Lake eutrophication and brownification downgrade availability and transfer of essential fatty acids for human consumption.

Fish are an important source of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) for birds, mammals and humans. In aquatic food webs, these highly unsaturated fatty acids (HUFA) are essential for many physiological processes and mainly synthetized by distinct phytoplankton taxa. Consumers at different trophic levels obtain essential fatty acids from their diet because they cannot produce these sufficiently de novo. Here, we evaluated how the increase in phosphorus concentration (eutrophication) or terrestrial organic matter inputs (brownification) change EPA and DHA content in the phytoplankton. Then, we evaluated whether these changes can be seen in the EPA and DHA content of piscivorous European perch (Perca fluviatilis), which is a widely distributed species and commonly consumed by humans. Data from 713 lakes showed statistically significant differences in the abundance of EPA- and DHA-synthesizing phytoplankton as well as in the concentrations and content of these essential fatty acids among oligo-mesotrophic, eutrophic and dystrophic lakes. The EPA and DHA content of phytoplankton biomass (mgHUFAg-1) was significantly lower in the eutrophic lakes than in the oligo-mesotrophic or dystrophic lakes. We found a strong significant correlation between the DHA content in the muscle of piscivorous perch and phytoplankton DHA content (r=0.85) as well with the contribution of DHA-synthesizing phytoplankton taxa (r=0.83). Among all DHA-synthesizing phytoplankton this correlation was the strongest with the dinoflagellates (r=0.74) and chrysophytes (r=0.70). Accordingly, the EPA+DHA content of perch muscle decreased with increasing total phosphorus (r2=0.80) and dissolved organic carbon concentration (r2=0.83) in the lakes. Our results suggest that although eutrophication generally increase biomass production across different trophic levels, the high proportion of low-quality primary producers reduce EPA and DHA content in the food web up to predatory fish. Ultimately, it seems that lake eutrophication and brownification decrease the nutritional quality of fish for human consumers.

Inferring Phytoplankton, Terrestrial Plant and Bacteria Bulk δ¹³C Values from Compound Specific Analyses of Lipids and Fatty Acids.

Stable isotope mixing models in aquatic ecology require δ13C values for food web end members such as phytoplankton and bacteria, however it is rarely possible to measure these directly. Hence there is a critical need for improved methods for estimating the δ13C ratios of phytoplankton, bacteria and terrestrial detritus from within mixed seston. We determined the δ13C values of lipids, phospholipids and biomarker fatty acids and used these to calculate isotopic differences compared to the whole-cell δ13C values for eight phytoplankton classes, five bacterial taxa, and three types of terrestrial organic matter (two trees and one grass). The lipid content was higher amongst the phytoplankton (9.5±4.0%) than bacteria (7.3±0.8%) or terrestrial matter (3.9±1.7%). Our measurements revealed that the δ13C values of lipids followed phylogenetic classification among phytoplankton (78.2% of variance was explained by class), bacteria and terrestrial matter, and there was a strong correlation between the δ13C values of total lipids, phospholipids and individual fatty acids. Amongst the phytoplankton, the isotopic difference between biomarker fatty acids and bulk biomass averaged -10.7±1.1‰ for Chlorophyceae and Cyanophyceae, and -6.1±1.7‰ for Cryptophyceae, Chrysophyceae and Diatomophyceae. For heterotrophic bacteria and for type I and type II methane-oxidizing bacteria our results showed a -1.3±1.3‰, -8.0±4.4‰, and -3.4±1.4‰ δ13C difference, respectively, between biomarker fatty acids and bulk biomass. For terrestrial matter the isotopic difference averaged -6.6±1.2‰. Based on these results, the δ13C values of total lipids and biomarker fatty acids can be used to determine the δ13C values of bulk phytoplankton, bacteria or terrestrial matter with ± 1.4‰ uncertainty (i.e., the pooled SD of the isotopic difference for all samples). We conclude that when compound-specific stable isotope analyses become more widely available, the determination of δ13C values for selected biomarker fatty acids coupled with established isotopic differences, offers a promising way to determine taxa-specific bulk δ13C values for the phytoplankton, bacteria, and terrestrial detritus embedded within mixed seston.

Suitability of Phytosterols Alongside Fatty Acids as Chemotaxonomic Biomarkers for Phytoplankton.

