目的nn 建立H2O2诱导视网膜色素上皮细胞ARPE-19(acute retinal pigment epithelial-19)细胞损伤模型, 研究蓝莓花青素对细胞的保护作用。 n方法nn 采用MTT法测定细胞存活率, 免疫荧光法测定细胞活性氧(reactive oxygen species, ROS)水平。以浓度0~3 200 μmol·L-1的H2O2为诱导剂, 刺激细胞2、6、24 h, 用1、5、10 mg·L-1花青素提取物(blueberry anthocyanin extract, BAE)预处理细胞6、24 h, 比较对细胞的保护作用, 确定H2O2的浓度及诱导时间。通过ARPE-19细胞损伤模型的建立, 研究蓝莓花青素对细胞的保护作用。 n结果nn 以800 μmol·L-1 H2O2刺激2 h为条件, 建立ARPE-19细胞损伤模型, 细胞存活率为63.69%。5 mg·L-1蓝莓花青素提取物、锦葵色素(malvidin, Mv)及其葡萄糖苷(malvidin-3-glucoside, Mv-3-glc)和半乳糖苷(malvidin-3-galactoside, Mv-3-gal)预处理6 h后, 细胞存活率明显提高(P n结论nn 蓝莓花青素可以有效抑制H2O2诱导产生的视网膜细胞氧化损伤, 其中主成分锦葵色素及其糖苷发挥重要作用。
