Chronic exposure of 2,2′,4,4′-tetrabromodiphenyl ether (PBDE-47) alters locomotion behavior in juvenile zebrafish (Danio rerio)

In the present study, we used zebrafish (Danio rerio) as a model to address possible effects of chronic exposure of polybrominated diphenyl ether (PBDE) flame retardants on locomotion behavior, body size, and gonad development in fish. Zebrafish were fed food dosed with PBDE-47 (control, solvent control, low, medium, and high dose groups) from 21 days post hatch (dph) to 90 dph. Fish locomotion parameters, including maximum swimming speed, total distance moved, and percent time active, were assessed using a video-based animal movement analysis system. At the end of the exposure, all fish were euthanized for length and weight measurement, and then subjected to either whole fish histological analysis or tissue PBDE-47 measurement. Survival, body size, and gonad histology were similar between the five groups. However, both total swimming distance and percent time active were negatively correlated with tissue PBDE-47 concentration and were significantly lower in the high dose group. Tissue levels of PBDE-47 in the exposed fish were comparable to that reported in previous field studies. In summary, this study showed that developmental exposure of PBDE-47 at an ecologically relevant level altered locomotion behavior without affecting body size or gonad development of zebrafish.

Polybrominated diphenyl ethers and polychlorinated biphenyls in sediments of southwest Taiwan: Regional characteristics and potential sources

Very little information is available on the contamination of coastal sediments of Taiwan by PBDEs and PCBs. In this study, we determined the concentrations of 19 PBDE and 209 PCB congeners in 57 surface sediment samples to identify the possible sources of PBDEs and PCBs. The total PBDE and PCB concentrations ranged from below detection limit to 7.73 ng/g and 0.88-7.13 ng/g, respectively; these values are within the ranges observed for most coastal sediments worldwide. The PBDE congeners were dominated by BDE-209 (50.7-99.7%), with minor contributions from penta- and octa-BDEs. The signatures of PCB congeners suggested that PCB residues in Kaohsiung coast may be the legacy of past use or the result of ongoing inputs from the maintenance, repair and salvage of old ships. Principal component analysis of the congener-specific composition of PBDEs and PCBs revealed distinct regional patterns that are related to the use of commercial products.

Relationships between persistent organic pollutants and carbonaceous materials in aquatic sediments of Taiwan

Recent studies have shown that many persistent organic pollutants (POPs, e.g., polychlorinated biphenyls (PCBs), polycyclic aromatic hydrocarbons (PAHs) and various pesticides), are strongly associated with carbonaceous materials (including organic carbon (OC) and black carbon (BC)). We hypothesize that carbonaceous materials can be used as a first-order pollution index, to indicate areas where POP pollution may require further investigation. We tested our hypothesis and found that strong, positive correlations between BC and OC contents versus the concentrations of PCBs (and PAHs) existed in estuarine sediments of the Danshui River in 2005 and 2008. Thus, our preliminary results demonstrate that POC and BC are potential indicators of the POP pollution potential in fluvial sediments of the Danshui River in Taiwan. This innovative approach can provide a simple, relatively inexpensive and expedient means to monitor concentrations of POPs in polluted aquatic sediments of Taiwan, and/or those having a legacy of POP inputs.

Polycyclic aromatic hydrocarbons in surface sediments of the East China Sea and their relationship with carbonaceous materials.

This study measured concentrations of polycyclic aromatic hydrocarbons (PAHs) in surface sediments in the East China Sea (ECS) to investigate possible sources and fate of PAHs. Total concentration of PAHs in the sediments of the ECS ranged from 22 to 244 ng g(-1), with the highest levels in the coastal area and outer shelf. The observed PAH results showed elevated levels in both inner and outer shelf areas, a finding that is different from predictions by an ocean circulation model, suggesting that terrestrial sources are important for PAH contaminations in the ECS, while sediment resuspension, tidal changes and lateral transport may be important in affecting the distribution of PAHs in the outer shelf. The distribution of PAHs in the surface sediments of the ECS is similar to the distribution of carbonaceous materials (e.g., particulate organic carbon and black carbon), suggesting that carbonaceous materials may strongly affect the distribution of PAHs.

Air-water exchange fluxes of polycyclic aromatic hydrocarbons in the tropical coast, Taiwan.

Air-water exchange fluxes of polycyclic aromatic hydrocarbons (PAHs) were simultaneously measured in air and water samples from two sites on the Kenting coast, located at the southern tip of Taiwan, from January to December 2010. There was no significant difference in the total PAH (t-PAH) concentrations in both gas and dissolved phases between these two sites due to the less local input which also coincided to the low levels of t-PAH concentration; the gas and dissolved phases averaged 1.29±0.59 ng m(-3) and 2.17±1.19 ng L(-1) respectively. The direction and magnitude of the daily flux of PAHs were significantly influenced by wind speed and dissolved PAH concentrations. Individual PAH flux ranged from 627 ng m(-2) d(-1) volatilization of phenanthrene during the rainy season with storm-water discharges raising dissolved phase concentration, to 67 ng m(-2) d(-1) absorption of fluoranthene during high wind speed periods. Due to PAH annual fluxes through air-water exchange, Kenting seawater is a source of low molecular weight PAHs and a reservoir of high molecular weight PAHs. Estimated annual volatilization fluxes ranged from 7.3 μg m(-2) yr(-1) for pyrene to 50 μg m(-2) yr(-1) for phenanthrene and the absorption fluxes ranged from -2.6 μg m(-2) yr(-1) for chrysene to -3.5 μg m(-2) yr(-1) for fluoranthene.

