THE DAY-OLD CHICK: A CRUCIAL HINGE BETWEEN BREEDERS AND BROILERS

The quality of the day-old broiler chick is foremost in the minds of many poultry managers. From fertilised ovum to the placement at the broiler farm, some factors such as the characteristics of the incubating eggs, the egg holding conditions, the incubation conditions and the conditions that exist between hatching and placement at the farm and their interactions may affect chick quality. It may be questioned if maximal hatchability is the best indicator for chick quality and/or post-hatching viability, even if optimal hatchability is considered a prerequisite for successful incubation. The environmental conditions during incubation (e.g. the temperature, humidity and ventilation rate (carbon dioxide concentration)) are not set independently from each other, although each may have its own optimum for hatchability and chick quality. These optima for each incubation factor may alter differentially according to the characteristics of the incubating egg. It is concluded that more independent control as well as more control of the variability of the classical physical conditions in the incubator are required in order to improve hatchability and chick quality.

Effect of thermal conditioning during embryonic development on aspects of physiological responses of broilers to heat stress

Abstract The purpose of the present study was to investigate the effect of thermal conditioning, (through exposure to heat stress), during pre-hatch development on some physiological responses of post-hatch broilers to a post-natal heat stress challenge. Exposure to heat stress at this stage, we hope, may possibly induce epigenetic heat adaptation. Incubating eggs were exposed to temperature of 39.0°C for 2 h from Day 13 to 17 of incubation. At 33, 35, 37, 39, 41 and 43 d of age, the broilers hatched from these eggs were housed individually in open-circuit respiration cells. The climatic chambers were set to 22°C and increased to 30°C for 4 h . O2 consumption and CO2 production of each chicken was monitored continuously in order to calculate the heat production. Blood samples were obtained before and during the 4 h heat stress. Thermal conditioning during incubation did not affect the plasma T4, corticosterone, glucose, uric acid and CK concentrations. Temperature challenge, decreased plasma T3 of broilers of both groups but the decrease was greater in pre-conditioned broilers compared with controls. A similar trend was observed for triglycerides. These changes did not affect total heat production. Since decreased T3 and triglyceride levels are part of the mechanisms for thermoregulation, these suggest that thermal conditioning during incubation can improve the broiler chicken capability for thermotolerance at later post-hatch age.

Critical Assessment of Chick Quality Measurements as an Indicator of Posthatch Performance

For hatcheries, not only is it important to have a high level of hatchability, but the quality of the chicks provided also has to be good, because broiler farmers are looking for chicks with a high growth potential, resulting in a greater slaughter yield at the end of the rearing period. However, chick quality has proven to be a difficult and subjective matter to define. Therefore, the aim of this study was to investigate the predictive value of different chick quality measurements for BW at slaughter age. Body weight, chick length, shank length, and toe length measurements as well as Tona score determination were performed on 1-d-old chicks and were linked to posthatch performance parameters. Different breeder lines (Cobb and Ross) and breeder ages (39, 42, and 53 wk of age) were used to investigate line and age effects. In addition, variability between people and repeatability in time of these quality measurements were determined. Body weight at 7 d of age appeared to be the best predictor of BW at slaughter age among all the quality measurements performed. Body weight at 1 d of age had the second greatest predictive value, closely followed by the ratio between BW at 1 d of age and chick length squared. Chick length and shank length both had low to no predictive value whatsoever for posthatch performance. The lack of significant correlations between the Tona score and posthatch performance could be explained by the absence of day-old chicks with anomalies (and thus a suboptimal Tona score) because a distinction had already been made, as is done in practice, between top-grade and lower grade chicks.

Delay in feed access and spread of hatch: importance of early nutrition

In a commercial hatchery, chicks (or poults) hatch over a 24–48 hour period. All chicks remain in the incubator until the majority of the chicks have emerged from the shell. Once removed from the incubator, the newly hatched chick has to undergo several hatchery treatments and is then transported before being placed on the broiler farm. This means that, under practical conditions, chicks are deprived of feed and water for up to 72 hours. In addition, the time of hatch within the hatching window and the spread of hatch cause variability in the amount of time that chicks are feed deprived. Literature on feed deprivation after hatch clearly demonstrates the detrimental effects of any delay in feed access on performance of the chicks with respect to growth, immune system activation, digestive enzyme stimulation and organ development. Improved management strategies, such as shortening the hatching window or the time to first feeding by specific management measures, provide an alternative in dealing with the negative effects caused by a delay in feed access. The development of pre-starter diets that better meet the needs of the newly hatched chicks or in ovo feeding to bridge the gap between hatch and first feeding provide other alternatives in overcoming these problems. However, speculation remains regarding the importance of in ovo or early feeding, or whether the in ovo or early feeding itself is responsible for the beneficial effects reported. The aim of the following review is to discuss the current status of research into early feeding and to stimulate future and further research regarding these topics.

