Intoxicação experimental por monofluoroacetato de sódio em bovinos: aspectos clínicos e patológicos

Sodium monofluoroacetate (MF) was identified, by cromatography, in three of the 12 plants that cause sudden death in cattle in Brazil, including Palicourea marcgravii, the most important plant of this group. A special kind of hydropic-vacuolar degeneration (HVD) of the distal convoluted uriniferous tubules was considered typical for the poisoning by some authors. The objective of this study was to verify if the ingestion of MF causes similar clinical signs and lesion in cattle poisoned by plants that cause sudden death. Six cows received orally 0.5 and 1.0mg/kg of M diluted in 50mL of distilled water. Clinically the animals presented tachycardia, engorgement and positive pulse of the jugular vein, abdominal breathing, swaying gait, subtle unbalance, and recumbency with head placed the on the flank. In the agonal phase all the animals in lateral recumbence presented muscular fasciculation, tonic contraction, peddling movements, opisthotonus, nystagmus, and died between 2 and 14 minutes. At postmortem examination, the heart auricles, jugular and pulmonary veins were moderately distended and engorged with blood. Slight to moderate edema of the subserosa was seen around the gall bladder and the duodenum near to the pancreas. At light microscopy, HVD associated with nuclear picnosis of the epithelial cells of the distal convoluted uriniferous tubules was present in all animals. Ultrastructurally, the cell lesion represents formation of cytosolic vacuoles, likely due to accumulation of water. Coagulation necrosis of individual or groups of hepatocytes and slight hepatic congestion secondary to the venous stasis were also observed. Hydropic-vacuolar degeneration has been observed in cases of poisoning by many substances, which cause acute tubular nephrosis, however not restricted to the distal renal tubules and without nuclear picnosis. This study demonstrates that the HVD in the kidney can be caused by MF and, in analogy, the compound should be considered responsible for the death of cattle that ingest toxic plants which cause sudden death in Brazil. Our results can be supportive to studies that will focus on the degradation of MF by rumen bacteria, what might have economic implications, as at least 500.000 cattle die annually by sudden death causing toxic plants in Brazil.

Intoxicação por antibióticos ionóforos em animais

The therapeutic use of ionophores in veterinary medicine has grown in the last years, with resultant increase in the risk of poisoning in animals. Ionophores are used as food additives as coccidiostacts in several animal species and growth promoter and bloat prevention in ruminants. The most often used ionophores are monensin, lasalocid, narasin and salinomycin. There is a great variation in the susceptibility to the toxic effect of ionophores in different animal species. Poisoning can occur when the dosage is too high or when not correct doses for a certain animal species are given. Cases of poisoning have been described in sheep, swine, horses, dogs and poultry. For horses ionophores are extremely toxic. The use of ionophores is only safe when used accordingly to the instructions of the manufacturer and especially for each animal species. In this paper the most important data regarding clinical-pathological and pathogenic aspects, and also the conditions in which the poisoning may occur are critically reviewed.

Intoxicação experimental com as folhas de Jatropha gossypifolia (Euphorbiaceae) em ovinos

Fresh green leaves of Jatropha gossypifolia (Euphorbiaceae) were lethal for sheep in single administrations of 40g/kg. The dose of 5g/kg did not cause poisoning, but intermediate doses caused death in part of the animals. The clinical course of poisoning was 6 to 22 days. The clinic and pathological picture in the experimental sheep was characterized by digestive, lung and heart disturbances, and also by slight microscopic liver and renal regressive alterations. These findings are similar to those observed in experiments with the seeds of Jatropha curcas in goats, sheep and calves, and with the fruits and leaves of Jatropha glauca and Jatropha aceroides in goats, performed by other authors. A comparison indicates that, independently of the plant species, the leaves of Jatropha spp. contain toxic compounds similar to those found in the seeds.

Trough a critical literature review, the main data of selenium poisoning in domestic animals are presented. Epidemiological, clinical, anatomic and histopathological aspects attributed to Alkali Disease, Blind Staggers, acute poisoning and focal symmetrical poliomielomalacia of swine are discussed. The main objective of the paper is to clarify obscure points of selenium poisoning, as well to point out the risks that supplementation with this element can represent.

