Differences in biological and behavioural characteristics of Harmonia axyridis (Coleoptera: Coccinellidae) according to colour patterns of elytra

Samples of overwintering populations of Harmonia axyridis were classified into five colour patterns, succinea 1, succinea 2, conspicua, spectabilis and axyridis. Generally, the succinea 1 colour pattern was dominant at six collecting sites in Daejon and Chungnam provinces of Korea and their occurrence proportion was over 50%. Non‐melanic forms such as succinea 1 and 2 were noted in about 90% of collected individuals. On the other hand, conspicua and spectabilis as melanic forms were dominant in the laboratory. One hypothesis is that the difference between field and laboratory populations is related to non‐random mating and environmental conditions. Practically, in mate‐choice experiments, most colour pattern ladybirds seemed to mate with the melanic form, especially the conspicua form. The body size (length) of H. axyridis in colour patterns was slightly different, but in females, there was no significant difference among colour patterns. However, both lengths of males were significantly different among colour patterns. In reproductive ability (fecundity), there was no difference among colour patterns.

Correlation of an Electrical Penetration Graph Waveform with Walking by Asian Citrus Psyllid, Diaphorina citri (Hemiptera: Psyllidae)

Electrical penetration graph (EPG) monitoring of insect feeding is a rigorous means of observing and quantifying the otherwise invisible feeding of piercing-sucking arthropods (Walker 2000). EPG is being used to study how the feeding behavior of the Asian citrus psyllid (ACP) (Diaphorina citri Kuwayama) changes in response to insecticide residues (Serikawa et al. 2010; Serikawa et al. in press), in an effort to improve the efficacy of wide spread insecticide treatments against this insect in Florida citrus. ACP is the vector of the putative causal agent of Huanglongbing (HLB) disease, Candidatus Liberibacter asiaticus, the cause of millions of dollars in damage to the citrus indus try in Florida (Muraro & Morris 2009). The pri mary means of disease management at this time is insecticide treatment of vectors; however, it is presently unknown whether insecticides prevent performance of the feeding behaviors of ACP re quired for pathogen transmission. Seasonal Occurrence, Development and Preference of Riptortus pedestris on Hairy Vetch

The seasonal occurrence of Riptortus pedestris Fabricius was investigated using pheromone traps baited with its aggregation pheromone in the fields with various crop composition (mixture culture of barley and hairy vetch in Asan, Chungnam; monoculture of hairy vetch in Yuseung, Daejeon) from mid-May to mid-July of seed gathering season in 2010. The invasion of R. pedestris into the experimental fields began to increase rapidly from late-June in which period hairy vetch and barley reach the beginning of seed formation stage. After seed gathering season in mid-July, the invasion rate of R. pedestris decreased, and the activity was continually observed till early-November. In addition to dominant R. pedestris species, 33 species of hemiptera including Apolyaus watajii, Dolycoris baccarum, Adelphocoris suturalis, and Yemma exlis were collected, which indicated abundant species diversity in the hairy vetch fields. In the laboratory, R. pedestris did not developed successfully to adult stage on food sources of hairy vetch, with decreasing survival rate after 4th instar and the failure of emergence to adult stage. Also, R. pedestris showed higher preference on soybeans than hairy vetch. Consequently, hairy vetch may be not true host for the development and survival of R. pedestris. It is considered that R. pedestris is a temporary visitor at the season of seed formation in hairy vetch fields.

Seasonal Fluctuation of Riptortus pedestris (Hemiptera: Alydidae) in Chungbuk Province

Seasonal fluctuations of Riptortus pedestris were investigated in four regions including two sites each at Mt. Yangseong (Munui-myeon, Cheongwon-gun), O-chang (Cheongwon-gun), and Jujung-dong (Cheongju) using aggression pheromone traps from April to November in 2010 and 2011. Aggression pheromone and aggression pheromone + soybean traps were set at all investigated sites, and the Mt. Yangseong A and B sites were investigated at a farmland (80 m, asl) and forest (200 and 300 m). The population density of R. pedestris was high in mid June, mid August, and late October in 2010 and in early May, mid June, mid September, and early October in 2011 with trivoltine. O-chang and Jujung-dong populations, which were distinguished in farmlands and forests, were highest from June to August in the farmland and in September in the forest. Similar numbers of R. pedestris were capture in the farmlands and the forest in June-August, September-November, respectively. From the results of the four regions, more R. pedestris adults were captured in the aggression pheromone + soybean trap than that in the pheromone trap. To investigate the migration route by altitude, 500 R. pedestris adults marked with fluorescent paint were released and re-caught insects were counted in traps after 10 and 20 days. The pattern of the re-caught R. pedestris indicated migration from the forest to farmlands during April-June. These results suggest that the insects did not migrate in August because food was plentiful in the forest at 200 m, but they moved to the forest during October due to the scarcity of food and for overwintering. The R. pedestris seasonal fluctuations in 2011 were affected heavily by the environment, particularly rain precipitation.

