Stomatal closure is induced by hydraulic signals and maintained by ABA in drought-stressed grapevine

Water saving under drought stress is assured by stomatal closure driven by active (ABA-mediated) and/or passive (hydraulic-mediated) mechanisms. There is currently no comprehensive model nor any general consensus about the actual contribution and relative importance of each of the above factors in modulating stomatal closure in planta. In the present study, we assessed the contribution of passive (hydraulic) vs active (ABA mediated) mechanisms of stomatal closure in V. vinifera plants facing drought stress. Leaf gas exchange decreased progressively to zero during drought, and embolism-induced loss of hydraulic conductance in petioles peaked to ~50% in correspondence with strong daily limitation of stomatal conductance. Foliar ABA significantly increased only after complete stomatal closure had already occurred. Rewatering plants after complete stomatal closure and after foliar ABA reached maximum values did not induced stomatal re-opening, despite embolism recovery and water potential rise. Our data suggest that in grapevine stomatal conductance is primarily regulated by passive hydraulic mechanisms. Foliar ABA apparently limits leaf gas exchange over long-term, also preventing recovery of stomatal aperture upon rewatering, suggesting the occurrence of a mechanism of long-term down-regulation of transpiration to favor embolism repair and preserve water under conditions of fluctuating water availability and repeated drought events.

Morpho-structural and physiological response of container-grown Sangiovese and Montepulciano cvv. (Vitis vinifera) to re-watering after a pre-veraison limiting water deficit

A better physiological and productive performance of cv. Montepulciano versus cv. Sangiovese under well-watered conditions has been recently assessed. The objective of this study was to verify that this behaviour is maintained when a pre-veraison deficit irrigation (vines held at 40% pot capacity from fruit-set to veraison) followed by re-watering (pot capacity reported at 90%). Single leaf assimilation rate and stomatal conductance, diurnal and seasonal whole-canopy net CO2 exchange (NCER) and water use efficiency were always higher in Sangiovese under deficit irrigation. Due to water shortage Montepulciano displayed a more compact growing habit due to decreased shoot and internode length. Sangiovese showed excellent recovery upon re-watering as NCER resulted to be higher than the pre-stress period; however, this might also relate to early and severe basal leaf yellowing and shedding. Early deficit irrigation affected xylem characteristics of Montepulciano more than in Sangiovese; vessel density increased (37 vs 29%, respectively, compared with well-watered vines) and the hydraulic conductance decreased more (-13 vs -3% respectively) compared with well-watered vines. Yield components and technological maturity were similar in the two cultivars, whereas Montepulciano grapes had lower anthocyanins and phenolics. Higher physiological and productive efficiency under non-limiting water conditions showed by Montepulciano compared with Sangiovese was basically reversed when both cultivars were subjected to an early deficit irrigation.

Postbudburst Spur Pruning Reduces Yield and Delays Fruit Sugar Accumulation in Sangiovese in Central Italy

The influence of pruning date on yield control and ripening rate of spur-pruned Sangiovese grapevines was investigated over two years (2013 and 2014). Winter pruning was applied on 1 or 4 Feb (mid dormancy); 1 or 5 March (late dormancy); 2 or 7 April (bud swell); 2 or 7 May (flowers closely pressed together); and 1 or 6 June (40 to 50% of flower caps fallen), respectively. Vine yield and fruit composition at harvest were not affected by shifting from the standard pruning dates of mid and late dormancy to the bud swell stage. In contrast, the number of inflorescences in compound buds was significantly reduced for vines pruned in early May. No inflorescences were retained on vines pruned at the beginning of June. Early May pruning reduced fruit set and berry weight and slowed fruit ripening compared to the other pruning dates. At harvest, must soluble solids and titratable acidity were 1.6 Brix lower and 1.8 g/L higher, respectively, for the May treatment compared to the standard pruning dates. The early May pruning dates also achieved higher total anthocyanins and phenolic concentrations than the standard pruning dates, indicating that this technique can potentially decouple the accumulation dynamics of these components. Further studies are needed to better calibrate winter pruning date for managing yield and berry maturation rate.