Intoxicação acidental por monensina em ovinos no Estado do Rio de Janeiro

An outbreak of monensin poisoning in sheep in the State of Rio de Janeiro is described. From 180 animals, eight died after they had been fed with ration containing the ionophore. The poisoning had a variable course and was clinically characterized by apathy, heart arrhythmia, myoglobinuria, incoordenation, incapacity of getting up, and sternal decubitus; one sheep aborted. The macroscopic lesions consisted of pale areas in the myocardium, hydroperitoneum, hydrothorax, and pulmonary edema. Histopathological examination revealed degenerative-necrotic alterations in heart and skeletal muscles. In the myocardium lesions were more severe and were characterized by multifocal necrosis with substitution of the myofibres by fibrous tissue and interstitial mononuclear infiltration. Proliferation of satellite cells and mononuclear inflammatory reaction in skeletal muscles were also verified. It seems that the exaggerated addition of sodic monensin, eventually associated with improper homogenization of monensin in the ration, was responsible for the excessive ingestion of monensin by some animals.

Biochemical and histological changes in liver of Nectomys squamipes naturally infected by Schistosoma mansoni.

The South American water rat Nectomys squamipes is a wild mammal reservoir of Schistosoma mansoni in Brazil. In the present study, wild rodents were collected in the field and categorized into two groups: infected and uninfected by S. mansoni. Blood was collected to analyze changes in the serum glucose level (mg/dL) and liver fragments were used to determine the hepatic glycogen content (mg of glucose/g tissue). The histological examination showed inflammatory granulomatous lesions in different phases of development in the liver of rodents naturally infected with S. mansoni, in some cases with total or partial occlusion of the vascular lumen. Early lesions were characterized by the presence of inflammatory infiltrate around morphologically intact recently deposited eggs. Despite the significance of these histological lesions, the biochemical changes differed in extent. N. squamipes naturally infected by S. mansoni showed no variation in hepatic glycogen reserves. These findings were accompanied by a significant increase in plasma glucose contents, probably as a consequence of amino acids deamination, which are degraded, resulting in the formation of intermediates used as precursors for the glucose formation, without compromising the reserves of liver glycogen. In the wild, naturally infected N. squamipes can maintain S. mansoni infections without undergoing alterations in its carbohydrate metabolism, which minimizes the deleterious effects of S. mansoni.

Avaliação lectino-histoquímica de fígado e rim de ovinos com fotossensibilização causada por Brachiaria decumbens

Inspite that Brachiaria spp. are most important forage grasses for beef cattle raising in Brazil, they can under certain conditions be toxic, cause outbreaks of hepatogenic photosensibilization and determine severe economic losses. Animals grazing toxic Brachiaria spp. commonly show in their liver foamy macrophages, isolated or grouped together, and crystals inside the biliary ducts. Steroidal saponins have been identified in these crystals and are liable for damage the liverleading to accumulation of phylloerytrin. Occasionally negative images of the crystals may be seen in the cytoplasm of foamy macrophages. The pathogenesis of formation and the type of material stored in the foamy cells is still unknown. The lectin histochemistry aims to assist in the detection of foamy macrophages and consequently in the diagnosis of the poisoning. Lectin histochemistry detects what kind of specific sugar residues exists in the cytoplasm of the foamy cells. The purpose of the present study was to identify the most suitable lectins the detection of steroidal saponins in the liver and kidney of sheep with photosensitization caused by Brachiara decumbens. Fragments of liver and kidney of fifteen sheep of both sexes and variable age, from Conceicao do Mato Dentro, Minas Gerais, southeastern Brazil, and one sheep kept free grazing Brachiaria spp. were evaluated by lectin histochemistry. Fourteen lectins were used (Con-A, DBA, SBA, PNA, SJA, RCA-I, UEA-I, WGA, SWGA, GSL, PSA, PHA-L, PHA-E and LCA). It was found that the PNA lectin detected in the liver of sheep with photosensitization, caused by ingestion of Brachiaria decumbens, showed specificity and great binding reactivity for foamy macrophages, and also specificity and slight binding reactivity for hepatocytes. The WGA lectin showed specificity and moderate binding reactivity for foamy macrophages of the liver, and also specificity and slight binding reactivity for hepatocytes. The SBA, GSL and LCA lectins showed specificity and moderate binding reactivity for foamy macrophages, but were not specific for hepatocytes. The PNA lectin showed greater binding reactivity in the kidney of the sheep. Lectin histochemistry evaluation had not yet been performed in liver and kidney of sheep with photosensitization caused by Brachiaria spp.

