Relationship between the microbiota in different sections of the gastrointestinal tract, and the body weight of broiler chickens

In the poultry industry, many efforts have been undertaken to further improve the growth performance of broilers and identification and modulation of body weight (BW)-related bacteria could be one of the strategies to improve productivity. However, studies regarding the relationship between microbiota and BW are scarce. The objective of the present study was to investigate the relationship between microbiota and BW in different sections of the gastrointestinal tract (GIT). A total of twenty 18-day-old birds were selected based on the BW, and samples were collected from the three different sections of the GIT, which included the crop, ileum and cecum. Bacterial genomic DNA was extracted from the samples, and the V4 region of 16S rRNA gene were amplified. Amplicons were sequenced on Illumina MiSeq, and microbial communities were analyzed by using QIIME. In principal coordinate analysis, bacterial communities were clustered into three groups, based on the sections of GIT. Several BW-related bacterial groups were identified from linear regression analysis. At the genus level, Streptococcus from the ileum as well as Akkermansia in both ileum and cecum, were negatively related to BW, whereas Bifidobacterium in the ileum and Lactococcus in the cecum showed a positive correlation. The results from the present study showed that particular bacterial communities in the GIT were related to BW, and the study has broadened the understanding of the intestinal microbial ecosystem in broiler chickens.

Evaluation of mycotoxin sequestering agents for aflatoxin and deoxynivalenol: an in vitro approach.

An experiment was conducted to determine the efficacy of mycotoxin sequestering agents for binding or degrading aflatoxin B1 (AFB1) and deoxynivalenol (DON) by an in vitro method. Ten toxin binder products including 5 bentonite clays (bentonite A, B, C, D, and E), 2 cellulose products (cellulose A and B), a yeast cell wall, an activated charcoal, and a mixture product containing minerals, microorganisms, and phytogenic substances were used in this experiment. An in vitro procedure was used to mimic the digestive process in pigs. The binding ability for AFB1 of the cellulose products was less compared with the values of other sequestering products (p < 0.05). The percent adsorption of AFB1 by bentonite clays, cellulose products, yeast cell wall product, activated charcoal product, and the mixture product were 92.5 (average of 5 bentonite products), −13.5 (average of 2 cellulose products), 92.7, 100.2, and 96.6, respectively. The respective values for DON were 3.24, 11.6, 22.9, 14.4, and 4.3. In conclusion, most toxin sequestering agents used in the present study had potential to bind AFB1 rather than DON based on the in vitro study which simulated the pH condition of the gastrointestinal tract of pigs.

Effects of Feeding Barley Naturally Contaminated with Fusarium Mycotoxins on Growth Performance, Nutrient Digestibility, and Blood Chemistry of Gilts and Growth Recoveries by Feeding a Non-contaminated Diet

The objectives of this study were to investigate the effects of feeding barley naturally contaminated with Fusarium mycotoxins on growth performance, vulva swelling, and digestibility of dry matter, organic matter, and crude protein of gilts and the recovery of gilts fed normal diets immediately after the exposure to contaminated diets by measuring growth performance and vulva swelling. In Exp. 1, four diets were prepared to contain 0%, 15%, 30%, or 45% contaminated barley containing 25.7 mg/kg deoxynivalenol and 26.0 μg/kg zearalenone. Sixteen gilts with an initial body weight (BW) of 33.3 kg (standard deviation = 3.0) were individually housed in a metabolism crate and assigned to 4 diets with 4 replicates in a randomized complete block design based on BW. During the 14-d feeding trial, individual BW and feed consumption were measured weekly and the vertical and horizontal lengths of vulva were measured every 3 d. From d 10, feces were collected by the maker-to-marker method for 4 d. Blood samples were collected on d 14. During the overall period, the average daily gain, average daily feed intake, and gain:feed of pigs linearly decreased (p<0.01) as the dietary concentration of contaminated barley increased. However, the digestibility of crude protein was linearly increased (p = 0.011) with the increasing amounts of contaminated barley. Increasing dietary Fusarium mycotoxin concentrations did not influence vulva size, blood characteristic as well as immunoglobulin level of pigs. In the Exp. 2, a corn-soybean meal-based diet was formulated as a recovery diet. Pigs were fed the recovery diet immediately after completion of the Exp. 1. During the 14-d of recovery period, the individual BW and feed consumption were measured weekly and the vertical and horizontal length of vulva were measured every 3 d from d 0. On d 7, the feed intake of pigs previously fed contaminated diets already reached that of pigs fed a diet with 0% contaminated barley and no significant difference in growth performance among treatments was observed during d 7 to 14 of the recovery period. In conclusion, increasing levels of mycotoxins in diets linearly decreased the growth performance of pigs, and these damages can be recovered in 7 d after the diet was replaced with a normal diet. The vulva size, blood characteristic, immune responses were not affected by increasing level of contaminated barley in the diets fed to pigs.

