増感作用の薬理学的研究(第5報)N, N-Dimethylethylenediamine誘導体のcatecholamine昇圧増強作用

1)N,N-dimethylethylenediamine誘導体12種のepinephrine昇圧増強作用を調べたところ,epinephrineの昇圧作用を著るしく増強する物質として, tripelennamine, N,N-dimethyl-N, N-dibenzylethylenediamine(I)およびN1,N1-dibenzyl-N2,N2-dimethyl-1,2-pro-panediamine(II)が見いだされた.2)cocaine,tripelennamine,(I)および(II)はnorepinephrineの昇圧作用も増強したが,(I)および(II)はnorepinephrineよりもepinephrineを強く増強し,tripelennamineおよびcocaineはnorepinephrineを強く増強した.3)(I)および(II)はtyramineの昇圧作用を増強したが, tripelennamineおよびcocaineはtyramineの昇圧作用を減弱した.4)(I),(II),tripelennamineおよびcocaineはisoproterenolおよびacetylcholineの降圧作用に影響を及ぼさなかった.5)histamineの降圧作用はtripelcenn-amineよって抑制きれたが,他の3つの化合物によって何ら影響を受けなかった.6)pithedratにおけるepinephrineの昇圧作用をcocaine,tripelennamine,(I)および(II)は増強した.7)reserpinized ratのepinephrineに対する感受性は未処置のものに較べて約8倍高かったが,(II),cocaineおよびtripelennamineはrescrpinized ratにおけるepinephrineの昇圧作用をさらに2倍増強した.8)後肢血管灌流実験で,(I),(II),tripelennamineおよびcocaineはepinephrine,norepinephrineの作用を増強した.しかも増強効果の絶対値は小さいながらepinephrineとnorepinephrine反応の増強作用の間には2)と同様な相対的な関係が得られた.以上の結果から,(I),(II),tripelennamineおよびcocaineは昇圧反応を増強し,その作用は末梢性であり,cocaineおよびtripelennamineの増強作用は交感神経終末におけるamine uptake機構の阻害,(I)および(II)の増強作用はそれ以外の機序によって生じると考えられる.またcocaineおよびtripelennamineの増強作用の一部としてuptake阻害以外の作用機序の関与している可能性も残されている.
