Factors Affecting Intestinal Health in Poultry

Abstract The gastrointestinal (GI) tract has the most extensive exposed surface in the body and is constantly exposed to a wide variety of potentially harmful substances. The GI tract acts as a selective barrier between the tissues of the bird and its luminal environment. This barrier is composed of physical, chemical, immunological, and microbiological components. A wide range of factors associated with diet and infectious disease agents can negatively affect the delicate balance among the components of the chicken gut and, as a result, affect health status and production performance of birds in commercial poultry operations. Phasing out of antibiotic growth promoters from poultry diets in Europe and recent moves toward reduction or removal of these compounds in other parts of the world including North America will likely change the microbial profile of the GI tract environment in commercial poultry. This paper reviews the GI tract from developmental, immunological, and microbial standpoints and then discusses factors that can affect health status of this system. Necrotic enteritis and coccidiosis and their interactions, and possible consequences of antibiotic growth promoter removal from poultry diets with respect to these diseases, are discussed in more detail.

Detection and Identification of Lactobacillus Species in Crops of Broilers of Different Ages by Using PCR-Denaturing Gradient Gel Electrophoresis and Amplified Ribosomal DNA Restriction Analysis

ABSTRACT The microflora of the crop was investigated throughout the broiler production period (0 to 42 days) using PCR combined with denaturing gradient gel electrophoresis (PCR-DGGE) and selective bacteriological culture of lactobacilli followed by amplified ribosomal DNA restriction analysis (ARDRA). The birds were raised under conditions similar to those used in commercial broiler production. Lactobacilli predominated and attained populations of 108 to 109 CFU per gram of crop contents. Many of the lactobacilli present in the crop (61.9% of isolates) belonged to species of the Lactobacillus acidophilus group and could not be differentiated by PCR-DGGE. A rapid and simple ARDRA method was developed to distinguish between the members of the L. acidophilus group. HaeIII-ARDRA was used for preliminary identification of isolates in the L. acidophilus group and to identify Lactobacillus reuteri and Lactobacillus salivarius. MseI-ARDRA generated unique patterns for all species of the L. acidophilus group, identifying Lactobacillus crispatus, Lactobacillus johnsonii, and Lactobacillus gallinarum among crop isolates. The results of our study provide comprehensive knowledge of the Lactobacillus microflora in the crops of birds of different ages using nucleic acid-based methods of detection and identification based on current taxonomic criteria.

Growth, efficiency, and yield of commercial broilers from 1957, 1978, and 2005

The effect of commercial selection on the growth, efficiency, and yield of broilers was studied using 2 University of Alberta Meat Control strains unselected since 1957 and 1978, and a commercial Ross 308 strain (2005). Mixed-sex chicks (n = 180 per strain) were placed into 4 replicate pens per strain, and grown on a current nutritional program to 56 d of age. Weekly front and side profile photographs of 8 birds per strain were collected. Growth rate, feed intake, and measures of feed efficiency including feed conversion ratio, residual feed intake, and residual maintenance energy requirements were characterized. A nonlinear mixed Gompertz growth model was used to predict BW and BW variation, useful for subsequent stochastic growth simulation. Dissections were conducted on 8 birds per strain semiweekly from 21 to 56 d of age to characterize allometric growth of pectoralis muscles, leg meat, abdominal fat pad, liver, gut, and heart. A novel nonlinear analysis of covariance was used to test the hypothesis that allometric growth patterns have changed as a result of commercial selection pressure. From 1957 to 2005, broiler growth increased by over 400%, with a concurrent 50% reduction in feed conversion ratio, corresponding to a compound annual rate of increase in 42 d live BW of 3.30%. Forty-two-day FCR decreased by 2.55% each year over the same 48-yr period. Pectoralis major growth potential increased, whereas abdominal fat decreased due to genetic selection pressure over the same time period. From 1957 to 2005, pectoralis minor yield at 42 d of age was 30% higher in males and 37% higher in females; pectoralis major yield increased by 79% in males and 85% in females. Over almost 50 yr of commercial quantitative genetic selection pressure, intended beneficial changes have been achieved. Unintended changes such as enhanced sexual dimorphism are likely inconsequential, though musculoskeletal, immune function, and parent stock management challenges may require additional attention in future selection programs.

