Tetrahedral DNA nanostructure-based microRNA biosensor coupled with catalytic recycling of the analyte.

MicroRNAs are not only important regulators of a wide range of cellular processes but are also identified as promising disease biomarkers. Due to the low contents in serum, microRNAs are always difficult to detect accurately . In this study, an electrochemical biosensor for ultrasensitive detection of microRNA based on tetrahedral DNA nanostructure is developed. Four DNA single strands are engineered to form a tetrahedral nanostructure with a pendant stem-loop and modified on a gold electrode surface, which largely enhances the molecular recognition efficiency. Moreover, taking advantage of strand displacement polymerization, catalytic recycling of microRNA, and silver nanoparticle-based solid-state Ag/AgCl reaction, the proposed biosensor exhibits high sensitivity with the limit of detection down to 0.4 fM. This biosensor shows great clinical value and may have practical utility in early diagnosis and prognosis of certain diseases.

A highly sensitive gold nanoparticle-based electrochemical aptasensor for theophylline detection

Theophylline is a common bronchodilator for the treatment of diseases like asthma, bronchitis and emphysema. However, it should be strictly used and monitored due to its toxicity when the concentration is above certain levels. In this work, an electrochemical biosensor for theophylline detection is proposed by recognition of RNA aptamer and gold nanoparticle (AuNP)-based amplification technique. First, RNA aptamer is splitted into two single-stranded RNA probes. One is hybridized with DNA tetrahedron and the resulted nanostructure is then immobilized onto a gold electrode; the other is modified on the surface of AuNPs which is also labeled with methylene blue (MB) as electrochemical species. The recognition process between the two RNA probes and theophylline causes the localization of AuNPs and the enrichment of MB on the electrode interface. A significant electrochemical response is thus generated which is related to the concentration of initial theophylline. This proposed aptasensor shows excellent sensitivity and selectivity which could also be applied in quantitatively detection of theophylline in serums samples.

Hydrothermal synthesis of N,S co-doped carbon nanodots for highly selective detection of living cancer cells

This study presents a facile synthesis method for the preparation of positively charged N,S co-doped carbon nanodots with excellent optical properties, and it develops a selective method for fluorescent detection of living cancer cells. The specific recognition is due to the application of an aptamer sequence, which shows high affinity and specificity to target cells. The aptamer is firstly labeled with BHQ and wraps around the carbon nanodots, then it finally quenches the fluorescence emission of the carbon nanodots. For the sensitive and selective analysis of target cells, the cells are simply mixed with the carbon nanodot-aptamer nanoconjugates, which are then centrifuged at a low speed. The recognition reaction between aptamer and target cells releases the quencher from the surface of the carbon nanodots and the centrifugation process enables the recovery of fluorescence intensity of the suspension, which reflects the level of initial cancer cells. The developed method is simple, highly selective and cost-effective, thus, it may be further exploited in clinical applications in the future.

One-pot synthesis of GSH-Capped CdTe quantum dots with excellent biocompatibility for direct cell imaging

In this work, we have developed one-pot aqueous synthesis of glutathione (GSH) binding CdTe quantum dots (QDs) for cell imaging. UV-Vis absorption spectrum, Fourier transform infrared spectrum, photoluminescence spectrum, and high-resolution transmission electron microscopy are applied to characterize the physical and chemical properties of the nanocomposites. The bioimaging efficiency of the GSH-capped CdTe QDs is further evaluated on Hela cells. The groups on the surface of QDs are able to bind to basic proteins, which are abundant in cell nuclei, enabling the application of QDs for direct cell imaging. Experimental results also indicate the GSH layer on the surface of QDs is able to reduce the cytotoxicity significantly. In conclusion, the as-prepared GSH-capped QDs are highly promising fluorescent probes for cell imaging in the near future.

Color Space Transformation-Based Smartphone Algorithm for Colorimetric Urinalysis

Urine strips are widely applied for rapid analysis of various indexes of urine for clinical examinations. The tests mainly rely on the application of a urine analyzer, which suffers several drawbacks and cannot meet the requirements of point-of-care testing (POCT). The integration of a smartphone with a biosensor has recently attracted great attention. We herein propose a human vision-based smartphone algorithm for colorimetric analysis of various urine indexes. A CIEDE2000 formula in CIELab color space is applied for the evaluation of color difference, which may greatly improve the analytical performances of urine strips. The proposed algorithm also possesses merits such as good accuracy, quantitative analysis, and limited calculation task, which is suitable for the application with smartphone platform. Experimental results demonstrate that the proposed method shows excellent reliability compared with the urine analyzer and some other algorithms. In addition, human real samples are successfully analyzed with excellent accuracy. Therefore, this work provides a convenient colorimetric tool for POCT urine analysis.
