Stress-induced brain dysfunction and functional analysis of stress-responsible transcription factor Npas4

胎生期から思春期に至る発育環境は,統合失調症などの精神疾患の発症に影響をおよぼすことが知られている.我々は,出生直後の擬似ウイルス感染,離乳後からの長期隔離飼育あるいは慢性拘束ストレスにより,成熟後のマウスの学習・記憶や情動行動に障害が生じ,海馬における神経新生にも異常が生じることを明らかにした.また,幼若期長期隔離飼育による学習・記憶障害や海馬における神経新生の異常に対して,フルオキセチンは改善効果を示した.ストレス誘発性脳機能障害に関連する遺伝子を同定するため,ストレスを負荷した動物の海馬における遺伝子発現を網羅的に解析した.その結果,離乳直後から4週間の隔離飼育ストレス負荷を行ったマウスではneuronal PAS domain 4(Npas4)およびnuclear receptor subfamily 4,group A,member 2(Nurr1)の遺伝子発現が有意に低下していることを見出した.ICR系マウスの海馬におけるNpas4 mRNAレベルはコルチコステロン投与により有意に低下し,副腎摘出処置により増加した.同様に,株化神経細胞をコルチコステロンで処理するとNpas4タンパク質の発現レベルは有意に減少した.Npas4過剰発現細胞では神経突起の伸展が促進され,リン酸化synapsin Iの増加およびCdk5タンパク質の発現増加が認められた.Neuo2A細胞におけるリン酸化synapsin Iの増加はCdk5阻害薬により拮抗された.これらの結果は,幼若期ストレス負荷は成熟後の脳機能障害を誘発し,その分子機序の一部としてNpas4が重要な役割を果たしている可能性を示唆している.

Transcriptional regulation of neuronal PAS domain 4 (NPAS4) by stress

Fluorescent In situ Hybridization (FISH) analysis is a powerful tool used in karyotyping, cytogenotyping, cancer diagnosis, species specification, and gene expression analysis. Although widely used, conventional FISH methods are cumbersome and time consuming. We have now developed a highresolution FISH protocol using exciton-controlled hybridization-sensitive fluorescent oligonucleotide (ECHO) probes. The new ECHO-FISH protocol takes 25 min from fixation to mounting, is highly reproducible, quantitative, and compatible with other fluorescent cellular labeling techniques. Using ECHO-FISH, we demonstrate detection of telomeric and centromeric DNA on metaphase mouse chromosomes. We also demonstrate detection of changes in poly (A)+ RNA concentration in both soma and dendrites of cultured hippocampal neurons that correspond to neuronal activity. The simplicity of ECHO-FISH method will facilitate quantitative genome-wide cytogenetic and gene expression analysis. Research fund: JSPS Grant-in-aid 22-00212.
