Functional models for iron-bleomycin

Abstract A new pentadentate ligand, N,N -bis(2-pyridylmethyl)- N -bis(2-pyridyl)methylamine (N4Py), has been prepared. The corresponding iron(II) complex, [N4PyFe(CH 3 CN)](ClO 4 ) 2 , serves as a functional model for the active site of iron-bleomycin. Upon treatment with H 2 O 2 the formation of a purple intermediate has been observed, which has been characterized as an iron(III)-hydroperoxide complex. This N4Py-iron-hydroperoxide system has proven to be an active oxidation catalyst.

Preparation and properties of iron carbonyl complexes of 2,5-dihydrothiophene-1 oxide

J.H. Eekhof; H. Hogeveen; Richard M. Kellogg; Ebe P. Schudde

Abstract On reaction with diironnonacarbonyl in refluxing ether, 2,5-dihydrothiophene-1-oxide forms an iron tricarbonyl complex in which the ligand has not undergone rearrangement. The iron tricarbonyl complex is formed from an intermediate that appears to be a tetracarbonyl complex in which the iron is bonded to the sulfoxide, but not to the double bond of 2,5-dihydrothiophene-1-oxide. Some reactions of the tricarbonyl complex have been examined.

A functional mimic of natural peroxidases: synthesis and catalytic activity of a non-heme iron/peptide hydroperoxide complex

Christin T. Choma; Ebe P. Schudde; Richard M. Kellogg; George T. Robillard; Ben L. Feringa

Site-selective attachment of unprotected peptides to a non-heme iron complex is achieved by displacing two halides on the catalyst by peptide caesium thiolates. This coupling approach should be compatible with any peptide sequence provided there is only a single reduced cysteine. The oxidation activity with hydrogen peroxide of the dipeptide–catalyst complex in water is retained, and shows similarities with oxidation mechanisms observed for natural oxidizing enzymes. The results pave the way for the future design of peroxidase mimics where the activity of the catalyst will be modulated by a designed protein matrix.