Bone Mineral Density and Breaking Strength of White Leghorns Housed in Conventional, Modified, and Commercially Available Colony Battery Cages

Limited opportunity for movement and load-bearing exercise for conventionally caged laying hens leads to bone loss and increased susceptibility to osteoporosis, bone fractures, and cage layer fatigue, all of which compromise hen welfare and have negative consequences for production. The objective of this study was to compare bone mineral density (BMD) and strength measures of White Leghorns housed in conventional battery cages (CONV), cages modified to incorporate a nest box and perch (MOD), and commercially available, furnished colony cages with (CWDB) or without (CWODB) a raised dust bath. Hens reared on floor litter were randomly allocated to 1 of 4 cage systems at 19 wk of age. Hen-day production and egg quality were measured between 20 and 64 wk. At 65 wk, hens were killed, and right femur, tibia, and humerus were excised. Bone mineral density was assessed using quantitative computed tomography, and breaking strength was measured with an Instron Materials Tester. In the femur and tibia, CONV hens exhibited lower total BMD, bone mass, cortical bone area, cortical bone mass, and bone-breaking strength than CWDB, CWODB, and MOD hens. Density and cross-sectional area of bone in the trabecular space was highest in CONV. In the humerus, total and cortical BMD and mass and breaking strength values were higher for colony-housed birds than hens in CONV and MOD. The MOD birds did not exhibit increased humeral BMD or strength measures over CONV hens. These findings provide evidence that hens housed in modified and colony cages, furnished systems that promote load-bearing movement, are better able to preserve cortical structural bone than conventionally caged hens and simultaneously have stronger bones. Furthermore, inclusion of raised amenities that encourage wing loading is necessary to reduce humeral cortical bone loss. The overall absence of correlation between egg production or quality and bone quality measures also suggests that improved bone quality in CWDB, CWODB, and MOD furnished cages is not the result of lowered egg production or quality.

The effect of calcium source and particle size on the production performance and bone quality of laying hens

The efficacy of 3 local limestone sources as potential Ca sources for laying hens was studied. Limestone sources were assessed for in vitro solubility. Four Ca sources (control, A, B, or C) and 2 particle size combinations (ground, 100% ground; or mixed, 67% ground+33% large particle) was used. The control consisted of a commercial ground limestone for the ground Ca source and oyster shell of the large particle Ca source. DeKalb laying hens were randomly placed in 32 battery cage units (n=12/cage). At 19 wk of age, hens received 1 of 8 Ca source and particle size treatments (4 replicates/treatment) until 74 wk. Egg production, feed consumption, BW, and egg quality were measured throughout. Bone mineral density (by quantitative computed tomography), breaking strength, ash, and Ca were assessed at the end of lay. In vitro solubility was dependent upon Ca source and particle size (P<0.0001) with oyster shell generally having a greater in vitro solubility than test limestone sources A, B, and C of similar particle sizes. Feed consumption (100 to 117 g/bird per day), BW (1,500 to 1,800 g), egg production (92% peak egg production), egg weight (55 to 67 g), and egg specific gravity (1.090 to 1.078) did not differ among hens fed the different Ca sources (P>0.05). Tibia bone mineral density indicated the oyster shell treatment had a lower trabecular density than Ca source A; however, all other Ca sources had similar bone mineral density for all measures (P>0.05). Hens fed the mixed Ca particle treatments consistently had greater feed consumption from 27 to 70 wk than those fed the 100% ground Ca source (P<0.05). Bone mineralization was enhanced in hens that received the mixed Ca particle treatments (P<0.05). Overall, the results of this study indicate that the local limestone sources A, B, and C would be suitable alternatives to current commercial sources of Ca for laying hens. In addition, large particle Ca did improve bone quality.

