KINETIC STUDIES ON METABOLISM OF LIPOPROTEIN LIPASE

Since Hahn demonstrated the clearing of alimentary lipemia by the intravenous injection of heparin, many excellent studies have been done on lipoprotein lipase (LPL). In these studies, however, LPL activities were compared in normal and pathological conditions at a certain time after heparin injection. If the level of LPL activity in the circulating blood changes continuously after heparin injection, under the influences of production and inactivation of LPL, it may be much more useful, under certain circumnstances, to measure the rate of change as well as the amount of such activity. One of the authors reported previously (1) the method of estimating the release or production rate and inactivation rate of LPL after heparin injection. This report will present the data on the metabolism of LPL activity in blood after the intravenous injection of heparin in normal subjects and patients with liver diseases, obesity, and coronary sclerosis, and in some animals.

SummaryUsing gastrin-like AOC-tetrapeptide the secretory function of the stomach of Japanese with endoscopically normal gastric mucosa was examined in a multicenter clinical study. Mean one hour secretion volume, mean maximal acidity, and mean maximal acid output were found to be 129.3 ±60.0 ml/hr, 103.7 ±32. 4 mEq/1, and 9.2 ±6.9 mEq/hr respectively. Results were analysed according to sex or age by analysis of variance and compared further with the corresponding data reported in the literature.

Abstract 1. 1. Molecular weight, shape, and electric charge may each play some role in determining the rate of disappearance of macromolecules from subcutaneous connective tissue. 2. 2. The state of the connective tissue, especially the volume and state of its ground substance, may modify the rate of disappearance of macromolecules from the tissue. 3. 3. There are at least three types of edema: tissue clearance of albumin was slow in lymphedema, normal in myxedema, and accelerated in other types of edema. The clearance rate was usually normal in scleroderma and rapid in hyperthyroidism and in edema of lymphosarcomatosis. 4. 4. The rate of clearance of albumin from subcutaneous tissue can be used as an indicator of lymphatic flow under usual conditions.

Collagenase-like peptidase activity in fibrotic liver tissue

Abstract Activities of an enzyme capable of digesting carbobenzoxy-glycyl-prolyl-leucyl-glycyl-proline proved to be significantly elevated in fibrotic and cirrhotic human liver tissues, as well as in experimentally induced fibrotic rat liver tissues. Hydroxyproline content was also elevated.

Dependency on Ca++ of ATP-stimulated uncoupled oxidation of succinate in rat liver mitochondria

Rat liver mitochondria depleted of endogenous substrates and high-energy phosphate bonds require exogenous ATP for the maximal uncoupled oxidation of succinate. The present experiments using a mannitol sucrose medium indicate that the effect of ATP is dependent upon the presence of low concentrations of Ca ++ (less than 5 × 10 −6 M). The requirement for Ca ++ was obvious irrespective of the presence or absence of Mg ++ , and of the species of anions (Cl − , HPO −− 4 , HAsO −− 4 , or acetate) in the buffer. Sr ++ , but not Mn ++ could replace Ca ++ . The Ca ++ -ATP effect was not obtained in the uncoupled oxidation of NAD-dependent substrates or of ascorbate in the presence of tetramethyl-P-phenylenediamine. The results of a kinetic analysis of the uncoupled succinate oxidation and of experiments with reagents which interfere with the mitochondrial Ca ++ translocation suggested that, in the presence of ATP, Ca ++ after binding on the outer surface of the inner membrane enhanced the mitochondrial succinoxidase activity by lowering the apparent Km for succinate.

Study on the gastric secretory function of Japanese with endoscopically normal gastric mucosa

According to the agreement made at the Research Committee of the Asian-Pacific Association Gastroenterology held in Tokyo in September 1966, the subcommittee for the study of gastric secretory function of normal Japanese was organized in December 1967 under the auspices of Japanese Society of Gastroenterology. To pick up cases for this study, it was agreed among members of the subcommittee that each case should be screened at least endoscopically and if possible, histologically by means of biopsy. Table 1 shows that 205 cases of endoscopically normal gastric mucosa were studied and they represent in general each age group and each sex properly except the seventh decade in female.

Prognosis of liver cirrhosis

Clinical surveys on 224 patients with liver cirrhosis were made to assess the recent advance in the treatment for liver cirrhosis. These patients consisted of those received treatment in our Department in the years from 1938 to 1965.SummaryClinical surveys on 224 patients with liver cirrhosis were made to assess the recent advance in the treatment for liver cirrhosis. These patients consisted of those received treatment in our Department in the years from 1938 to 1965.Of the principal causes of death in cirrhotics studied, hepatic coma has recently decreased and bleeding from the alimentary tract has increased. Cachexia or infection as the cause of death has also decreased.Such an improvement is likely to be attributable to introducing a high protein, high caloric diet. Alternatively, the therapeutic use of potent diuretic drugs has brought success in the treatment of ascites and edema, however, there has been found no betterment in the survival rate of cirrhotics with ascites.Among the abnormal findings which were shown in cirrhotics, icterus index more than 30, bromsulphalein retention exceeding 30 percent in 45 min, albumin/globulin ratio less than 0.5, zinc sulfate test more than 30, and reduced albumin less than 1.5 g/dl were of much omnious significance. The majority of those died within a year. Prothrombin time, total protein values, and transaminase activities in sera were not to be related to the prognosis.Most of deaths without any marked abnormalities in laboratory findings died of massive hemorrhage from the alimentary tract.

Effect of Xylitol on Pyridine Nucleotide Levels in the Liver of Rats

Recently there have been several reports that xylitol stimulates steroid synthesis (in vitro [1]) as well as reduction of methemoglobin in erythrocytes [2]. It has been assumed that these stimulations might be induced, at least in part, by altered pyridine nucleotide levels in each of the organ tissues. Actually, the conversion of xylitol to L- or D-xylulose within the cell is accompanied by the reduction of pyridine nucleotides. Therefore the effects of xylitol and other related compounds on the pyridine nucleotide levels have been studied in rat liver slices, and the results are presented in this paper.
