Occurrence of Galβ(1å3)GalNAc-SerThr in the linkage region of polygalactosamine containing fungal glycoprotein from Cordyceps ophioglossoides

The nitrous acid deamination liberated the N-acetyl galactosamine containing glycoproteins from galactosminoglycan (CO-N) which has been isolated from Cordyceps ophioglossoides culture. Mild alkaline borohydride degradation of the purified glycoprotein released oligosaccharide alditols. The smallest oligosaccharide alditol was characterized to be Gal beta (1----3)-GalNAcol. This result indicated that polygalactosamine part attached to protein part via Gal beta (1----3) GalNAc-Ser/Thr as the linkage region. To our knowledge, this is the first report of Gal-GalNAc-Ser/Thr linkage from fungal glycoprotein.

Abstract The structures and absolute configuration of α-kessyl alcohol (I) and kessyl glycol (II), sesquiterpenic constituents of Japanese valerian roots, have been established on the basis of chemical and spectroscopic data.

胆汁・膵液を回腸下部に分泌するように外科的手術を施して, 消化吸収能を障害させたラット (MWラット) を作製し, このラットに乳タンパク質の酵素加水分解物 (MPH) を摂取させた場合の体内利用性や栄養状態に及ぼす影響を, 基質である乳タンパク質 (MP) を対照として比較検討した。1) エネルギー効率において, MW-MPH群はMW-MP群に比べ高値を示す傾向にあった。また体重変化でも, とくに術後早期においてMW-MPH群がMW-MP群に比べ高値を示す傾向にあった。2) 生物価および窒素出納において, MW-MPH群はMW-MP群に比べ有意な高値を示した。また, 尿中3-メチルヒスチジン排泄量および3-メチルヒスチジン/クレアチニン比において, MW-MPH群はMW-MP群に比べて有意な低値を示した。3) 血清総タンパク濃度およびアルブミン濃度において, MW-MPH群はMW-MP群に比べ有意な高値を示した。以上の結果から, 乳タンパク質加水分解物であるMPHはその基質であるMPに比べ, 体内利用性をはじめとする栄養学的有効性が認められ, このことから消化管術後の消化吸収障害下で投与する窒素源として, MPHはMPに比べ有用である可能性が示唆された。