Aborto em bovinos devido à intoxicação por Tetrapterys acutifolia (Malpighiaceae)

Tetrapterys acutifolia Cav. (fam. Malpighiacae) stands out among the most important toxic plants of the Brazilian Southeast Region. These plants are responsible for a significant number of deaths in over 1-year-old cattle, especially in the states of Rio de Janeiro and Sao Paulo. This study aims to demonstrate that T. acutifolia is also able to induce abortion in cattle and to characterize the clinic-pathological alterations in cows and fetuses. Four cows, kept on pasture were used in the experiments which were performed on a farm in the county of Barra do Pirai, RJ, Brazil. The sprouts and young leaves of T. acutifolia were collected from the vicinity shortly before disposed in a trough. The experimental cows ingested the plant material at doses of 2.5g/kg/day (one cow), 5g/kg/day (two cows) and 10g/kg/day (one cow) until abortion occurred. All four cows aborted. The cows showed cardiac arrhythmia, muscular tremors, anorexia, ascites, distended jugular veins, edema of the sternal region and dewlap, and abortion (23 to 76 days after the beginning of ingestion of the plasnt) from the 5th to the 28th day of the experiment. Only one cow which received 10g/kg/day died with characteristic signs of heart failure, 36 days after she had aborted. Gross examination of the fetuses revealed hydrothorax, hydropericardium, hydroperitoneum, nutmeg appearance of the liver, few petechiae and ecchymoses in the epicardium, and pale areas in cross sections of the myocardium. The main postmortem findings in the cow that died were hydrothorax, hydropericardium and hydroperitoneum, engorged vessels at the heart base, exceedingly pale heart muscle with marked spots and stripes in cross sections. Distended jugular veins and severe edema in the sternal region and the abdominal wall were also observed. Histological examination of the fetal hearts revealed interstitial edema with incipient fibrosis and degenerative/necrotic changes of the myocytes. In the succumbed cow was found interstitial fibrosis, as well as necrotic areas and/or necrosis of individual myocytes in extensive portions of the heart, besides passive congestion of the liver. This study shows that Tetrapterys acutifolia can induce abortion in cattle and, depending on the dosage, can cause death.

Accidental and experimental salinomycin poisoning in rabbits

An outbreak of salinomycin poisoning in rabbits is described. At least 27 out of 2,000 rabbits reared on a farm died after the coccidiostatic drug sulfaquinoxaline was substituted by salinomycin in the feed. An average of 26.9ppm salinomycin was detected in the ration given to the rabbits. Clinical signs included anorexia, apathy and bradykinesia, which progressed to incoordination and recumbency. Gross lesions consisted of pale areas in the skeletal muscles. The histopathological findings showed severe necrotic degenerative myopathy in association with infiltration of neutrophils and macrophages. One rabbit exhibited similar alterations in the myocardium. Mineralization was observed in the affected skeletal muscles in some cases. In order to verify if the poisoning was due to salinomycin, 20 rabbits were divided into five groups and a ration containing the drug at doses of 10, 25, 50, 75 and 100ppm was given. The administration of doses higher than 50ppm resulted in manifestation of the clinical signs seen in the outbreak of poisoning. It was concluded, that probably an error related to the mixture of salinomycin in the feed was the cause of deaths in the spontaneous outbreak of poisoning on the rabbit farm.

Hipervitaminose D em animais

Through revision of the literature, data are presented about vitamin D metabolism and the toxicological, clinical, biochemical, macro and microscopic, ultrastructural, immunhistochemical and radiographic aspects in animals of different species poisoned natural and experimentally by the vitamin. We aimed to show the existence of many lacunae in the knowledge of physiological and pathological tissue mineralization, especially regarding the hormonal metabolism of vitamin D, and to alert for risk of the poisoning.