Effects of Some Pesticides on Development of Ascaris suum Eggs

To evaluate the effects of pesticides to parasite eggs, Ascaris suum eggs were incubated with 5 different pesticides (1:1,500-1:2,000 dilutions of 2% emamectin benzoate, 5% spinetoram, 5% indoxacarb, 1% deltamethrin, and 5% flufenoxuron; all v/v) at 20℃ for 6 weeks, and microscopically evaluated the egg survival and development on a weekly basis. The survival rate of A. suum eggs incubated in normal saline (control eggs) was 90±3% at 6 weeks. However, the survival rates of eggs treated with pesticides were 75-85% at this time, thus significantly lower than the control value. Larval development in control eggs commenced at 3 weeks, and 73±3% of eggs had internal larvae at 6 weeks. Larvae were evident in pesticide-treated eggs at 3-4 weeks, and the proportions of eggs carrying larvae at 6 weeks (36±3%-54±3%) were significantly lower than that of the control group. Thus, pesticides tested at levels similar to those used in agricultural practices exhibited low-level ovicidal activity and delayed embryogenesis of A. suum eggs, although some differences were evident among the tested pesticides.

Characteristics of feeding behaviors of Myzus persicae (Hemiptera: Aphididae) depending on inflow concentrations of dinotefuran.

The changes of feeding pattern and the amount of insecticide penetrated into the leaf were monitored for 420 min after treatment of three concentrations of dinotefuran. At about 30 min after treatment of insecticide, the overall feeding pattern in EPG (Electrical Penetration Graph) monitoring was started to change, and there was significant difference in proportions of NP (non-penetration), PA (pathway activity), and phloem-feeding patterns among the different concentrations of dinotefuran treatment. Especially, as the amount of insecticide penetrated into a leaf increased, the reactive behaviors against this insecticide such as withdrawal of proboscis and movements of stylet of aphid were more quickly exhibited. And also, total time at which the proboscis of the aphids did not penetrate the plant was getting longer. The amount of dinotefuran penetrated into a leaf was monitored with HPLC. When the recommended concentration (100 mg/L) of the insecticide was treated, 5.247.24 mg/L of the insecticide was detected from the leaf, and the proboscis of the aphids was apparently withdrawn from the leaf at approximately 60 minutes after treatment of this insecticide.

Differentiation in Feeding Behaviour of Biotypes B and Q of Bemisia tabaci (Homoptera: Aleyrodidae) against Three Insecticides

The feeding behaviors of 2 biotypes (type B and Q) of tobacco whitefly, Bemisia tobaci, were monitored using EPG technique on tomato and pepper plants treated 3 insecticides for controlling whiteflies, for examples, acetamiprid, spinosad and thiamethoxam. After treatment of three insecticides with recommended concentrations to tomato and pepper plants, EPG waveforms were recorded during 6 hours. The characteristic patterns of feeding behaviors investigated were as follows; time consumed by withdrawal of proboscis, total non-penetration time, total stylet pathway pattern time and total phloem feeding time. There was somewhat difference among 3 insecticides tested. As a result of investigation of total duration showed the stylet pathway activity due to the reaction against all tested insecticides, the Q biotype showed fewer time than those from the B biotype. The B biotype showed more frequent stylet pathway activity patterns during whole recording time and a shorter phloem ingestion time than those from the Q biotype. In result of prior up (non-penetration) time representing the reaction against the insecticide treated, the time of B biotype was more faster than that of the Q biotype, so it was considered that the B biotype was more sensitive to the tested insecticides. Therefore, our results revealed a clear difference in feeding behaviour between the B and Q biotypes of B. tabaci. Also, it was investigated that B biotype was susceptible to the 3 insecticides.

Quantitative evaluation of viability- and apoptosis-related genes in Ascaris suum eggs under different culture-temperature conditions.