Bioavailability of D-methionine relative to L-methionine for nursery pigs using the slope-ratio assay

This experiment was conducted to determine the bioavailability of D-methionine (Met) relative to L-Met for nursery pigs using the slope-ratio assay. A total of 50 crossbred barrows with an initial BW of 13.5 kg (SD = 1.0) were used in an N balance study. A Met-deficient basal diet (BD) was formulated to contain an adequate amount of all amino acids (AA) for 10–20 kg pigs except for Met. The two reference diets were prepared by supplementing the BD with 0.4 or 0.8 g L-Met/kg at the expense of corn starch, and an equivalent concentration of D-Met was added to the BD for the two test diets. The pigs were adapted to the experimental diets for 5 d and then total but separated collection of feces and urine was conducted for 4 d according to the marker-to-marker procedure. Nitrogen intakes were similar across the treatments. Fecal N output was not affected by Met supplementation regardless of source and consequently apparent N digestibility did not change. Conversely, there was a negative linear response (P < 0.01) to Met supplementation with both Met isomers in urinary N output, which resulted in increased retained N (g/4 d) and N retention (% of intake). No quadratic response was observed in any of the N balance criteria. The estimated bioavailability of D-Met relative to L-Met from urinary N output (g/4 d) and N retention (% of intake) as dependent variables using supplemental Met intake (g/4 d) as an independent variable were 87.6% and 89.6%, respectively; however, approximately 95% of the fiducial limits for the relative bioavailability estimates included 100%. In conclusion, with an absence of statistical significance, the present study indicated that the mean relative bioequivalence of D- to L-Met was 87.6% based on urinary N output or 89.6% based on N retention.

Additivity of Ileal Crude Protein Digestibility and Comparison of Digestibility with Methodological Consideration in Broilers

This study was aimed at evaluating the additivity of crude protein digestibility in mixed diets of corn and soybean meal (SBM), and comparing direct and indirect methods for evaluating crude protein (CP) digestibility. Totally, five hundred and twenty-five 18-day-old broiler chickens were grouped into 7 blocks based on body weight, and randomly allocated to 6 treatment groups in a randomized complete block design. The basal diet, diet 3, was corn-SBM-based, containing 65% corn and 28% SBM. Diets 1 and 5 contained corn and SBM, respectively, as the sole CP source. To use the difference method, 2 diets, diets 2 and 4, were prepared by mixing corn and SBM at the expense of the basal diet, respectively. Diet 2 contained 79% corn and 14% SBM, and diet 4 contained 32.5% corn and 34% SBM. To evaluate the additivity of digestibility values, the difference between measured values for the mixed diets (diets 2, 3, and 4) and predicted values calculated using the measured values for diets 1 and 5 was examined. The apparent (AID) and standardized (SID) ileal digestibility of CP in SBM differed between the direct and indirect methods; however, corresponding digestibility did not differ for corn. Additionally, the predicted and measured digestibility of both AID and SID differed in diets 2 and 3, implying that the digestibility values obtained from diets 1 and 5 were not additive for mixed diets. In conclusion, this study showed that digestibility evaluated by direct and indirect methods depends on the ingredients having different CP concentrations, and this finding may be considered to improve the accuracy of feed formulation for broiler chickens. Different Coefficients and Exponents for Metabolic Body Weight in a Model to Estimate Individual Feed Intake for Growing-finishing Pigs.

