Bioaccumulation of Heavy Metals in Blood and Tissue of Striped Mullet in Two Italian Lakes

Water and sediment from Faro and Ganzirri lakes were tested for cadmium, mercury, and lead. These heavy metals have been assessed in serum and muscles of Striped Mullet Mugil cephalus. A haemogram was performed to find effects of heavy metals on haematological variables. Students t-test showed higher Pb levels in sediments of Ganzirri Lake than Faro Lake. Two-way analysis of variance showed higher Cd and Pb concentration in muscles samples of Striped Mullet from Ganzirri Lake than from Faro Lake. Significant differences were found among haematological parameters as well. Mullet from Ganzirri Lake had lower white and red blood cell and trombocyte counts and higher mean corpuscular haemoglobin and mean corpuscular haemoglobin concentration than those from Faro Lake. Our results indicate a significant presence of lead in the environmental produces an accumulation of this metal in fish muscles.

Influence of different salinity on haematological and biochemical parameters of the widely cultured mullet, Mugil cephalus

This study investigates and reports on the effects of salinity levels on haematological and biochemical parameters of the widely cultured mullet, Mugil cephalus. For this purpose, 45 mullet caught in Faro Lake (Italy) were acclimated and fed with commercial diet for 20 days prior to the start of experiments that lasted for 20 days. After acclimation, fish were divided into three equal groups and subjected to 25‰ (Group A), 45‰ (Group B) and 35‰ of salinity (Group C), respectively. Statistical analysis and linear regression showed a significant influence of salinity levels on erythrocyte, haemoglobin, haematocrit, leucocytes and trombocytes and on all biochemical parameters except on osmolarity. These results add to our understanding of the effects of salinity levels on the physiological and biochemical parameters of Mugil cephalus and, in particular, improve our ability to select salinity levels for optimal growth for aquaculture and breeding of this species.

Effect of storage time on haematological parameters in mullet, Mugil cephalus

The aim of the present study was to assess the effect of storage time at +4 °C on red blood cell count (RBC), haematocrit (Hct), haemoglobin (Hb), white blood cell count (WBC), thrombocyte count (TC), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) in mullet (Mugil cephalus) using an automatic method. After blood collection (T0), all samples were analyzed using both the manual and automatic method. To test the validation of the automatic method, a paired t‐test was applied, and no statistical difference was observed. The samples were successively divided into four different aliquots and stored at +4 °C to assess the haematological parameters using the automatic method. The first aliquot was refrigerated for 6 h, the second one for 24 h, the third one for 48 h and the last one at for 72 h. One‐way repeated‐measures ANOVA showed a significant effect of storage time (P < 0.05) on Hb, WBC, TC, MCH and MCHC. These results suggest that haematological parameters can be assessed within 6 h from blood collection when samples are stored at +4 °C because long‐term storage modifies the results of the analyses. Further studies on these parameters could be still needed in various fish species to validate an appropriate method for haematological analysis useful not only for the evaluation of the health status of animal living in captivity and in aquaculture but also to have reliability environmental haematological biomarkers. Age-related changes of serum mitochondrial uncoupling 1, rumen and rectal temperature in goats

Thermoregulatory processes are induced not only by exposure to cold or heat but also by a variety of physiological situations including age, fasting and food intake that result in changes in body temperature. The aim of the present study was to evaluate the differences in serum mitochondrial uncoupling protein 1 (UCP1), rumen temperature (TRUMEN) and rectal temperature (TRECTAL) values between adult and kids goats. Ten adult male Maltese goats aged 3-5 years old (Group A) and 30 male kids, raised for meat, were enrolled in this study. The kids were equally divided into 3 groups according to their age: Group B included kids aged 3 months, Group C included kids aged 4 months and Group D included kids aged 5 months. Blood samples and measurements of TRUMEN and TRECTAL were obtained from each animal. One-way repeated measures analysis of variance (ANOVA) was applied to evaluate the effect of age on the studied parameters. Statistically significant higher serum UCP1 levels (P<0.001) were found in Group A as compared to Groups B, C and D. Higher TRUMEN values (P<0.001) were found in Group A than in Groups B, C and D, and in Group B than in Groups C and D. Group A showed lower TRECTAL values (P<0.001) than Groups B, C and D. The Pearsons Correlation test was applied to assess significant relationship among studied parameters showing a statistically significant negative correlation between the values of TRECTAL and serum UCP1 in all studied Groups (P<0.001). These results indicate that goats have good control of body temperature suggesting that further details about the thermogenic capacity and the function of UCP1 in kids and adult goats are worth exploring.

Dynamic modulation of platelet aggregation, albumin and nonesterified fatty acids during physical exercise in Thoroughbred horses

The effect of exercise on platelet aggregation, albumin and nonesterified fatty acids (NEFAs) values and the correlation among these parameters were evaluated in ten clinically healthy and regularly trained Thoroughbred horses. All horses were subjected to two simulated races. Blood samples were collected by jugular venipuncture before and after the first simulated race (T0PRE and T0POST), every 7 days at rest condition for a month (T1R-T2R-T3R), and before and after the second simulated race (T4PRE and T4POST) in order to assess platelet aggregation, albumin and nonesterified fatty acids (NEFAs) levels. One-way analysis of variance showed a significant effect of exercise (P<0.01) on platelet aggregation, albumin and NEFAs values. A negative correlation between platelet aggregation and albumin or NEFAs values, and a positive correlation between albumin and NEFAs values, were found both at T0POST and T4POST (P<0.05). These findings are likely related to dynamic physiological adaptations to exercise that allow re-establishment of the homeostatic equilibrium of the organism.