Omega-3-enriched broiler meat: 1. Optimization of a production system

Consumer awareness of the health benefits of n-3 fatty acids is growing and is driving consumer demand for enriched food products. Enrichment of meat with n-3 fatty acids is an opportunity for the broiler production sector to add value to their product, but enrichment can increase the cost of production. A study was conducted to determine an optimal production strategy for n-3 enrichment of broiler meat using ground full-fat flaxseed. Low and high levels of dietary flaxseed (10 and 17%, respectively) were fed to broilers for 8 lengths of time (0, 4, 8, 12, 16, 20, 24, or 35 d) before processing at 35 d. Increasing the level or duration decreased feed intake, BW, and the percentage yield of carcass and breast. Flaxseed level and duration of feeding increased feed conversion ratios and the cost of production. Feeding flaxseed at 10 and 17% increased breast n-3 fatty acid levels by 7.65 and 13.70 mg/100 g of meat per day, respectively. In breast meat, the threshold level of 300 mg/g, required in Canada for labeling foods as a source of n-3 fatty acids, was reached at 12.1 and 24.1 d in the high and low flaxseed treatments, respectively. This was due primarily to a tripling of alpha-linolenic acid (18:3n-3) levels in the breast meat. Levels of the long-chain n-3 fatty acid eicosapentaenoic acid increased significantly in the breast meat with increased level and duration of flax feeding, indicating that birds were able to desaturate and elongate alpha-linolenic acid to eicosapentaenoic acid. To minimize cost, while achieving adequate breast meat n-3 enrichment, carcass weight, and meat yield, feeding 10% flaxseed for 24.1 d before processing is recommended as an optimal breast meat n-3 enrichment strategy. The optimal thigh meat n-3 enrichment strategy was to feed 10% flaxseed for 4.54 d before processing.

Detection, Characterization, and In Vitro and In Vivo Expression of Genes Encoding S-Proteins in Lactobacillus gallinarum Strains Isolated from Chicken Crops

ABSTRACT Thirty-eight isolates of Lactobacillus gallinarum cultured from the crops of broiler chickens were screened for the presence of genes encoding S-layer proteins. All of the isolates had two S-protein genes, which were designated Lactobacillus gallinarum S-protein (lgs) genes. One gene in each isolate was either lgsA or lgsB. The Lactobacillus isolates were further characterized by pulsed-field gel electrophoresis of DNA digests, which grouped the isolates into 17 genotypes (strains). The second gene in each of eight representative strains was sequenced and shown to differ among strains (lgsC, lgsD, lgsE, lgsF, lgsG, lgsH, and lgsI). The genome of each strain thus encoded a common S-protein (encoded by either lgsA or lgsB) and a strain-specific S-protein. The extraction of cell surface proteins from cultures of the eight strains showed that each strain produced a single S-protein that was always encoded by the strain-specific lgs gene. Two of the strains were used to inoculate chickens maintained in a protected environment which were Lactobacillus-free prior to inoculation. DNAs and RNAs extracted from the digesta of the chickens were used for PCR and reverse transcription-PCR, respectively, to demonstrate the presence and transcription of lgs genes in vivo. In both cases, only the strain-specific gene was transcribed. Both of the strains adhered to the crop epithelium, consistent with published data predicting that S-proteins of lactobacilli are adhesins. The results of this study provide a basis for the investigation of gene duplication and sequence variation as mechanisms by which bacterial strains of the same species can share the same habitat.

Comparative feeding value of extruded and nonextruded wheat and corn distillers dried grains with solubles for broilers

The feeding value of extruded and nonextruded wheat and corn distillers dried grains with solubles (DDGS) for broilers was evaluated in 2 experiments. In experiment 1, male broilers (n=360) housed in battery cages were fed assay diets that included either 15 or 30% wheat or corn DDGS (extruded or not) in relation to a basal diet from d 21 to 28. Birds were killed on d 28 and ileal digesta was collected to establish the apparent ileal digestibility (AID) coefficients of energy and nutrients for test ingredients using the difference method based on 5 cages of 8 birds per diet. In experiment 2, a 42-d study compared the growth performance of broilers fed phase diets including 0, 5, or 10% wheat or corn DDGS, based on 4 pens of 55 birds per diet×sex combination. Diets within phase were formulated to have a similar content of AME, CP, and digestible lysine. Breast meat weight and yield were determined on d 37 by sampling 5 birds per pen. In experiment 1, at 15% inclusion, AID coefficients of most amino acids were higher for corn DDGS than for wheat DDGS (P<0.05). At 30% inclusion, however, there were fewer differences in AID between corn and wheat DDGS. Twin-screw extrusion increased the AID of AA in both corn and wheat DDGS by 10 to 34% (P<0.05). In experiment 2, there was no adverse effect of including corn or wheat DDGS at up to 10% of the diet on pen average daily weight gain, feed disappearance, feed efficiency, breast meat weight, or yield. In conclusion, extrusion increased the feeding value of DDGS. The AID coefficients for amino acids were similar between corn and wheat DDGS. We also confirmed that either corn or wheat DDGS can be included at up to 10% of wheat-based broiler diets without affecting growth performance or breast meat yield.