Physiological parameters and protective energy dissipation mechanisms expressed in the leaves of two Vitis vinifera L. genotypes under multiple summer stresses

Photosynthetic performances and energy dissipation mechanisms were evaluated on the anisohydric cv. Sangiovese and on the isohydric cv. Montepulciano (Vitis vinifera L.) under conditions of multiple summer stresses. Potted vines of both cultivars were maintained at 90% and 40% of maximum water availability from fruit-set to veraison. One week before veraison, at predawn and midday, main gas-exchange and chlorophyll fluorescence parameters, chlorophyll content, xanthophyll pool and cycle and catalase activity were evaluated. Under water deficit and elevated irradiance and temperature, contrary to cv. Montepulciano and despite a significant leaf water potential decrease, Sangioveses leaves kept their stomata more open and continued to assimilate CO2 while also showing higher water use efficiency. Under these environmental conditions, in comparison with the isohydric cv. Montepulciano, the protective mechanisms of energy dissipation exerted by the anisohydric cv. Sangiovese were: (i) higher stomatal conductance and thermoregulation linked to higher transpiration rate; (ii) greater ability at dissipating more efficiently the excess energy via the xanthophylls cycle activity (thermal dissipation) due to higher VAZ pool and greater increase of de-epoxidation activity.

Malate as substrate for catabolism and gluconeogenesis during ripening in the pericarp of different grape cultivars

Malate is accumulated in grape pericarp until the start of ripening and then it is dissimilated. One aim of this study was to determine if the potential contribution of stored malate to the substrate requirements of metabolism in ripening grape pericarp is dependent on the cultivar. Two Vitis vinifera L. cultivars which accumulated different amounts of malate and had ripening periods of a different length were compared. The potential contribution of stored malate over the whole period of ripening was around 20 % in the cv. Sagrantino and 29 % in the cv. Pinot Noir. The contribution was higher in Pinot Noir because it contained more malate and had a shorter ripening period. A second aim of this study was to evaluate the contribution of gluconeogenesis to the amount of sugar accumulated in the pericarp. If all the dissimilated malate was utilized by gluconeogenesis, then the maximum contribution of stored malate to the total amount of sugar accumulated in the pericarp over the whole period of ripening was around 2.4 % in Sagrantino and 2.9 % in Pinot Noir. However, the actual contribution was only about 0.1–0.6 % in both cultivars because the majority of stored malate was not utilized by gluconeogenesis. However, it is likely that the actual contribution is much lower. This suggests that the function of gluconeogenesis is not to support accumulation of sugars in the fruits, but probably it plays other roles.

Distinct transcriptome responses to water limitation in isohydric and anisohydric grapevine cultivars

BackgroundGrapevine (Vitis vinifera L.) is an economically important crop with a wide geographical distribution, reflecting its ability to grow successfully in a range of climates. However, many vineyards are located in regions with seasonal drought, and these are often predicted to be global climate change hotspots. Climate change affects the entire physiology of grapevine, with strong effects on yield, wine quality and typicity, making it difficult to produce berries of optimal enological quality and consistent stability over the forthcoming decades.ResultsHere we investigated the reactions of two grapevine cultivars to water stress, the isohydric variety Montepulciano and the anisohydric variety Sangiovese, by examining physiological and molecular perturbations in the leaf and berry. A multidisciplinary approach was used to characterize the distinct stomatal behavior of the two cultivars and its impact on leaf and berry gene expression. Positive associations were found among the photosynthetic, physiological and transcriptional modifications, and candidate genes encoding master regulators of the water stress response were identified using an integrated approach based on the analysis of topological co-expression network properties. In particular, the genome-wide transcriptional study indicated that the isohydric behavior relies upon the following responses: i) faster transcriptome response after stress imposition; ii) faster abscisic acid-related gene modulation; iii) more rapid expression of heat shock protein (HSP) genes and iv) reversion of gene-expression profile at rewatering. Conversely, that reactive oxygen species (ROS)-scavenging enzymes, molecular chaperones and abiotic stress-related genes were induced earlier and more strongly in the anisohydric cultivar.ConclusionsOverall, the present work found original evidence of a molecular basis for the proposed classification between isohydric and anisohydric grapevine genotypes.

Double-Pruning Grapevines as a Management Tool to Delay Berry Ripening and Control Yield

Sangiovese vines mechanically spur-pruned during dormancy in February were manually finished either immediately or post budburst to test the potential of a ‘double-pruning’ approach to delay fruit sugar accumulation and limit yield. The treatments were applied in 2014, 2015, and 2016 at BBCH-0 as standard hand-finishing on dormant buds (SHF), and as late (LHF) and very late (VLHF) hand-finishing at BBCH-14 and BBCH-19, i.e., when the two apical shoots on the mechanically-shortened canes were ~10 and 20 cm long, respectively. While yield per vine was drastically reduced in the VLHF treatment (−43% versus SHF) due to high incidence of unsprouted (blind) nodes, lower shoot fruitfulness, and berries per cluster, yield reduction in LHF was −22% versus SHF due only to the incidence of unsprouted nodes. While the fruit ripening profile was not significantly modified in VLHF compared to SHF, in data pooled over three seasons, LHF delayed basic fruit composition at harvest, producing fruit with less total soluble solids, lower pH, and greater acidity, but more phenolics than SHF. Overall, LHF proved to be effective at reducing yield per vine to a level that did not require expensive cluster thinning. By reducing berry sugar accumulation, it has the potential to produce wines with lower alcohol and higher phenol content. Noteworthy too is its potential to delay harvest date or increase crop hanging time under specific vineyard conditions.

