Integrated Microsystem of Isothermal Amplification of DNA and Electrophoresis on a Microfabricated Plastic Chip for Detection of Specific Gene and Analysis of Genetic Materials

We have successfully realized the on-chip integration of isothermal polymerase chain reaction (PCR) and the subsequent analysis of the amplification products by electrophoresis. In our experiments, loop-mediated isothermal amplification (LAMP) has been adopted, which can not only avoid the thermal cycling, but also offer higher specific amplification than traditional PCR. After the amplification, the products could be either analyzed by the integrated microchip electrophoresis or directly observed by the naked eyes by adding some fluorescent dye like SYBR Green I. This integrated microsystem has proven extremely useful for the fast, sensitive, and simple detection of some specific sequences of DNA.
