Comparison of allele-specific PCR, created restriction-site PCR, and PCR with primer-introduced restriction analysis methods used for screening complex vertebral malformation carriers in Holstein cattle

Complex vertebral malformation (CVM) is an inherited, autosomal recessive disorder of Holstein cattle. The aim of this study was to compare sensitivity, specificity, positive and negative predictive values, accuracy, and rapidity of allele-specific polymerase chain reaction (AS-PCR), created restriction-site PCR (CRS-PCR), and PCR with primer-introduced restriction analysis (PCR-PIRA), three methods used in identification of CVM carriers in a Holstein cattle population. In order to screen for the G>T mutation in the solute carrier family 35 member A3 (SLC35A3) gene, DNA sequencing as the gold standard method was used. The prevalence of carriers and the mutant allele frequency were 3.2% and 0.016, respectively, among Holstein cattle in the Thrace region of Turkey. Among the three methods, the fastest but least accurate was AS-PCR. Although the rapidity of CRS-PCR and PCR-PIRA were nearly equal, the accuracy of PCR-PIRA was higher than that of CRS-PCR. Therefore, among the three methods, PCR-PIRA appears to be the most efficacious for screening of mutant alleles when identifying CVM carriers in a Holstein cattle population.

KESİM KUZUSU ELDE ETMEK AMACIYLA ALMAN SİYAH BAŞLI ETÇİ KOÇLARLA BİRLEŞTİRİLEN SAKIZ x KIVIRCIK (F1) KOYUNLAR İLE KIVIRCIK VE TÜRK MERİNOSU KOYUNLARIN DÖLVERİMİ VE SÜT VERİMİ ÖZELLİKLERİ

Ozet Bu arastirma Kivircik koyunlarin, dolverimi ve sut verimi ozellikleri yonunden gelistirilmesi icin Sakiz koclarla melezlenmesi ile elde edilen F ] koyunlarin, ayni surude yetistirilen Kivircik ve Turk Merinosu koyunlar ile dolverimi ve sut verimi ozellikleri yonunden karsilastirmali olarak degerlendirilmesi amaciyla yurutulmustur. Iki yetistirme doneminde izlenen dolverimi ozelliklerinden dogum orani ve bir dogumdaki kuzu sayisi ortalamalari Sakiz x Kivircik (F 1 ) koyunlar icin %69.05 ve 1,31, Kivircik koyunlar icin %72.63 ve 1.40, Turk Merinosu koyunlar icin ise %81,41 ve 1.36 bulunmustur. Sut verimi ve laktasyon suresi F 1 koyunlar icin 85.61 kg ve 137 gun, Kivircik koyunlar icin 82.85 kg ve 135 gun ve Turk Merinosu koyunlar icin 97.02 kg ve 141 gun olarak saptanmistir. Arastirma bulgulari degerlendirildiginde, erken yaslardaki Sakiz x Kivircik (F 1 ) koyunlarin bir doguma dusen kuzu sayisi ve sut verimi yonunden ayni yastaki Kivircik koyunlardan yuksek ve Turk Merinosu koyunlarla benzer duzeylerde oldugu sonucuna varilmistir. Genel suru ortalamalari yonunden ise erken yaslardaki F 1 koyunlarin Kivircik ve Turk Merinosu koyunlarla benzer sonuclar verdigi belirlenmistir. Anahtar Kelimeler: Sakiz. THE FERTILITY AND MILK PRODUCTION TRAITS OF CHIOS X KIVIRCIK (F 1 ) EWES MATED WITH GERMAN BLACK-HEADED MUTTON RAMS TO PRODUCE CROSSBRED SLAUGHTER LAMBS, IN COMPARISON WITH KIVIRCIK AND TURKISH MERINO EWES Summary This study was carried out to investigate the fertility and milk production characteristics of F 1 ewes, which were produced by the mating of Kivircik ewes with Chios rams, in comparison with purebred Kivircik and Turkish Merino ewes. The rate of ewes lambing and litter size were 69.05% and 1.31 for Sakiz x Kivircik (F 1 ) ewes, 72.63% and 1.40 for Kivircik ewes and 81.41% and 1.36 for Turkish Merino ewes. The milk production and lactation lengths determined in this study for Sakiz x Kivircik (F 1 ), Kivircik and Turkish Merino ewes were 85.61 kg and 137 days, 82.85 kg and 135 days and 97.02 kg and 141 days, respectively. When the results of this study are evaluated, it was concluded that in terms of the litter size and milk yield the Sakiz x Kivircik (F,) ewes at early ages were higher than the Kivircik ewes at the same ages and were similar to Turkish Merino ewes. With regard to the general flock averages the F 1 ewes at early ages were found to be similar to Kivircik and Turkish Merino ewes. BROİLER ÜRETİMİNDE ZEOLİTİN ALTLIK MATERYALİ OLARAK KULLANILMA OLANAKLARI

Ozet Broiler yetistiriciliginde altliga zeolit ilavesinin, altlik kalitesi uzerine etkileri ve ayni altligin yeniden kullanilabilirliginin arastirildigi bu calismada; buyume hizi, yasama gucu, yemden yararlanma, verim indeksi degerleri ile altlik nemi, pHsi, amonyak duzeyi ve total bakteri sayisi uzerindeki etkileri incelenmistir-Altliga zeolit ilavesinin ilk uretim doneminde buyume hizi uzerine olumlu bir etki sagladigi, aynca zeolitli ayni altligin yeniden kullaniminin incelenen ozellikler uzerine olumsuz yonde bir etki de olusturmadigi belirlenmistir. Yasama gucu, yemden yararlanma gibi ozellikler uzerinde olumsuz bir etki saptanmamis olup, altlik kalitesi ile ilgili olarak da nem ve pH acisindan az da olsa avantaj saglanabildigi soylenebilir. Anahtar Kelimeler: Broiler, altlik, zeolit, ayni altligin yeniden kullanimi EFFECTS OF ZEOLITE ADDITION TO BEDDING ON BROILER REARING Summary In this study, the effects of zeolite addition to broiler bedding and their re-using possibility were investigated. Also, the growth rate, survival ability, feed conversation, productability, indexes bedding humidity, pH, ammonia values and total bacterial content were all measured and considered in the study. It was observed that, zeolite addition to bedding made a beneficial effect on growth rate, and re-using of the same bedding did not cause any negative effect on the measured subjects. Survival rate and feed conversation were not affected negatively and it was concluded that zeolite has improved the bedding quality relating to the humidity and pH of the bedding.