Leukocyte modifications during the first month after foaling in mares and their newborn foals

During early post-partum period both neonatal foals and peripartum mares are most susceptible to diseases. The aim of this study was to establish physiologic modifications of leukogram during the first month after foaling in mares and their newborn foals. To this end blood samples were collected from nine mares and nine foals (T0-T10), every three days from the 1st day until the 30th day after foaling. Samples were analysed for white blood cell (WBC) count and differential leucocyte counts. Two-way repeated measure analysis of variance (ANOVA) showed, in postpartum mares WBC showed significant higher values at T0 (9.02±0.76) in respect to other time points, and at T2 (8.08±0.53) and T3 (7.92±0.59) compared to T1 (6.98±0.43), whereas in foals lower WBC values at T0 (6.11±0.49) compared to other experimental periods except T1 (6.90±0.94), and at T1 compared to T8 (7.95±0.61) and T10 (7.90±0.36) were observed. The differential leucocyte counts showed significant modifications in the percentage of neutrophils (π<0.001) and lymphocytes (p<0.001) both in postpartum mares and in foals during the experimental period. Furthermore ANOVA showed significant differences between postpartum mares and foals (P<0.01) in all studied parameters, and between postpartum mares and control mares in WBC and neutrophils values. The obtained results provide suitable information about the influence of foaling on leukogram of periparturient mares and reveal WBC dynamics in newborn foals during the first month post-partum.

Photic entrainment of daily rhythm pattern of locomotor activity in sea bass (Dicentrarcus labrax)

The aim of this study was to evaluate the circadian rhythm of daily locomotor activity in sea bass exposed to three different artificial lighting regimes including 12/12, 24/0, and 0/24 L/D periods, and scheduled daytime feeding. Daily rhythm of locomotor activity during 12/12 L/D was observed, whereas locomotor activity recorded during 24/0 and 0/24 L/D resulted arrhythmic. Statistical differences in mesor values among the photoperiods and between light and dark phases of each photoperiod were found. During the 12/12 and 0/24 L/D, the fish were active mostly during the light phase. During 24/0 L/D, a phase inversion occurred. The results showed that photoperiod is a dominant synchronizer in sea bass, and that the appetite in this fish follows daily rhythms which does not match the imposed feeding schedule, suggesting the absence or the lability of internal process that couples feeding cycles and biological rhythms.

Application of a combined global positioning and heart rate monitoring system in jumper horses during an official competition - A preliminary study.

The aim of this study was to investigate whether a combined global positioning system (GPS)/heart rate (HR) monitoring system is a valuable tool to assess, step by step, the physiological response of HR and its relationship with speed in healthy horses competing in an official show jumping class. Six mares performing a standardised warm-up and jumping course were monitored using a HR/GPS device. Venous blood lactate (BL), assessed before and after exercise, showed a significant increase (P = 0.0004) following the physical effort. One-way analysis of variance (ANOVA) showed significant changes in HR throughout the experimental period. The analysis of HR data recorded during the warm-up jumping stage showed significantly higher HR (P = 0.001) in the recovery period compared to the related jumping phase. Shifting the fence height from 100 cm to 125 cm during the warm-up jumps was also found to cause a significant increase (P = 0.016) in HR. According to these preliminary results, the simultaneous logging of heart rate and speed has the potential to be a reliable and powerful technique for field testing that can help in the monitoring of the horses response to jumping effort during training and competition.

Peripheral blood and head kidney haematopoietic tissue response to experimental blood loss in mullet (Mugil cephalus)

Abstract Changes in haematological parameters and differential leukocyte counts of peripheral blood and head kidney haematopoietic tissue of mullet were assessed after an experimental anaemia, induced by caudal blood extraction. Thirty fish were used and divided into two groups: Group A (control group n = 20) and Group B (experimental group n = 10). From Group A, 10 fish were sacrificed to obtain head kidney control; from the 10 remaining, blood samples were collected by caudal venipuncture. From Group B, blood samples were collected five times once a week (T0, T1, T2, T3, T4) to induce experimental blood loss. At the end of the experimental period fish were sacrificed and the head kidneys were isolated. For all fish, haemogram and manual differential leukocyte counts were determined in peripheral blood. Reduced red blood cell count (RBC), haematocrit (Hct), haemoglobin (Hb), thrombocyte count (TC), lymphocytes and increased mean corpuscular haemoglobin (MCH), mean corpuscular haemoglobin concentration (MCHC), monocytes and neutrophil values were found at T1 with respect to T0. Successively, until T3, RBC, Hct, Hb, TC and lymphocyte values showed an increase in peripheral blood. A decrease of haematological parameters in peripheral blood after the last bleeding time (T4) was observed. Cell identification and differential counts on head kidneys were performed and a significant increase of lymphocytes, monocytes and early unidentified blast cells was observed. A decrease in RBC, neutrophils, eosinophils, TC and a total absence of basophils were found. These results indicated that mullet are able to recover from anaemia caused by various adverse environmental and/or nutritional factors.

The influence of acute handling stress on some blood parameters in cultured sea bream (Sparus aurata Linnaeus, 1758)

The effect of acute handling stress on haematological profile, blood glucose and lactate (secondary stress markers) in cultured sea bream Sparus aurata was evaluated. Sixty six Sparus aurata were used and equally divided into two groups (A and B). Group A was not subjected to stress, Group B was subjected to acute handling stress. From each fish, biometric data and blood samples were collected to evaluate haematological profile, blood glucose and lactate. Unpaired t-test Student was applied to evaluate possible differences in parameters between the two groups. Red blood cells, haematocrit, haemoglobin, white blood cells (WBC), glucose and lactate showed an increase (P<0.05) in Group B compared with Group A, while mean corpuscular volume decreased (P<0.05) in Group B. The results highlight the role of studied parameters in monitoring the stressful conditions of aquaculture production which affect animal welfare and fish products quality.
