Marine and ice-core records show that the Earth has experienced a succession of glacials and interglacials during the Quaternary (last ∼2.6 million years), although it is often difficult to correlate fragmentary terrestrial records with specific cycles. Aminostratigraphy is a method potentially able to link terrestrial sequences to the marine isotope stages (MIS) of the deep-sea record. We have used new methods of extraction and analysis of amino acids, preserved within the calcitic opercula of the freshwater gastropod Bithynia, to provide the most comprehensive data set for the British Pleistocene based on a single dating technique. A total of 470 opercula from 74 sites spanning the entire Quaternary are ranked in order of relative age based on the extent of protein degradation, using aspartic acid/asparagine (Asx), glutamic acid/glutamine (Glx), serine (Ser), alanine (Ala) and valine (Val). This new aminostratigraphy is consistent with the stratigraphical relationships of stratotypes, sites with independent geochronology, biostratigraphy and terrace stratigraphy. The method corroborates the existence of four interglacial stages between the Anglian (MIS 12) and the Holocene in the terrestrial succession. It establishes human occupation of Britain in most interglacial stages after MIS 15, but supports the notion of human absence during the Last Interglacial (MIS 5e). Suspicions that the treeless ‘optimum of the Upton Warren interstadial’ at Isleworth pre-dates MIS 3 are confirmed. This new aminostratigraphy provides a robust framework against which climatic, biostratigraphical and archaeological models can be tested.

Aminostratigraphies of Quaternary non-marine deposits in Europe have been previously based on the racemization of a single amino acid in aragonitic shells from land and freshwater molluscs. The value of analysing multiple amino acids from the opercula of the freshwater gastropod Bithynia, which are composed of calcite, has been demonstrated. The protocol used for the isolation of intra-crystalline proteins from shells has been applied to these calcitic opercula, which have been shown to more closely approximate a closed system for indigenous protein residues. Original amino acids are even preserved in bithyniid opercula from the Eocene, showing persistence of indigenous organics for over 30 million years. Geochronological data from opercula are superior to those from shells in two respects: first, in showing less natural variability, and second, in the far better preservation of the intra-crystalline proteins, possibly resulting from the greater stability of calcite. These features allow greater temporal resolution and an extension of the dating range beyond the early Middle Pleistocene. Here we provide full details of the analyses for 480 samples from 100 horizons (75 sites), ranging from Late Pliocene to modern. These show that the dating technique is applicable to the entire Quaternary. Data are provided from all the stratotypes from British stages to have yielded opercula, which are shown to be clearly separable using this revised method. Further checks on the data are provided by reference to other type-sites for different stages (including some not formally defined). Additional tests are provided by sites with independent geochronology, or which can be associated with a terrace stratigraphy or biostratigraphy. This new aminostratigraphy for the non-marine Quaternary deposits of southern Britain provides a framework for understanding the regional geological and archaeological record. Comparison with reference to sites yielding independent geochronology, in combination with other lines of evidence, allows tentative correlation with the marine oxygen isotope record.

Between the Levant and the Indian sub-continent only a few Acheulean sites have been documented, hampering models of hominin dispersals. Here we describe the first Acheulean sites to be discovered in the Nefud Desert of northern Arabia. The four sites occur in a variety of settings including adjacent to an alluvial fan drainage system, at a knappable stone source, and on the margins of endorheic basins. We discuss the implications of the sites for hominin landscape use, in particular the preferential transport and curation of bifaces to fresh water sources. The bifaces correspond to the Large Flake middle Acheulean in the Levantine sequence. The sites occupy a gap in the distribution of the Acheulean across the Saharo-Arabian arid belt, and as such have implications for dispersal routes between Africa and Asia.

The charge-flipping algorithm has been adapted to allow symmetry constraints to be included during the solution of structures from diffraction data. Rather than impose symmetry at the start of the algorithm, which is known to cause the process to stagnate, it is shown that the algorithm must be allowed to find an intermediate stable solution first. Although care is needed when using this modified algorithm, the improvement in the fidelity of the structure solution is considerable.

The Pleistocene archaeological record of the Arabian Peninsula is increasingly recognized as being of great importance for resolving some of the major debates in hominin evolutionary studies. Though there has been an acceleration in the rate of fieldwork and discovery of archaeological sites in recent years, little is known about hominin occupations in the Pleistocene over vast areas of Arabia. Here we report on the identification of five new Middle Palaeolithic sites from the Nejd of central Arabia and the southern margins of the Nefud Desert to the north. The importance of these sites centers on their diversity in terms of landscape positions, raw materials used for lithic manufacture, and core reduction methods. Our findings indicate multiple hominin dispersals into Arabia and complex subsequent patterns of behavior and demography.

