS. According to our data, GluN1 and GluN2A were mainly responsible for neuronal cell death as evidenced by neutral red uptake, whereas GluN2A was involved in TCS-induced caspase-3-dependent apoptosis. We suggest that TCS-evoked apoptosis and neurotoxicity could be related to transient degradation of NMDAR subunits in mouse neurons. Furthermore, recycling of NMDAR subunits in response to TCS is possible. Because transfections with specific siRNA did not completely abolish the effects of TCS as compared to cells transfected with negative siRNA in this study, other NMDAR-independent mechanisms of TCS action are also possible.

The objective of this study was to determine the chemical composition in the dried leaves and the content of bioactive compounds, antioxidant activity as well as the identification of polyphenolic compounds not only in extracts but also in infusions prepared from leaves of selected strains of buckwheat. To the best of our knowledge, it is the first comprehensive and in-depth study, which focuses on identification and quantitative determination of individual polyphenolic compounds of buckwheat leaves. Furthermore, the aim of presented research is to analyse the new, unknown strains of buckwheat. A statistically significant effect of strain on the content of individual components was observed. The extracts contained more polyphenols and were characterized by higher antioxidant activity than infusions. Strains DW15 and DWO Karzel were characterized by one of the highest contents of polyphenols, dietary fibre, protein, ash and antioxidant activity. Phenolic acids (3,5-diCQA, ferulic acid, sinapinic acid, p-coumaric acid), flavanones (hesperidin), flavonoids (isorhamnetin) as well as flavanols (catechin) and flavonols (rutin, quercetin) in the extracts and infusions, were found. Both extracts and infusions were the richest in ferulic acid. These studies indicate that the buckwheat leaves can be used for preparation of various types of tea and can also be introduced for consumers as the source of bioactive compounds.

This study was designed to determine the effect of feeding female rats with low-calcium diet containing one of three fructan sources (Jerusalem artichoke, yacon, Beneo Orafti Synergy1), on selected bone parameters. Growing Wistar rats were fed modified AIN-93xa0G diet enriched in fructan sources (8%), added alone or as a strawberry sorbet ingredient. Two of eight groups were a validation model, where the positive control group was fed with recommended calcium dose in the diet (RCD), and negative one – with low calcium diet (LCD). After 12xa0weeks, femoral Ca content, bone densitometry, architecture and hardness were examined. The positive effects on femoral Ca content and cortical thickness, area and content in distal part of bone was observed after feeding animals diet enriched in Jerusalem artichoke sorbet. Beneficial action on other bone tomographic parameters (particularly trabecular volumetric bone mineral density) in this part of femur were associated mainly with the consumption of the diet with sorbet containing yacon. Our results showed an important role of diet containing frozen strawberry desserts enriched in fructan sources in the maintenance of healthy bones of growing organism. It may suggest possible synergisms between fructans and bioactive substances of strawberry.
