Plasmid-mediated control of nodulation in Rhizobium trifolii

The nodulation ability was effectively eliminated from different Rhizobium trifolii strains incubated at elevated temperature (Żurkowski and Lorkiewicz, 1978). Non-nodulating (Nod-) mutants were stable and no reversion of Nod- to Nod+ phenotype was observed. Strains R. trifolii 24 and T12 which showed a high percentage of elimination of nodulation ability were examined in detail. Two plasmids were detected in strain 24 using neutral and alkaline sucrose gradient centrifugation of plasmid preparations. Molecular weights of the plasmids pWZ1 and pWZ2 were 460 Mdal and 190 Mdal, respectively. Rhizobium lysates labeled with 3H-thymidine and ultracentrifuged in caesium chloride — ethidium bromide gradients demonstrated a 40% reduction of the plasmid DNA content in R. trifolii 24 Nod- mutants in comparison with the nodulating wild type strain 24. It was found further that non-nodulation of mutants 24 Nod- was due to the absence of plasmid pWZ2. Sucrose gradient data also demonstrated that strain T12 contained two plasmids with molecular weights corresponding to those of pWZ1 and pWZ2, respectively. In Nod- mutant clones derived from strain T12, pWZ2 plasmid was missing.

Chemical analysis of Azospirillum lipopolysaccharides

Lipopolysaccharides (LPS) were extracted by hot phenol-water from five strains each of Azospirillum lipoferum and Azospirillum brasilense. Rhamnose, glucose, glucosamine and 3-deoxy-d-mannooctulosonic acid were comon sugar constituents of all LPS preparations. 2-O-Mefucose, 3-O-Me-fucose, 3-O-Me-rhamnose and 2-O-Megalactose were found in LPSs of some A. brasilense strains. Fatty acid spectra from all LPSs studied were almost identical with predominance of 3-hydroxymyristic and 3-hydroxypalmitic acids. 3-Hydroxypalmitic acid was the only amide-linked fatty acid. Lipopolysaccharides isolated from A. brasilense showed higher heterogeneity in sugar composition than those from A. lipoferum.

Siderophore production and utilization byRhizobium trifolii

SummarySeveral strains ofRhizobium trifolii were tested for their ability to synthesize and utilize phenolate or hydroxamate types of siderophores. None of the nodulating strains ofR. trifolii was able to produce detectable amounts of siderophores. Only the non-nodulating strainR. trifolii AR6 formed a phenolate siderophore, which stimulated the growth of the siderophore-negative mutant AR65. Other strains ofR. trifolii could not utilize iron from exogenously supplied Desferal, pseudobactin or citrate. The siderophore fromR. trifolii AR6 and 2,3-dihydroxybenzoic acid slightly stimulated the growth of someR. trifolii strains.

Nif plasmid from Lignobacter

Lignobacter strain K17 is able to degrade aromatic compounds and to fix atmospheric nitrogen. It was proved that capacity for nitrogen fixation by Lignobacter was plasmid mediated. Plasmid pUCS100 (17.5 Mdal) carrying nif genes was transferred from Lignobacter to Escherichia coli SK1592 and Salmonella typhimurium. The transposon Tn9 was translocated to pUCS100 to facilitate selection of Nif+ bacteria. E. coli SK1592 harboring the new plasmid (pUCS101) reduced acetylene under anaerobic conditions. Plasmids pUCS100 and pUCS101 were not stably maintained in E. coli and S. typhimurium.

Plasmid-determined Alterations of Salmonella typhimurium Lipopolysaccharides

SummarySalmonella typhimurium Rc902 infected with derepressed ColIb mutants gave rise to changes in the composition of bacterial lipopolysaccharides (LPS). Bacteria carrying ColIbdrd7, derepressed in transfer, exhibited a marked decrease in the content of all 0-side-chain sugars of LPS. Similar effects were found upon the introduction of R64-11, also derepressed in transfer. In LPS of S. typhimurium containing ColIbdrd2, derepressed in colicin synthesis, a decrease of abequose content associated with an increase of glucose level was observed. Bacteria carrying the wild-type ColIb, the revertant of a drd mutant to the wild type, or the non colicinogenic strain resulting from the elimination of ColIbdrd2, showed no changes in the sugar composition of LPS.

Transfer of nodulation ability in Rhizobium using R68.45 derived plasmids

SummaryNodulation ability was transferred from Rhizobium meliloti L5.30 to the non-nodulating mutant Rhizobium trifolii 24K using plasmid R68.45. Transconjugants selected for the carbenicillin resistance (cbr) marker became simultaneously capable of nodulating clover and showed changes in phage sensitivity. Besides the indigenous plasmid of 90 MD (pUCS201), the nodulating transconjugants harbored the newly introduced plasmid pUCS202 (ca. 40 MD). After treatment of the transconjugants with curing agents the simultaneous loss of antibiotic resistance and ability to form nodules were associated with the disappearance of pUCS202. nod and cbr genes were cotransferred into R. trifolii strains by conjugation and transformation. There is genetic evidence that the nod gene(s) was integrated into R68.45.

Role of noncarbohydrate substitutions of Rhizobium exopolysaccharide in nodulation process

Exopolysaccharides (EPS) of nodulating strains of Rhizobium trifolii and Rhizobium leguminosarum added to red clover seedlings before inoculation reduced the number of nodules. The inhibition of the nodulation was correlated with the amount of EPS. The preparations of EPS from mutants defective in early stages of nodulation (Roa- or Hac-) did not affect the nodulation, whereas EPS from mutants deficient in late stages (post Hac-) exerted an inhibitory effect.Inactive preparation of EPS contained less O-acetyl groups and pyruvic acid residues. Deacetylation and depyruvylation of EPS from R. trifolii Nod+ abolished it inhibitory effect. It was concluded that noncarbohydrate substitutions (acetate, pyruvate) are involved in EPS effect.

Restriction enzyme analysis of the Plasmid ColIb DNA

SummaryPlasmid ColIb (61.5 Mdal) was digested with restriction enzymes EcoRI and HindIII. The DNA digestion products were separated by electrophoresis on 1.2% agarose gels. There were identified 22 fragments of ColIb DNA generated by the endonuclease EcoRI and 21 fragments produced by HindIII. Molecular weights of the fragments were estimated. The total molecular weight of the fragments generated by EcoRI was 61.42 Mdal and for HindIII fragments 62.79 Mdal.

Detection of small cryptic plasmids in Salmonella typhimurium strain LT2

Small cryptic plasmids of molecular weights ranging from 1 to 3 Mdal were detected by electron microscopy in Salmonella typhimurium strain LT2 (ColIb). They were divided into different size classes. Two of the cryptic plasmids were transferred simultaneously with ColIb to Escherichia coli.

Bidirectional Replication of the Chromosome in Rhizobium trifolii

SummaryCulture of the thymine-dependent mutant of Rhizobium trifolii T37 was synchronized with phenethanol. During bacterial synchronous growth with synchronized replication of DNA, cells were differentially labeled using subsequently 3H-thymidine of low and high specific activity. The grain tracks produced in autoradiographs of chromosomes were denser on both ends than in the middle. In control experiments with bacteria labeled only with 3H-thymidine of low specific activity, the grain density was uniform throughout the grain track. The results constitute clear evidence of bidirectional replication of R. trifolli chromosome.
