In 2016, the order Mononegavirales was emended through the addition of two new families (Mymonaviridae and Sunviridae), the elevation of the paramyxoviral subfamily Pneumovirinae to family status (Pneumoviridae), the addition of five free-floating genera (Anphevirus, Arlivirus, Chengtivirus, Crustavirus, and Wastrivirus), and several other changes at the genus and species levels. This article presents the updated taxonomy of the order Mononegavirales as now accepted by the International Committee on Taxonomy of Viruses (ICTV).

Molecular detection of two adenoviruses associated with disease in Australian lizards.

We give the first published description of the pathology and molecular findings associated with adenovirus infection in lizards in Australia. A central netted dragon (Ctenophorus nuchalis) exhibited severe necrotising hepatitis with abundant intranuclear inclusion bodies within hepatocytes and rarely within intestinal epithelial cells. Polymerase chain reaction (PCR) using pooled tissues yielded an amplicon that shared strong nucleotide identity with an agamid adenovirus (EU914203). PCR on the liver of a bearded dragon (Pogona minor minor) with illthrift, coccidiosis, nematodiasis and hepatic lipidosis yielded an amplicon with strong nucleotide identity to a helodermatid adenovirus (EU914207).

Paramyxoviruses in reptiles: A review

In 1972, an outbreak of neurorespiratory disease in a Swiss serpentarium formed the basis for the first description of a paramyxovirus isolated from a reptile. In the forty years since this outbreak, there have been over 50 published reports about reptilian paramyxoviruses from all over the world. The majority of these investigations have concerned themselves with ferlaviruses (sometimes previously referred to as ophidian paramyxoviruses, or OPMV). The biology of these viruses is reviewed and this is followed by a review of the clinical findings that are associated with ferlaviral infection and the various diagnostic tests that are used to identify infected reptiles. Recently, a second, and highly divergent, reptilian paramyxovirus, Sunshine virus, was described in Australian pythons, so it is an opportune time to reflect on the paramyxoviruses that infect reptiles.

Is Wildlife Fertility Control Always Humane

Simple Summary There are various fertility control methods (modalities) currently available that aim to reduce the abundance of problematic free-ranging mammalian wildlife. Here, we propose that dissimilarities in the mechanism of action indicate these methods produce great variation in animal welfare outcomes. We present a framework to assist managers in minimising animal welfare risks. Abstract Investigation of fertility control techniques to reduce reproductive rates in wildlife populations has been the source of much research. Techniques targeting wildlife fertility have been diverse. Most research into fertility control methods has focused upon efficacy, with few studies rigorously assessing animal welfare beyond opportunistic anecdote. However, fertility control techniques represent several very different mechanisms of action (modalities), each with their own different animal welfare risks. We provide a review of the mechanisms of action for fertility control methods, and consider the role of manipulation of reproductive hormones (“endocrine suppression”) for the long-term ability of animals to behave normally. We consider the potential welfare costs of animal manipulation techniques that are required to administer fertility treatments, including capture, restraint, surgery and drug delivery, and the requirement for repeated administration within the lifetime of an animal. We challenge the assumption that fertility control modalities generate similar and desirable animal welfare outcomes, and we argue that knowledge of reproductive physiology and behaviour should be more adeptly applied to wild animal management decisions. We encourage wildlife managers to carefully assess long-term behavioural risks, associated animal handling techniques, and the importance of positive welfare states when selecting fertility control methods as a means of population control.

Sunshine virus in Australian pythons.

Sunshine virus is a recently discovered novel paramyxovirus that is associated with illness in snakes. It does not phylogenetically cluster within either of the two currently accepted paramyxoviral subfamilies. It is therefore only distantly related to the only other known genus of reptilian paramyxoviruses, Ferlavirus, which clusters within the Paramyxovirinae subfamily. Clinical and diagnostic aspects associated with Sunshine virus are as yet undescribed. The objective of this paper was to report the clinical presentation, virus isolation, PCR testing and pathology associated with Sunshine virus infection. Clinical records and samples from naturally occurring cases were obtained from two captive snake collections and the archives of a veterinary diagnostic laboratory. The clinical signs that are associated with Sunshine virus infection are localised to the neurorespiratory systems or are non-specific (e.g. lethargy, inappetence). Out of 15 snakes that were infected with Sunshine virus (detected in any organ by either virus isolation or PCR), the virus was isolated from four out of ten (4/10) sampled brains, 3/10 sampled lungs and 2/7 pooled samples of kidney and liver. In these same 15 snakes, PCR was able to successfully detect Sunshine virus in fresh-frozen brain (11/11), kidney (7/8), lung (8/11) and liver (5/8); and various formalin-fixed paraffin-embedded tissues (7/8). During a natural outbreak of Sunshine virus in a collection of 32 snakes, the virus could be detected in five out of 39 combined oral-cloacal swabs that were collected from 23 of these snakes over a 105 day period. All snakes that were infected with Sunshine virus were negative for reovirus and ferlavirus by PCR. Snakes infected with Sunshine virus reliably exhibited hindbrain white matter spongiosis and gliosis with extension to the surrounding grey matter and neuronal necrosis evident in severe cases. Five out of eight infected snakes also exhibited mild bronchointerstitial pneumonia. Infection with Sunshine virus should be considered by veterinarians investigating disease outbreaks in snakes, particularly those that are associated with neurorespiratory disease.