The Middle to Later Stone Age transition in Africa has been debated as a significant shift in human technological, cultural, and cognitive evolution. However, the majority of research on this transition is currently focused on southern Africa due to a lack of long-term, stratified sites across much of the African continent. Here, we report a 78,000-year-long archeological record from Panga ya Saidi, a cave in the humid coastal forest of Kenya. Following a shift in toolkits ~67,000 years ago, novel symbolic and technological behaviors assemble in a non-unilinear manner. Against a backdrop of a persistent tropical forest-grassland ecotone, localized innovations better characterize the Late Pleistocene of this part of East Africa than alternative emphases on dramatic revolutions or migrations.Most of the archaeological record of the Middle to Later Stone Age transition comes from southern Africa. Here, Shipton et al. describe the new site Panga ya Saidi on the coast of Kenya that covers the last 78,000 years and shows gradual cultural and technological change in the Late Pleistocene.

Background and aims: Understanding the aetiology, clinical presentation and prognosis of type 2 diabetes (T2D) and optimizing its treatment might be facilitated by biomarkers that help predict a person’s susceptibility to the risk factors that cause diabetes or its complications, or response to treatment. The IMI DIRECT (Diabetes Research on Patient Stratification) Study is a European Union (EU) Innovative Medicines Initiative (IMI) project that seeks to test these hypotheses in two recently established epidemiological cohorts. Here, we describe the characteristics of these cohorts at baseline and at the first main follow-up examination (18-months). Materials and methods: From a sampling-frame of 24,682 European-ancestry adults in whom detailed health information was available, participants at varying risk of glycaemic deterioration were identified using a risk prediction algorithm and enrolled into a prospective cohort study (n=2127) undertaken at four study centres across Europe (Cohort 1: prediabetes). We also recruited people from clinical registries with recently diagnosed T2D (n=789) into a second cohort study (Cohort 2: diabetes). The two cohorts were studied in parallel with matched protocols. Endogenous insulin secretion and insulin sensitivity were modelled from frequently sampled 75g oral glucose tolerance (OGTT) in Cohort 1 and with mixed-meal tolerance tests (MMTT) in Cohort 2. Additional metabolic biochemistry was determined using blood samples taken when fasted and during the tolerance tests. Body composition was assessed using MRI and lifestyle measures through self-report and objective methods. Results: Using ADA-2011 glycaemic categories, 33% (n=693) of Cohort 1 (prediabetes) had normal glucose regulation (NGR), and 67% (n=1419) had impaired glucose regulation (IGR). 76% of the cohort was male, age=62(6.2) years; BMI=27.9(4.0) kg/m2; fasting glucose=5.7(0.6) mmol/l; 2-hr glucose=5.9(1.6) mmol/l [mean(SD)]. At follow-up, 18.6(1.4) months after baseline, fasting glucose=5.8(0.6) mmol/l; 2-hr OGTT glucose=6.1(1.7) mmol/l [mean(SD)]. In Cohort 2 (diabetes): 65% (n=508) were lifestyle treated (LS) and 35% (n=271) were lifestyle + metformin treated (LS+MET). 58% of the cohort was male, age=62(8.1) years; BMI=30.5(5.0) kg/m2; fasting glucose=7.2(1.4)mmol/l; 2-hr glucose=8.6(2.8) mmol/l [mean(SD)]. At follow-up, 18.2(0.6) months after baseline, fasting glucose=7.8(1.8) mmol/l; 2-hr MMTT glucose=9.5(3.3) mmol/l [mean(SD)]. Conclusion: The epidemiological IMI DIRECT cohorts are the most intensely characterised prospective studies of glycaemic deterioration to date. Data from these cohorts help illustrate the heterogeneous characteristics of people at risk of or with T2D, highlighting the rationale for biomarker stratification of the disease - the primary objective of the IMI DIRECT consortium. In recent years pastoral regions of western China have been subjected to significant spatial transformation in the name of economic development and environmental protection. Scholarly accounts of these regions have often focused on the state’s efforts to sedentarise herding households; this article, however, examines the significance of the administrative recategorisation of a pastoral district and the relocation of its centre, in line with the state’s policy of creating towns in rural areas. Drawing on ethnographic fieldwork in Alasha, in the west of Inner Mongolia, I show how this particular transformation involved the combination of elements from two distinct spatial formations which characterised state territorialisation in this part of China in the early socialist period: the pastoral district or commune, and the military-agricultural colony. While much recent literature has highlighted the enduring legacy of pre-socialist spatiality in the face of the modern state’s projects of spatial reconfiguration, this article attends to the ways in which the spatial transformations of the early socialist period continue to reverberate today. I show how, for local ethnic Mongolians, the meanings inscribed upon the landscape during this period, and the infrastructural orientations which were established then, today sit awkwardly with official visions of an urban future.

This book explores new approaches to the remarkably detailed information that archaeologists now have for the study of our early ancestors. Rather than explaining the archaeology of stones and bones as the product of group decisions, the contributors investigate how individual action created social life. This challenge to the accepted standpoint of the Palaeolithic brings new models and theories into the period; innovations that are matched by the resolution of data preserving individual action among the stones and bones. The volume brings together examples from recent excavations such as Boxgrove, Schoningen and Blombos Cave and the analyses of artefacts from Middle and Early Upper Pleistocene excavations in Europe, Africa and Asia.
