3-Imidazolylmethylaminophenylsulfonyltetrahydroquinolines, a novel series of farnesyltransferase inhibitors

Charles Z. Ding; John T. Hunt; Carol Ricca; Veeraswamy Manne

Design, synthesis and structure-activity relationship of a series of 3-imidazolylmethylaminophenylsulfonyltetrahydroquinolines as farnesyltransferase inhibitors are presented. A working pharmacophore of inhibiting farnesyltransferase by this series of inhibitors is proposed.

A convenient diastereoselective synthesis of β-Aryl-β-aminoalcohols

Charles Z. Ding

Abstract β-Aryl-β-aminoalcohols (1) were prepared from N-diphenylmethylene benzylamines (2) diastereoselectively in a one-pot procedure.

A convenient Synthesis of 6-Substituted-2,2-Dimethyl-2H-1-Benzopyrans

Charles Z. Ding

Abstract Various 2,2-dimethyl-6-functional-2H-1-benzopyrans were prepared via a halogen-metal exchange reaction followed by treatment with electrophiles. These 6-functionalized chromenes are important synthetic intermediates for preparation of benzopyran-based potassium channel openers.

Synthesis of 4-(N-alkyl-N-heteroaryl)amino-3,4-dihydro-3-hydroxy-2,2-dimethyl-2H-1-benzopyran-6-carbonitrile derivatives via an unusual 1,4-oxygen to nitrogen heteroaryl migration

Charles Z. Ding; Arthur V. Miller

Abstract Various heteroaryls were found to undergo 1,4-oxygen to nitrogen migration (e.g. 4→1 ) under acidic conditions. This unprecedented migratin provides a convenient access to 4-(N-alkyl-N-heteroaryl)amino-3,4-dihydro-3-hydroxy-2,2-dimethyl-2H-1-benzopyran-6-carbonitrile derivatives, which are otherwise difficult to prepare.

The design, synthesis and structure-activity relationships of a novel series of 3,4-disubstituted pyrrolidine acid analogs as PPAR ligands is outlined. In both the 1,3- and 1,4-oxybenzyl pyrrolidine acid series, the preferred stereochemistry was shown to be the cis-3R,4S isomer, as exemplified by the potent dual PPARα/γ agonists 3k and 4i. The N-4-trifluoromethyl-pyrimidinyl pyrrolidine acid analog 4i was efficacious in lowering fasting glucose and triglyceride levels in diabetic db/db mice.