The composition and abundance of phytoplankton is an important factor defining ecological status of marine and freshwater ecosystems. Chemotaxonomic markers (e.g., pigments and fatty acids) are needed for monitoring changes in a phytoplankton community and to know the nutritional quality of seston for herbivorous zooplankton. Here we investigated the suitability of sterols along with fatty acids as chemotaxonomic markers using multivariate statistics, by analyzing the sterol and fatty acid composition of 10 different phytoplankton classes including altogether 37 strains isolated from freshwater lakes. We were able to detect a total of 47 fatty acids and 29 sterols in our phytoplankton samples, which both differed statistically significantly between phytoplankton classes. Due to the high variation of fatty acid composition among Cyanophyceae, taxonomical differentiation increased when Cyanophyceae were excluded from statistical analysis. Sterol composition was more heterogeneous within class than fatty acids and did not improve separation of phytoplankton classes when used alongside fatty acids. However, we conclude that sterols can provide additional information on the abundance of specific genera within a class which can be generated by using fatty acids. For example, whereas high C16 ω-3 PUFA (polyunsaturated fatty acid) indicates the presence of Chlorophyceae, a simultaneous high amount of ergosterol could specify the presence of Chlamydomonas spp. (Chlorophyceae). Additionally, we found specific 4α-methyl sterols for distinct Dinophyceae genera, suggesting that 4α-methyl sterols can potentially separate freshwater dinoflagellates from each other.

An urban boreal lake basin as a source of CO2 and CH4

Up to now, carbon gas fluxes from urban lakes in the boreal zone have seldom been studied. In summer 2005 we investigated fluxes from an urban boreal lake basin in southern Finland with long history of eutrophication and anoxia. Hypolimnetic CO₂ and CH₄ concentrations were high compared to other boreal lakes. During the open-water period, the lake basin acted as a source of CO₂ and CH₄ with fluxes of 2.10 mol m(-2 )and 0.04 mol m(-2), respectively. Despite the high oxidation rate (83%), CH₄ flux was higher than in other lakes and CH₄ contributed 33% [ corrected] to Global Warming Potential. The ratio of carbon emission to accumulation was 4, i.e. emissions were an important route for carbon departure but less so than in rural lakes. Since the lake oxygen conditions affected nutrient availability, there was a positive feedback from hypolimnion to carbon uptake, which was reflected in gas concentrations.

The Importance of Phytoplankton Biomolecule Availability for Secondary Production

The growth and reproduction of animals is affected by their access to resources. In aquatic ecosystems, the availability of essential biomolecules for filter-feeding zooplankton dependents greatly on phytoplankton. Here, we analyzed the biochemical composition, i.e., the fatty acid, sterol and amino acid profiles and concentrations as well as protein, carbon, nitrogen and phosphorus content of 17 phytoplankton monocultures representing the seven most abundant phytoplankton classes in boreal and sub-arctic lakes. To examine how the differences in the biochemical composition between phytoplankton classes affect their nutritional quality for consumers, we assessed the performance of Daphnia, on these diets. Furthermore, we defined the most important biomolecules regulating the somatic growth and reproduction of Daphnia, expecting that higher concentrations of certain biomolecules are needed for reproduction than for growth. Finally, we combined these results with phytoplankton field data from over 900 boreal and sub-arctic lakes in order to estimate whether the somatic growth of Daphnia is sterol-limited when the natural phytoplankton communities are cyanobacteria-dominated. Our analysis shows that Daphnia grows best with phytoplankton rich in sterols, ω-3 fatty acids, protein and amino acids. Their reproduction follows food sterol and ω-3 concentration as well as C:P-ratio being two times higher in Daphnia feeding on cryptophytes than any other diet. Interestingly, we found that a high dietary ω-6 fatty acid concentration decreases both somatic growth and reproduction of Daphnia. When combined with phytoplankton community composition field data, our results indicate that zooplankton is constantly limited by sterols in lakes dominated by cyanobacteria (≥ 40 % of total phytoplankton biomass), and that the absence of cryptophytes can severely hinder zooplankton production in nature.