Effects of ad libitum feeding on performance of different strains of broiler breeders

Tolerance to ad libitum feeding was compared in three genotypes of broiler breeder hens: a standard broiler breeder fed ad libitum (SA) or restricted (SR), a slow growing ‘label’ broiler breeder (L) and an experimental dwarf heavy broiler breeder (E). Two similar experiments were conducted in two distinct research centres. Feed intake and body weight were measured every 3 weeks from hatch to 40 to 49 weeks of age. Egg production and egg abnormalities were recorded. The number of yellow follicles in ovaries was counted at the age of 32 weeks. Body weight was stabilised at 2·2, 3·7 and 5·4 kg after 24 weeks of age in L, E and SA hens, respectively. Growth of the SR hens was similar to that of L up to 20 weeks and stabilised at a similar level to that of E hens after 30 weeks of age. Sexual maturity was delayed by 6 weeks in restricted breeders compared to ad libitum fed hens that started to lay at 20 weeks. SA hens had low egg production and a high proportion of defective eggs, which was largely compensated for by feed restriction. However, productivity of SR hens remained lower than that of L breeders. Compared to the low viability and reproductive fitness observed with SA hens, the E dwarf broiler breeder tolerated ad libitum feeding and had better egg production, fewer egg abnormalities and yellow follicles per ovary and a higher egg production. However, laying rate was still lower than that of the SR and L groups. Energy conversion (kJ/g egg) from 32 to 40 weeks of age was much higher in the SA group than in the other three groups. The feasibility of feeding a dwarf broiler breeder ad libitum calls for further research on implications of specific IGF and GH-receptor expression at the level of the ovary in dw chickens.

Acclimation to Heat During Incubation. 1. Embryonic Morphological Traits, Blood Biochemistry, and Hatching Performance

Eggs obtained from broiler breeders at 32 (young), 42 (middle aged), and 65 wk (old) were used to measure the effects of heat acclimation during incubation on morphological, physiological, and metabolic traits at internal pipping (IP) and at hatch. All eggs were from the same stock, and hatching performance was also evaluated. Eggs from each breeder age were incubated at control (CONT) or 38.5 degrees C for 6 h daily from d 10 to 18 of incubation (HA). On d 10 after heat exposure and on d 14, absolute and proportional weights were significantly lower for HA than CONT embryos. By the time of hatching, HA chicks were heavier than CONT chicks, which suggested accelerated growth. This effect was consistent across ages. Liver and heart weights were lower for HA than CONT chicks. At IP, pH was similar for HA and CONT embryos, whereas pO(2) and Na(+) were significantly higher and pCO(2), HCO(3)-, and K(+) significantly lower for HA than CONT embryos. Blood pH was higher in embryos from older than for younger and mid-aged parents at IP. At hatch there was no effect of heat acclimation for blood HCO(3)-, Na(+), and K(+) levels, whereas plasma triglyceride and T(3) levels were higher and plasma uric acid, glucose, and lipid peroxidation levels were lower for HA than CONT chicks. Embryonic mortality was similar among parental ages for CONT. In contrast for HA, embryonic mortality from older parents was higher than for younger and middle-aged parents. A delay in external pipping and hatching time with high incubation temperature was consistent across the breeder ages. It was concluded that lower blood pCO(2), HCO(3)-, K(+), and higher pO(2) at IP stage, plus increased plasma triglyceride concentrations at hatch, indicate adaptive responses of embryos.

Vinculin and cellular retinol-binding protein-1 are markers for quiescent and activated hepatic stellate cells in formalin-fixed paraffin embedded human liver.

