Effect of osmotic stress and sodium nitroprusside pretreatment on proline metabolism of wheat seedlings

Effect of osmotic stress and sodium nitroprusside (SNP, NO donor) pretreatment on growth and proline metabolism of wheat seedlings was investigated. Polyethylene glycol 6000 treatment for 2, 4 and 6 d could be termed as mild, moderate and severe stress, respectively, according to decrease in the relative water content. Severe osmotic stress significantly decreased the growth and photochemical efficiency, and increased proline content due to activation of its synthesis. 0.2 mM SNP pretreatment enhanced growth of wheat seedlings, increased variable to maximum fluorescence ratio (Fv/Fm) and fluorescence yield, while decreased proline content. However, 2 mM SNP retarded the seedlings growth and chlorophyll a fluorescence, and increased proline accumulation. Our results showed that NO might be involved in the regulation of osmotic stress in a concentration-dependent manner.

The identification of flavonoids and the expression of genes of anthocyanin biosynthesis in the chrysanthemum flowers

In order to provide additional information on the coloration of chrysanthemum flowers, the flavonoid composition and the expression of six structural genes involved in anthocyanin pathway in the ray florets of a pink flowering (cv. H5) and two white flowering (cvs. Keikai and Jinba) Chrysanthemum grandiflorum cultivars were examined. HPLCDAD/ESI-MSn analysis showed that cyanidin 3-O-(6″-O-malonylglucoside) and cyanidin 3-O-(3″,6″-O-dimalonylglucoside) were the two major flavonoids presented in H5, while white flowering cultivars contained flavones instead of anthocyanins. Nine flavone derivatives were detected in the three cultivars, the amount of each flavone varied upon cultivars, and seven of these were identified as luteolin 7-O-arabinosylglucuronide, apigenin 7-O-glucoside, luteolin 7-O-malonylglucoside, apigenin 7-O-malonylglucoside, chrysoeriol 7-O-malonylglucoside, acacetin 7-O-rutinoside and acacetin 7-O-malonylglucoside. The two white flowering cultivars showed similar total flavonoid content, which was about two fold higher than that in H5. A high expression of the genes encoding dihydroflavonol 4-reductase and 3-O-glucosyltransferase was detected only in H5 but not in Keikai or Jinba. Chalcone synthase, chalcone isomerase, flavanone 3-hydroxylase, and flavonoid 3′-hydroxylase were expressed in all flowers, suggesting that the lack of anthocyanin in white flowering cultivars cannot be due to any blockage of their expression.

Uni-Directional Transportation on Peristome-Mimetic Surfaces for Completely Wetting Liquids

Liquid uni-directional transport on solid surface without energy input would advance a variety of applications, such as in bio-fluidic devices, self-lubrication, and high-resolution printing. Inspired by the liquid uni-directional transportation on the peristome surface of Nepenthes alata, here, we fabricated a peristome-mimicking surface through high-resolution stereo-lithography and demonstrated the detailed uni-directional transportation mechanism from a micro-scaled view visualized through X-ray microscopy. Significantly, an overflow-controlled liquid uni-directional transportation mechanism is proposed and demonstrated. Unlike the canonical predictions for completely wetting liquids spreading symmetrically on a high-energy surface, liquids with varied surface tensions and viscosities can spontaneously propagate in a single preferred direction and pin in all others. The fundamental understanding gained from this robust system enabled us to tailor advanced micro-computerized tomography scanning and stereo-lithography fabrication to mimic natural creatures and construct a wide variety of fluidic machines out of traditional materials.

Responses to drought stress in two poplar species originating from different altitudes.

Cuttings of Populus kangdingensis and Populus cathayana, originating from high and low altitudes in the eastern Himalaya, respectively, were examined during one growing season in a greenhouse to determine their responses to drought stress (soil moisture decreased from 100 to 55 or 25 % field capacity). Compared to control plants grown under 100 % field capacity, those poplars grown under 55 and 25 % field capacity possessed lower increases in height and stem diameter, and higher contents of soluble sugars, free proline, malondialdehyde (MDA) and hydrogen peroxide, and higher activities of catalase (CAT), superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX) and glutathione reductase (GR). Compared with P. cathayana with greater leaf area, P. kangdingensis with greater root/shoot ratio exhibited lower MDA and H2O2 contents, higher soluble sugar and free proline contents, and higher activities of CAT, SOD, POD, APX and GR. These results suggested that P. kangdingensis was more drought tolerant than P. cathayana.

