Abstract The assay of galactose-1-phosphate uridyltransferase (E.C. 2.7.7.12: UDP-glucose: α- d -galactose-1-phosphate uridyltransferase) in red and white cells of blood is reported using [14C]galactose-1-phosphate and UDPglucose as substrates and measuring the 14C activity in the nucleotides. The reaction requires for optimal activity a concentration of 3.2 mM galactose-1-phosphate and 1.6 mM UDPG with the pH maintained at 8.40.

Regain of biological activity of gonadotropin from choriocarcinoma.

Abstract Gonadotropin from patients with choriocarcinoma was investigated to find means to regain biological activity which was lost during and after purification by diethylaminoethyl-cellulose chromatography. The loss was not due to changes in the content of N-acetyl neuraminic acid. Regain of biological activity as tested by the mouse uterine weight gain method was effected by incubating the gonadotropin in 20 mM sodium phosphate buffer at pH 7.2 containing 3.5 mM sodium dodecyl sulfate, 25 mM oxidized glutathione, and 6.0 mM ethylenediamine tetra-acetate for 24 hours at 37°C. The regain was 50- to 85-fold. The possible mechanisms are discussed.