Assessing and mitigating post-operative castration pain in Bos indicus cattle

Cattle on pastoral land are subject to potentially painful husbandry procedures. In northern Australia, these practices generally occur once a year after the muster and the procedures are usually performed on animals older than 6 months of age. It is seldom that any pain mitigating medications are administered at this time and there is increasing concern that this perceived impost on the animal’s welfare will become more significant at both an economic and cultural level. There is a need to investigate the possibility of using simple, cost-effective, readily available medications, administered using relatively quick and easily taught techniques to increase the chance of industry adoption. This research used castration of 6–8-month-old Bos indicus calves as the test model because it is arguably one of the most common and most painful procedures endured by the animals. Forty-eight, 6–8-month-old Brahman bulls were surgically castrated after random assignment to six different treatment groups (n = 8): no castration Control (NC); castration (C); castration with post-operative meloxicam (CMpost-op); castration with lignocaine (CL); castration with lignocaine and post-operative meloxicam (CLMpost-op); castration with pre-operative meloxicam (CMpre-op). Serial measures of weight, blood cortisol concentrations, balk score, crush score, and daily activity (steps taken, number and duration of rest bouts) were taken. Linear mixed modelling was used to compare experimental groups. Bulls that were castrated and did not receive post-operative meloxicam (C and CL) had significantly lower average weights at all time points (Days 0–13, P < 0.001) of 170.9 kg and 168.6 kg, respectively, than those that did (CMpost-op and CLMpost-op) at 174.7 kg and 173.7 kg, respectively. When lignocaine was not administered before castration, Day 1 post-operative cortisol concentrations were significantly lower when post-operative meloxicam was provided than when it was not (CMpost-op: 47.2 nmol/L vs C: 65.5 nmol/L). When post-operative meloxicam was not given, animals that were given lignocaine before castration had significantly lower cortisol concentrations than those that received no lignocaine (CL: 52.7 nmol/L vs C: 65.5 nmol/L). Animals that were given pre-operative meloxicam had, on average, significantly lower Day 1 post-operative cortisol concentrations than animals that received no lignocaine or meloxicam (CMpre-op: 44.8 nmol/L vs C: 65.5 nmol/L). Analysis of Day 1 (the day following castration) data showed that animals that were given pre-operative meloxicam (CMpre-op) were significantly more active than animals in all other treatment groups (P = 0.038). It was concluded that 6–8-month old, Bos indicus bulls benefit from the administration of both lignocaine before surgery and peri-operative meloxicam. The analgesic methods used are readily adoptable by industry with respect to cost, efficiency and degree of technical skill required.

Animal welfare and the use of procedural documents: Limitations and refinement

Abstract. Increased scrutiny of animal welfare in wildlife management has seen a recent proliferation in the use of procedural documents (standard operating procedures, codes of practice etc.). Some procedural documents are presumed to represent ‘best practice’ methods, whereby adherence to prescribed inputs is explicitly purported to generate humane outcomes. However, the relationship between what is done to animals (inputs) and what they experience (outputs), as assessed by animal-based measures, has received little attention. Procedural documents are commonly developed in the absence of empirical animal-based measures, creating uncertainty in animal welfare outcomes. Prescribed procedures are valuable as guidelines for standardising methodology, but the development of ‘welfare standards’ that focus on desired thresholds for animal-based measures offers many advantages for improving animal welfare. Refinement of the use of procedural documents in wildlife management is required to ensure they generate desirable outcomes for animals, and do not preclude the development of improved methods.

