Antiviral flavonoids from the root bark of Morus alba L.

A prenylated flavonoid, moralbanone, along with seven known compounds kuwanon S, mulberroside C, cyclomorusin, eudraflavone B hydroperoxide, oxydihydromorusin, leachianone G and alpha-acetyl-amyrin were isolated from the root bark of Morus alba L. Leachianone G showed potent antiviral activity (IC(50) = 1.6 microg/ml), whereas mulberroside C showed weak activity (IC(50) = 75.4 microg/ml) against herpes simplex type 1 virus (HSV-1). Their structures were elucidated by spectroscopic methods.

Anti-oxidative, anti-inflammatory and hepato-protective effects of Ligustrum robustum

Aqueous extract of processed leaves of Ligustrum robustum could dose-dependently scavenge superoxide radicals, inhibit lipid peroxidation, and prevent AAPH-induced hemolysis of red blood cells. In comparison with green tea, oolong tea and black tea, processed leaves of L. robustum exhibited comparable antioxidant potency in scavenging superoxide radicals and in preventing red blood cell hemolysis. By activity-guided fractionation, a glycoside-rich fraction named fraction B2 was separated and demonstrated to possess strong antioxidant effect. It was evaluated for its anti-inflammatory and hepato-protective activities. A single oral dose of fraction B2 at 0.5 g/kg could provide 51.5% inhibition on the vascular permeability change induced by intraperitoneal injection of acetic acid, but it could not inhibit croton oil-induced ear edema. On the other hand, fraction B2 exhibited moderate hepato-protective effect. Intragastric application of fraction B2 at 1.25, 2.5 or 5 g/kg 6 h after carbon tetrachloride administration could reduce the elevations of serum levels of aminotransferases (AST and ALT). Also, liver integrity was preserved, as liver sections from rats post-treated with fraction B2 showed a milder degree of fatty accumulation and necrosis. These results offer partial support to the traditional uses of the leaves of L. robustum as Ku-Ding-Cha.

Secoiridoid constituents from the fruits of Ligustrum lucidum

Two secoiridoid glucosides, lucidumosides A and B, as well as six known glucosides, oleoside dimethyl ester, ligustroside, oleuropein, nuezhenide, isonuezhenide, and neonuezhenide, were isolated from the fruits of Ligustrum lucidum. Their structures were elucidated by spectroscopic methods.

Molecular Structures and Antiviral Activities of Naturally Occurring and Modified Cassane Furanoditerpenoids and Friedelane Triterpenoids from Caesalpinia minax

Further investigation of the active components of the chloroform fraction of the seeds of Caesalpinia minax led to the isolation of a new cassane furanoditerpenoid, caesalmin H (1), together with two known furanoditerpenoid lactones, caesalmin B (2) and bonducellpin D (3). Reduction of the naturally abundant caesalmin D (9), E (10) and F (11) resulted in three new furanoditerpenoid derivatives 4-6. Phytochemical study of the stem of the same plant and subsequent reduction afforded two friedelane triterpenoids (7-8), which were identified by spectroscopic methods. Compounds 1-2 and 4-8 were corroborated by single crystal X-ray analysis. The factors governing the reduction of cassane furanoditerpenoids and friedelane triterpenoids were investigated by correlating the crystallographic results with density functional theory. The inhibitory activities of 2-8 on the Para3 virus were evaluated by cytopathogenic effects (CPE) reduction assay.

Antiviral constituents against respiratory viruses from Mikania micrantha.

Phytochemical investigation of the dried aerial parts of Mikania micrantha led to the isolation of a new sesquiterpene, 3beta-acetoxy-1,10-epoxy-4-germacrene-12,8;15,6-diolide (1), along with six known constituents: 1,10-epoxy-4-germacrene-12,8;15,6-diolide (2), dihydromikanolide (3), potassium mikanin 3-sulfate (4), mikanin (5), alpinetin (6), and ergosta-7,22-dien-3beta-ol (7). Their structures were elucidated by spectroscopic methods, and the molecular structures and stereochemistry of sesquiterpene lactones 1-3 were revealed by single-crystal X-ray analysis. Compound 2 showed moderate activity against respiratory syncytial virus (IC(50) = 37.4 uM) and parainfluenza type 3 virus (IC(50) = 37.4 uM) with a therapeutic index (TI) of 16.0 for both compounds. Compound 4, the main component of M. micrantha, exhibited inhibitory activity against parainfluenza type 3 virus with IC(50) (19.7 uM) and TI (24.0) values comparable to those of ribavirin, serving as a positive control.
