A water-insoluble glucan, PFPSIN, has been isolated from the aqueous extract of an edible mushroom Pleurotus florida. On the basis of total acid hydrolysis, methylation analysis, periodate oxidation, Smith degradation, and (13)C NMR experiments, the repeating unit of the polysaccharide was established as Conformational analysis revealed the triple helical conformation of this glucan.

The water-soluble glucan was obtained from Pleurotus florida fruit bodies by hot water extraction, ethanol precipitation, DEAE cellulose dialysis and Sephadex G-75 gel filtration. The structural information of the glucan was achieved by chemical (hydrolysis, methylation, periodate oxidation) and spectroscopic (1H and 13C) analyses, indicated a repeating unit built up of (1→6)-linked D-glucose. An immunoenhancing polysaccharide isolated from the hot aqueous extract of mature pods of Moringa oleifera was found to contain only D-glucose as a monosaccharide constituent. On the basis of total acid hydrolysis, methylation analysis, periodate oxidation and NMR studies (1H and 13C) the structure of the repeating unit of the polysaccharide was assigned and indicated only α-(1→4) linked Glucan. This polysaccharide showed significant macrophage activity through the release of nitric oxide on MOUSE monocyte cell line (J744.1) and also enhanced the number of albumin/ globulin percentage, the phagocyte cells and respiratory burst cells in fish system.