Ascaris suum eggs are inactivated by composting conditions; however, it is difficult to find functional changes in heat-treated A. suum eggs. Here, unembryonated A. suum eggs were incubated at 20℃, 50℃, and 70℃ in vitro, and the gene expression levels related to viability, such as eukaryotic translation initiation factor 4E (IF4E), phosphofructokinase 1 (PFK1), and thioredoxin 1 (TRX1), and to apoptosis, such as apoptosis-inducing factor 1 (AIF1) and cell death protein 6 (CDP6), were evaluated by real-time quantitative RT-PCR. No prominent morphological alterations were noted in the eggs at 20℃ until day 10. In contrast, the eggs developed rapidly, and embryonated eggs and hatched larvae began to die, starting on day 2 at 50℃ and day 1 at 70℃. At 20℃, IF4E, PFK1, and TRX1 mRNA expression was significantly increased from days 2-4; however, AIF1 and CDP6 mRNA expression was not changed significantly. IF4E, PFK1, and TRX1 mRNA expression was markedly decreased from day 2 at 50℃ and 70℃, whereas AIF1 and CDP6 mRNA expression was significantly increased. The expressions of HSP70 and HSP90 were detected for 9-10 days at 20℃, for 3-5 days at 50℃, and for 2 days at 70℃. Taken together, incremental heat increases were associated with the rapid development of A. suum eggs, decreased expression of genes related to viability, and earlier expression of apoptosis-related genes, and finally these changes of viability- and apoptosis-related genes of A. suum eggs were associated with survival of the eggs under temperature stress.

Assessment of the Microbiological Quality of Vegetable from Urban Community Gardens in Korea

Many community gardens in large cities worldwide grow vegetables; however, no information regarding the levels of sanitary indicator bacteria and prevalence of foodborne pathogens in vegetables grown in urban community gardens is available. To evaluate the microbiological quality of vegetables from urban community gardens in Korea, 530 samples (nine types of vegetable, including Chinese cabbage, lettuce, radish leaves, spinach, mustard leaves, crown daisy, leek, Korean cabbage, and chicory) were collected at 11 urban community gardens in Seoul, Korea from September through October 2012. The levels of total aerobic bacteria, Escherichia coli, total coliforms, Salmonella spp. Listeria monocytogenes, and E. coli O157:H7 were evaluated quantitatively and/or qualitatively. The mean numbers of total aerobic bacteria and coliforms were 6.3 log CFU/g (range 3.8-8.1 log CFU/g) and 4.3 log CFU/g (range 2.1-6.4 log CFU/g), respectively. Total coliforms were detected on 67% of whole vegetables. Chicory showed the highest number of total aerobic bacteria and coliforms, whereas the lowest number of coliforms was detected on leeks. E. coli was detected on 2.3% of whole vegetables, including lettuce, radish leaves, mustard leaves, and chicory; however, foodborne pathogenic bacteria were not detected on any of the vegetable samples using this highly sensitive and validated procedure. Based on these findings, the presence of coliforms and E. coli demonstrates that opportunity for improvement of microbiological safety exists throughout the produce production chain, although no major foodborne pathogens were present in vegetables grown in urban community gardens.

Comparison of Life Table and Feeding Behavior of Resistance and Susceptible Population of the Green Peach Aphid, Myzus persicae (Hemiptera: Aphididae) against Etofenprox

For the comparing of mortality of the resistance and susceptible population of Myzus persicae, etofenprox was treated in the recommended concentration of 200ppm. Mortalities of resistance population were 16.7 and 36.7%, and susceptible population were 86.7 and 86.7% after 24 and 48 hours treatment, respectively. For the detect of cross resistance to other pyrethroids, 6 pyrethroids were examined. Mortalities of susceptible and resistance populations were 90 and 31% to deltamethrin, 92 and 23% to lambda cyhalothrin, 81 and 14% to cypermethrin, 70 and 20% to -cypermethrin, 29 and 28% to fenpropathrin, 84 and 29% to fenvalerate, respectively. It was showed that resistance populations were generally resistive to other pyrethroids. On the other hands, for recognized ecological characteristic of M. persicae susceptible and resistance populations life table was tested on the pepper leaves in the petri dish and on the plant in the pot. This results were showed that intrinsic rate of increase (), net reproduction number () and generation time in day () were significantly different between two population in both tested. However, life span and reproduction period were slightly different between them. Otherwise, feeding behaviors were tested using EPG technique with non- and treated etofenprox. First potential drop time of susceptible and resistance population was 73.5 and 257.9 sec with non-treated and 93.3 and 1076.2 sec after treated, respectively. Electrical probing signals were 8.2 and 48.8 times with susceptible and resistance individuals after treated etofenprox, respectively. It was supposed that the resistance is more probings than susceptible population. After treated, total feeding time have more 6,728.9 sec on resistance than 965.5 sec on susceptible population. So, total non penetration time of susceptible population was 3,000 sec longer than resistance population.