Hepatic stellate cells (HSCs) have important roles in the pathogenesis of liver fibrosis and cirrhosis. As response to chronic injury HSCs are activated and change from quiescent into myofibroblast-like cells. Several HSC-specific markers have been described in rat or mouse models. The aim of our work was to identify the best marker(s) for human HSCs. To this end we used the automated high throughput NexES IHC staining device (Ventana Medical Systems) to incubate sections under standardized conditions. Formalin fixed paraffin embedded (FFPE) normal and diseased human livers were studied. With immunohistochemistry we examined the expression of synemin, desmin, vimentin, vinculin, neurotrophin-3 (NT-3), α-smooth muscle actin (α-SMA), cellular retinol-binding protein-1 (CRBP-1), glial fibrillary acidic protein (GFAP), cysteine- and glycine-rich protein 2 (CRP2), and cytoglobin/stellate cell activation-associated protein (cygb/STAP). This is the first study in which a series of HSC markers is compared on serial FFPE human tissues. CRBP-1 clearly stains lobular HSCs without reacting with smooth muscle cells (SMCs) and shows variable cholangiocyte positivity. Vinculin has a similar staining pattern as CRBP-1 but additionally stains SMCs, and (myo)fibroblasts. In conclusion, we therefore propose to use CRBP-1 and/or vinculin to stain HSCs in human liver tissues.

Comparison of a modern broiler and layer strain during embryonic development and the hatching process.

1. This research focused on the embryonic development of broiler and layer embryos. 2. Egg, embryo and yolk weights were measured and partial pressure of gases in the air cell and blood were analysed at several embryonic ages. The static stiffness of the eggshell was measured before the start of incubation and at embryonic day (ED) 18 to register the change in shell strength. Times of internal pipping (IP), external pipping (EP) and hatch were recorded. Plasma corticosterone, triiodothyronine and thyroxine concentrations were determined. 3. Relative egg weight loss was higher in layer eggs. Before ED16, layer embryos showed a slower development which was reflected in lower (relative) embryo weight, lower air cell and blood pCO2 and higher air cell O2. From ED16 onwards, relative growth rate accelerated in the layer strain; as a consequence the difference in relative yolk-free chick weight at hatch had disappeared between strains. 4. Differences in physiological events necessary for hatching (thyroid hormones, corticosterone, air cell pCO2) are most probably responsible for the observed differences in timing of pipping and hatching events between layer and broilers.

Effect of chronic hypoxia during embryonic development on physiological functioning and on hatching and post-hatching parameters related to ascites syndrome in broiler chickens

The present study was designed to investigate the effect of different atmospheric pressure on the endogenous functions of broiler chickens during embryonic, hatching and growing periods related to ascites. Eggs from a commercial broiler line were incubated in two similar commercial incubators at high and low altitudes. The effect on embryonic development and physiological functions including hatching parameters, incidence of ascites and growth performance were examined. Embryos incubated at high altitude had higher plasma tri-iodothyronine, thyroxine, corticosteroid and lactic acid levels, and hatched earlier than those incubated at low altitude. Embryonic mortality was higher at high altitude. Chickens that had been incubated at high altitude showed less right ventricular hypertrophy and ascites mortality than those incubated at low altitude. It was concluded that different atmospheric pressure during incubation interacts with the endocrine functions of the embryo and hence affects hatching parameters, thereby influencing ascites susceptibility.

Acid-base balance in chicken embryos (Gallus domesticus) incubated under high CO2 concentrations during the first 10 days of incubation.

Recent studies show the importance of differential CO2 levels during the first half of incubation of chicken eggs on embryonic and postnatal growth. However, it is not known how external higher CO2 levels affect embryonic acid-base balance. In this study, the effect of an early rise in CO2, between 25th and 96th hour of incubation to 1.5% and maintained at that level until 240 h of incubation, was investigated on air cell gases, blood gas parameters from ED10 onwards and on embryonic growth and hatching parameters. Higher external CO2 concentrations resulted in a faster acidification of albumen resulting in a faster decrease of albumen pH with development, illustrating the capacity of albumen to cope with higher environmental CO2. Moreover, PCO2 in blood was higher in CO2 incubated embryos at embryonic day 10 and 11 but without a change in blood pH. The additional increase in plasma HCO3- concentration at day 10 and 11 was responsible for buffering the higher PCO2 in CO2 incubated embryos in order to stabilize pH. However, effects of hypercapnia on blood acid-base parameters extinguished 2 days after termination of high CO2 incubation. Embryonic growth was modestly accelerated which was reflected in higher embryonic weights at day 6 and 10 and a significant earlier hatching; hatchling weights were not different between treatment groups.