Leaf Removal and Cluster Thinning Efficiencies Are Highly Modulated by Environmental Conditions in Cool Climate Viticulture

One of several challenges in cool climate viticulture with a short growing season is to consistently reach a uniform, optimal fruit technological maturity at harvest before the first autumn frost. Weather conditions in Michigan from veraison to harvest are highly variable and unpredictable among years, constraining the preharvest assessment of fruit quality for grapegrowers and wineries. Under these environmental conditions, cluster thinning and leaf removal are commonly adopted viticultural techniques to enhance fruit ripening. Cluster thinning consists of a selective elimination of clusters to optimize the source/sink ratio of the vine. Cluster zone leaf removal induces changes in the fruit microenvironment, particularly solar radiation, temperature, and aeration. In this work, we evaluated the effects of cluster thinning and cluster zone leaf removal, applied separately in combination at veraison, on Cabernet franc in two consecutive years, 2011 and 2012. The two seasons had very distinct weather patterns from veraison to harvest. Fruit maturity was enhanced at 15 to 20 days after veraison in both years by these viticultural techniques, but with very different dynamics. The combination of leaf removal and cluster thinning led to greater fruit uniformity and better chemical composition at harvest in 2011, a year characterized by low heat accumulation after veraison. In 2012, when heat accumulation and mean temperatures after veraison were higher than in 2011, no differences were observed among treatments.

Enhancement of Fruit Technological Maturity and Alteration of the Flavonoid Metabolomic Profile in Merlot (Vitis vinifera L.) by Early Mechanical Leaf Removal

Removal of basal leaves near blooms inevitably affects grapevine balance and cluster microclimate conditions, improving fruit quality. Mechanization of this practice allows growers to save time and resources, but to our knowledge, it has not yet been compared with the manual application of this practice in a cool-climate region where seasonal temperatures frequently limit fruit technological maturity and phenolic ripening in red Vitis vinifera cultivars. In our research, berry sugar concentration was highest with prebloom mechanical treatment (PB-ME). Furthermore, metabolomics analysis revealed that PB-ME favored the accumulation of significantly more disubstituted anthocyanins and flavonols and OH-substituted anthocyanins compared with manual application. Given that vine balance was similar between treatments, increased ripening with PB-ME is likely due to enhanced microclimate conditions and higher carbon partitioning through a younger canopy containing basal leaf fragments proximal to fruit. This information provides an important strategy for consistently ripening red Vitis vinifera cultivars in cool climates.

Changes in within-shoot carbon partitioning in Pinot noir grapevines subjected to early basal leaf removal

Early leaf removal significantly alters the source-sink balance within grapevine shoots, leading to a reduction in fruit set. However, no research has previously examined the conditions controlling this process in terms of carbon allocation among major sink organs following defoliation. In this study, the impact of defoliation at bloom on the distribution dynamics of leaf assimilates among clusters and growing shoot apices was investigated on Vitis vinifera, cv. Pinot noir, grown in Michigan, a cool climate viticultural region. Three levels of defoliation: no leaves removed (LR-0); six leaves removed from six basal nodes (LR-6); and ten leaves removed from ten basal nodes (LR-10), were imposed at full bloom. A 13C pulsing was performed 1 week after the treatment application to the defoliated shoots. Single leaf gas exchange (Pn), diurnal changes of the leaf net CO2 assimilation rate, carbon distribution, fruit-set, yield, and fruit composition were measured. Higher Pn was recorded in diurnal measurements of gas exchange in leaf removal (LR) treatments compared to LR-0. The shoot apex of LR-10 experienced the highest 13C allocation (%) after 3 and 7 days following the carbon pulsing. LR-10 had lower percentage of 13C allocated to clusters, which decreased fruit set by 60%, compared to the control, and enhanced the concentration of phenolic compounds in fruit. Alteration of carbon portioning among shoot sink organs indicated that an increasing severity of leaf removal significantly reduced fruit set, and was linearly correlated to shoot apex sink strength, which occurred at the expense of the cluster.