Growth, abscisic acid content, and carbon isotope composition in wheat cultivars grown under different soil moisture

Changes in dry matter accumulation and allocation, abscisic acid content and carbon isotope composition of three wheat cultivars from dry, middle and wet climate regions were recorded at full maturity after exposure to different watering regimes (100, 50 and 25 % field capacity). Compared with the wet climate cultivar, the dry climate cultivar showed lower stem height, total leaf area, total dry biomass and total grain dry mass, and higher root/shoot ratio, abscisic acid content and carbon isotope composition under all watering regimes. Both water-limited treatments significantly reduced leaf growth and increased dry matter allocation into the roots leading to a significant raise of root/shoot ratio in all cultivars tested. In addition, drought affected morphological and physiological properties more in the dry climate cultivar than in the wet climate cultivar.

Different responses of two contrasting wheat genotypes to abscisic acid application

Purpose of this study was to investigate different responses of two wheat genotypes (Triticum aestivum L.) from the wet and dry climate regions to exogenous abscisic acid (ABA) application under well-watered and water-stressed conditions. Exogenous ABA was applied to the leaves by spraying and changes in dry matter accumulation and allocation, endogenous ABA content and carbon isotope ratio (δ13C) were monitored. The ABA application significantly decreased stem height, total biomass, total leaf area, total grain mass and leaf area/mass ratio, and significantly increased root/aboveground biomass ratio, endogenous ABA content and δ13C under well-watered and water-stressed conditions. Compared with the wet climate genotype, the dry climate genotype was more responsive to exogenous ABA application, resulting in lower stem height, total biomass, total leaf area, total grain mass and leaf area/mass ratio, and higher root/aboveground biomass ratio, endogenous ABA content and δ13C under all experimental treatments.

Time-Dependent Liquid Transport on a Biomimetic Topological Surface

Liquid drops impacting on a solid surface is a familiar phenomenon. On rainy days, it is quite important for leaves to drain off impacting raindrops. Water can bounce off or flow down a water-repellent leaf easily, but with difficulty on a hydrophilic leaf. Here, we show an interesting phenomenon in which impacting drops on the hydrophilic pitcher rim of Nepenthes alata can spread outward to prohibit water filling the pitcher tank. We mimic the peristome surface through a designed 3D printing and replicating way and report a time-dependently switchable liquid transport based on biomimetic topological structures, where surface curvature can work synergistically with the surface microtextures to manipulate the switchable spreading performance. Motived by this strange behavior, we construct a large-scaled peristome-mimetic surface in a 3D profile, demonstrating the ability to reduce the need to mop or to squeegee drops that form during the drop impacting process on pipes or other curved surfaces in food processing, moisture transfer, heat management, etc.

Liquids Unidirectional Transport on Dual-Scale Arrays

Liquids unidirectional transport has cutting-edge applications ranging from fog collection, oil-water separation, to microfluidic devices. Despite extensive progresses, existing man-made surfaces with asymmetric wettability or micro/nanoscales structures are still limited by complex fabrication techniques or obscure essential transport mechanisms to achieve unidirectional transport with both high speeds and large volumes. Here, we demonstrate the three-dimensional printed micro/macro dual-scale arrays for rapid, spontaneous, and continuous unidirectional transport. We reveal the essential directional transport mechanism via a Laplace pressure driven theory. The relationship between liquid unidirectional transport and surface morphology parameter is systematically explored. Threshold values to achieve unidirectional transport are determined. Significantly, dual-scale arrays even facilitate liquids uphill running, microfluidics patterning, and liquid shunting in target directions without external energy input. Free combination of dual-scale island arrays modules, just like LEGO bricks, achieves fast liquid transport on demand. This dual-scale island array can be used to build smart laboratory-on-a-chip devices, printable microfluidic integration systems, and advanced biochemistry microreactors.