Estimation of feed intake (FI) for individual animals within a pen is needed in situations where more than one animal share a feeder during feeding trials. A partitioning method (PM) was previously published as a model to estimate the individual FI (IFI). Briefly, the IFI of a pig within the pen was calculated by partitioning IFI into IFI for maintenance (IFIm) and IFI for growth. In the PM, IFIm is determined based on the metabolic body weight (BW), which is calculated using the coefficient of 106 and exponent of 0.75. Two simulation studies were conducted to test the hypothesis that the use of different coefficients and exponents for metabolic BW to calculate IFIm improves the accuracy of the estimates of IFI for pigs, and that PM is applied to pigs fed in group-housing systems. The accuracy of prediction represented by difference between actual and estimated IFI was compared using PM, ratio (RM), or averaging method (AM). In simulation studies 1 and 2, the PM estimated IFI better than the AM and RM during most of the periods (p<0.05). The use of 0.60 as the exponent and the coefficient of 197 to calculate metabolic BW did not improve the accuracy of the IFI estimates in both simulation studies 1 and 2. The results imply that the use of 197 kcal×kg BW0.60 as metabolizable energy for maintenance in PM does not improve the accuracy of IFI estimations compared with the use of 106 kcal×kg BW0.75 and that the PM estimates the IFI of pigs with greater accuracy compared with the averaging or ratio methods in group-housing systems.

Strategy to Improve the Productivity of Broilers: Focusing on Pre-Starter Diet

Ninety-five percent of the farms are contracted with Integration Company. According to the Korean broiler performance index, broilers in Korea are marketed at 32 days with 1.52 kg of body weight. In contrast, the market age and body weight of broilers are 47 days/2.8 kg in the United States and 42 days/2.5 kg in Europe. Because of the younger market age of the Korean broiler, the pre-starter feed is important. Chicks exhibit poor absorption of dietary nutrients up to 7 days after hatching due to an immature digestive system and low enzyme secretion rate and activity. At the beginning of hatching, chicks obtain their nutrients from the egg yolk sac. It is highly recommended that chicks, after consuming the nutrients in the egg yolk sac, are given supplemented pre-starter feed to increase early growth rates and improve the performance of broiler production. Pre-starter nutrient requirements are not expressed in NRC, so Korean feeding standards for poultry and commercial breeding companies determine the nutrient requirements of pre-starter broiler chickens. Three approaches are followed to formulate specially designed pre-starter feeds for broiler chicks: (i) selective use of raw materials, (ii) proper standards of nutrient supply, and (iii) application of feed additives such as exogenous enzymes. In the selection of raw materials, those with high digestibility can be used. The absorption rate of carbohydrates in grains can be increased through feed processing at high temperature and high pressure. Soy proteins and fish meal can also be added as protein sources. As an energy source, vegetable oils are preferred over animal fats because of the former’s high digestibility. It is suggested that the levels of proteins and amino acids are higher in pre-starter feed than in starter feed. With regard to energy, the sources of energy are more important than the levels of energy in feed. Feed additives such as exogenous enzymes can be used to improve nutrient digestibility. In addition, organic acids and plant extracts can be used as alternatives to animal growth promoters to stimulate immunity and prevent diseases. The growth performance of broilers is affected by various factors, such as management and disease control, in addition to the nutritional strategy; however, nutritional strategies play an important role in improving the productivity of broilers. Therefore, nutritional strategies, along with management and disease control, are required for improving the productivity of broilers in Korea.