Marine Cryptophytes Are Great Sources of EPA and DHA

Microalgae have the ability to synthetize many compounds, some of which have been recognized as a source of functional ingredients for nutraceuticals with positive health effects. One well-known example is the long-chain polyunsaturated fatty acids (PUFAs), which are essential for human nutrition. Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) are the two most important long-chain omega-3 (ω-3) PUFAs involved in human physiology, and both industries are almost exclusively based on microalgae. In addition, algae produce phytosterols that reduce serum cholesterol. Here we determined the growth rates, biomass yields, PUFA and sterol content, and daily gain of eight strains of marine cryptophytes. The maximal growth rates of the cryptophytes varied between 0.34–0.70 divisions day−1, which is relatively good in relation to previously screened algal taxa. The studied cryptophytes were extremely rich in ω-3 PUFAs, especially in EPA and DHA (range 5.8–12.5 and 0.8–6.1 µg mg dry weight−1, respectively), but their sterol concentrations were low. Among the studied strains, Storeatula major was superior in PUFA production, and it also produces all PUFAs, i.e., α-linolenic acid (ALA), stearidonic acid (SDA), EPA, and DHA, which is rare in phytoplankton in general. We conclude that marine cryptophytes are a good alternative for the ecologically sustainable and profitable production of health-promoting lipids.

Methanotrophic activity in relation to primary and bacterial production in a boreal humic lake

Microbial decomposition of organic matter causes hypoxic and anoxic conditions in the hypolimnion of polyhumic lakes during summer and winter stratification periods. The concentration of eH4, an end product of anaerobic decomposition of organic matter, may as high as 250mmol m-3 in the hypolimnion of these Jakes (HUTTUNEN et al. 2002, HUOTARI & KANKAALA unpubl.). A large proportion of the eH4 produced is oxidized to eo2 by methanotrophs or converted to microbial biomass (Ruoo & HAMILTON 1974, BASTVIKEN et al. 2003), while the rest is degassed into the atrnosphere mainly during the spring and autumn turnover periods (RIERA et al. 1999). Stable carbon isotope (ô13C) composition of zooplankton has given rise to a hypothesis that methanotrophs may be a significant source of energy and carbon in the food webs o f humic lakes (JONES et al. 1999), where the algal primary production is insufficient to support the productivity of zooplankton (ÜJALA & SALONEN 2001). In 2002 we studied e~ oxidation in relation to primary and bacterial production in the water column of a boreal, polyhumic Lake Valkea-Kotinen. The lake has been intensively monitored since 1990 by Finnish Environment Institute and Lanuni Biological Station (see KESKITALO et al. 1998).

Simulated eutrophication and browning alters zooplankton nutritional quality and determines juvenile fish growth and survival

Abstract The first few months of life is the most vulnerable period for fish and their optimal hatching time with zooplankton prey is favored by natural selection. Traditionally, however, prey abundance (i.e., zooplankton density) has been considered important, whereas prey nutritional composition has been largely neglected in natural settings. High‐quality zooplankton, rich in both essential amino acids (EAAs) and fatty acids (FAs), are required as starting prey to initiate development and fast juvenile growth. Prey quality is dependent on environmental conditions, and, for example, eutrophication and browning are two major factors defining primary producer community structures that will directly determine the nutritional quality of the basal food sources (algae, bacteria, terrestrial matter) for zooplankton. We experimentally tested how eutrophication and browning affect the growth and survival of juvenile rainbow trout (Oncorhynchus mykiss) by changing the quality of basal resources. We fed the fish on herbivorous zooplankton (Daphnia) grown with foods of different nutritional quality (algae, bacteria, terrestrial matter), and used GC‐MS, stable isotope labeling as well as bulk and compound‐specific stable isotope analyses for detecting the effects of different diets on the nutritional status of fish. The content of EAAs and omega‐3 (ω‐3) polyunsaturated FAs (PUFAs) in basal foods and zooplankton decreased in both eutrophication and browning treatments. The decrease in ω‐3 PUFA and especially docosahexaenoic acid (DHA) was reflected to fish juveniles, but they were able to compensate for low availability of EAAs in their food. Therefore, the reduced growth and survival of the juvenile fish was linked to the low availability of DHA. Fish showed very low ability to convert alpha‐linolenic acid (ALA) to DHA. We conclude that eutrophication and browning decrease the availability of the originally phytoplankton‐derived DHA for zooplankton and juvenile fish, suggesting bottom‐up regulation of food web quality.