Comparison of bones of 4 strains of laying hens kept in conventional cages and floor pens

The maintenance of bone strength has been an important issue in the debate over cage use for laying hens. Bone strength depends on adequate mechanical load and cages restrict movement. Four laying crosses (Lohmann White, Lohmann Brown, H&N White, and Rhode Island Red × Barred Plymouth Rock cross hens) were housed in conventional cages or in floor pens equipped with perches and nest boxes to measure the effect of the housing system on bone strength. Approximately 15 hens of each genotype from each housing system were killed at 50 wk of age and the radius and tibia of each were removed for analysis. There were no differences between the Lohmann White and H&N White (White Leghorn) hens, likely because of their similar genetic background. The Lohmann Brown and the cross hens (brown-egg layers) were larger and they had heavier bones, but the bone density was not different from that of the other lines. The radius was heavier for hens kept in floor pens than for those kept in cages, but the tibia was not. When hens were kept in floor pens, both bones had greater cortical bone density and cross-sectional area, but the difference between housing systems in cortical bone cross-sectional area was much greater for the radius than it was for the tibia. Although the movement of hens in cages is limited, they spend a great deal of time standing, which puts a mechanical load on the tibia. Hens in floor pens are able to stretch their wings or fly, in contrast to hens kept in cages, which likely explains why the difference between housing systems in cortical bone was greater for the radius than for the tibia.

Nutritive value of single-screw extruded and nonextruded triticale distillers dried grains with solubles, with and without an enzyme complex, for broilers

The nutritive value of triticale distillers dried grains with solubles (DDGS) for broilers was investigated in 2 experiments. In experiment 1, four hundred male broilers housed in battery cages were fed diets including 15 or 30% triticale DDGS (extruded or not) or a basal diet, supplemented with or without a multi-enzyme complex from d 21 to 28. Birds were killed and ileal digesta was collected on d 28 to establish the apparent ileal nutrient digestibility (AID) coefficients for both assay diets and DDGS as test ingredients based on 5 cages per diet. In experiment 2, a 42-d performance study compared growth phase-specific diets formulated to similar levels of AME, CP, and digestible lysine with graded levels (0, 5, or 10%) of triticale DDGS inclusion based on a minimum of 4 pens per diet x sex combination. Breast muscle weight and percentage yield were determined on d 37 by sampling 5 birds per pen. In experiment 1, there was a significant (P < 0.05) DDGS level of inclusion x enzyme interaction for CP, lysine, methionine, tryptophan, isoleucine, histidine, and phenylalanine, such that the AID increased with enzyme supplementation based on 15% but not 30% DDGS inclusion. At 15% DDGS inclusion, enzyme supplementation increased the AID of these nutrients in DDGS between 6 and 19 percentage units. Extrusion of triticale DDGS increased (P < 0.05) the AID of GE, CP, methionine, tryptophan, branched-chain amino acids, and phenylalanine between 3 and 8 percentage units. In experiment 2, feeding up to 10% triticale DDGS had no adverse effect on feed intake, weight gain, or feed efficiency of broilers compared with controls over the 42-d study. Feeding up to 10% triticale DDGS did not affect breast weight or yield on d 37. In conclusion, feed enzyme complex supplementation and extrusion both increased the nutritive value of triticale DDGS for broilers. Triticale DDGS can be fed at up to 10% of practical broiler diets without adverse effect on performance and breast muscle yield.