An animal welfare assessment framework for helicopter darting: a case study with a newly developed method for feral horses

Abstract Context. Helicopter darting (chemical immobilisation) is a very useful technique for large wild herbivores, such as feral horses (Equus caballus). There is currently no reliable framework to report on the animal welfare impacts of helicopter darting methods. Aim. The aim of this study was to develop an animal welfare assessment framework for helicopter darting methods, using quantifiable parameters, and to test it with a case study using a newly developed feral horse capture technique. Methods. Quantifiable animal welfare parameters were recorded for 11 feral horses captured using a traditional helicopter darting method in north-western Australia in October 2014. Welfare parameters chosen focused on quantifying the duration of procedures and the frequency of adverse events. They included chase time (CT; min) before darting, induction time (IT; min) between darting and recumbency, recumbency time (RT; min), total time (TT; CT+IT+RT; min), repeat-darting rate (animals requiring >1 dart; %), target zone accuracy rate (darts striking the intended anatomical area; %) and mortality rate (at time of capture and 14 days post-capture; %). Results. Median CT was 2 min, median IT was 19 min, median RT was 16 min and median TT was 38 min. Repeat-darting rate was 45%, target zone accuracy rate was 53% and mortality rates (time of capture and 14 days post-capture) were zero. Conclusions. Animal welfare parameters can be quantified for helicopter darting through estimation of the duration of procedures and the frequency of adverse events. Use of this framework will allow the identification of parameters requiring refinement for newly developed helicopter darting techniques. Implications. Animal welfare parameters are particularly important for helicopter-based darting methods. Pilot studies, using quantified parameters, should be performed for newly developed capture techniques before they are approved for large-scale programs.

Diagnostic investigation of new disease syndromes in farmed Australian saltwater crocodiles (Crocodylus porosus) reveals associations with herpesviral infection

Since 2006, 3 new disease syndromes have emerged in farmed saltwater crocodiles (Crocodylus porosus) in the Northern Territory of Australia. We describe the syndromes through a retrospective study of laboratory findings from 187 diagnostic cases submitted to Berrimah Veterinary Laboratories between 2005 and 2014. The first syndrome was characterized by conjunctivitis and/or pharyngitis (CP), primarily in hatchlings. Herpesviruses were isolated in primary crocodile cell culture, or were detected by PCR directly from conjunctiva or pharyngeal tissue, in 21 of 39 cases of CP (54%), compared with 9 of 64 crocodiles without the syndrome (14%, p < 0.0001). Chlamydiaceae were detected by PCR in conjunctiva or pharyngeal tissue of 55% of 29 CP cases tested, and of these, 81% also contained herpesvirus. The second syndrome occurred in juveniles and growers exhibiting poor growth, and was characterized histologically by systemic lymphoid proliferation and nonsuppurative encephalitis (SLPE). Herpesviruses were isolated or detected by PCR from at least 1 internal organ in 31 of 33 SLPE cases (94%) compared with 5 of 95 crocodiles without the syndrome (5%, p < 0.0001). The third syndrome, characterized by multifocal lymphohistiocytic infiltration of the dermis (LNS), occurred in 6 harvest-sized crocodiles. Herpesviruses were isolated from at least 1 skin lesion in 4 of these 6 cases. Although our study revealed strong associations between herpesvirus and the CP and SLPE syndromes, the precise nature of the role of herpesvirus, along with the pathogenesis and epidemiology of the syndromes, requires further investigation.

Mechanical nociceptive threshold testing in Bos indicus bull calves

The aim of this prospective, controlled, randomised trial was to develop a technique for mechanical nociceptive threshold testing (NTT) to assess pain in Bos indicus bull calves undergoing surgical castration. Analgesia was provided by 0.5 mg/kg subcutaneous (SC) meloxicam (M) and/or 2 mg/kg of intra-testicular and SC (at the surgery sites) lidocaine (L). Forty-eight Brahman bull calves at 6–8 months of age were divided into six study groups, each with eight animals: no surgery control; surgical castration (C) without analgesia; C and Mpre-op; C and Mpost-op; C, L and Mpost-op; C and L. Mechanical NTT was performed the day before surgery (Day –1) and on Days 1, 2, 6, 10 and 13 after surgery. A handheld manual pneumatic device with a 1-mm (diameter) blunt pin was used to deliver a mechanical stimulus to a maximum of 27 Newtons either side of the most dorsal aspect of the sacrum. The most frequent responses to the mechanical stimulus were lifting or kicking of the leg on the same side as the stimulus (31%) and stepping away from the stimulus (24.9%). Data were analysed with a mixed effect linear model with the nociceptive threshold (NT) as the response variable and day and analgesic treatment as predictors (P < 0.05 was considered significant). For all groups, there was a trend towards decreasing NT over the study period but there were no significant differences between groups. Step down model selection with day, batch and treatment terms revealed a significant effect of day (P < 0.001) and batch (P = 0.007). Mechanical NTT for assessment of pain in Bos indicus bull calves requires further refinement to determine if this is a useful method of pain assessment.