Diffusive exchange of PAHs across the air–water interface of the Kaohsiung Harbor lagoon, Taiwan

Instantaneous air-water polycyclic aromatic hydrocarbons (PAHs) exchange fluxes were calculated in 22 pairs of ambient air and water samples from Kaohsiung Harbor lagoon, from December 2003 to January 2005. The highest net volatilization (3135 ng m(-2) day(-1)) and absorptive (-1150 ng m(-2) day(-1)) fluxes in the present study were obtained for the three-ring PAH phenanthrene on 7 April and 27 January 2004, respectively. All PAH diffusive fluxes for three-ring PAHs except phenanthrene were mainly volatilization exchange across the air-water interface. Phenanthrene and the four-ring PAHs were absorbed primarily from the atmosphere and deposited to the surface water, although some minor volatilization fluxes were also observed. Differences in flux magnitude and direction between the dry and wet seasons were also evident for PAHs. Strong absorptive/weaker volatilization PAH fluxes occurred in the dry season, but the opposite was found in the wet season. The mean daily PAH diffusive fluxes were an in flux of -635 ng m(-2) day(-1) in the dry season and an efflux of 686 ng m(-2) day(-1) in the wet season. The integrated absorbed and emitted fluxes of PAHs for harbor lagoon surface waters in the dry and wet seasons were 3.1 kg and 3.4 kg, respectively. Different from water bodies located in temperate zone, phenanthrene diffusive fluxes in Kaohsiung Harbor lagoon was favored in volatilization from surface waters during the wet season (April to September) because of scavenging by precipitation and dilution by prevailing southwesterly winds. In addition, this study used both of salinity and temperature to improve estimation of Henrys law constants (H) of PAHs in a tropical coastal area and show that correction for salinity produced 13-15% of differences in H values.

Seasonality of diffusive exchange of polychlorinated biphenyls and hexachlorobenzene across the air-sea interface of Kaohsiung Harbor, Taiwan.

Gaseous and dissolved concentrations of polychlorinated biphenyls (PCBs) and hexachlorobenzene (HCB) were measured in the ambient air and water of Kaohsiung Harbor lagoon, Taiwan, from December 2003 to January 2005. During the rainy season (April to September), gaseous PCB and HCB concentrations were low due to both scavenging by precipitation and dilution by prevailing southwesterly winds blown from the atmosphere of the South China Sea. In contrast, trace precipitation and prevailing northeasterly winds during the dry season (October to March) resulted in higher gaseous PCB and HCB concentrations. Instantaneous air-water exchange fluxes of PCB homologues and HCB were calculated from 22 pairs of air and water samples from Kaohsiung Harbor lagoon. All net fluxes of PCB homologues and HCB in this study are from water to air (net volatilization). The highest net volatile flux observed was +172 ng m(-)(2) day(-1) (dichlorobiphenyls) in December, 2003 due to the high wind speed and high dissolved concentration. The PCB homologues and HCB fluxes were significantly governed by dissolved concentrations in Kaohsiung Harbor lagoon. For low molecular weight PCBs (LMW PCBs), their fluxes were also significantly correlated with wind speed. The net PCB and HCB fluxes suggest that the annual sums of 69 PCBs and HCB measured in this study were mainly volatile (57.4 x 10(3) and 28.3 x 10(3) ng m(-2) yr(-1), respectively) and estimated yearly, 1.5 kg and 0.76 kg of PCBs and HCB were emitted from the harbor lagoon surface waters to the ambient atmosphere. The average tPCB flux in this study was about one-tenth of tPCB fluxes seen in New York Harbor and in the Delaware River, which are reported to be greatly impacted by PCBs.

Bioaccumulation of persistent organic pollutants in stranded cetaceans from Taiwan coastal waters.

This study focuses on analyzing PBDEs in the liver, muscle, and blubber tissues of stranded dolphins (Stenella attenuate) on the Taiwan coast to determine and compare the PBDE levels and distributions among tissue types. Total concentrations of 19 PBDEs (ΣPBDE) in male dolphins (9.97 to 436ng/g fat) were significantly higher than in female animals (2.73 to 89.5ng/g fat), implying gender variation in bioaccumulation and the possibility of generation transfer from mother to fetus during pregnancy. The levels of contamination varied among tissue type; contamination was higher in blubber than that in muscle or liver, suggesting a possible transformation and redistribution of these compounds in body burden. Aside from gender and tissue type, ΣPBDE concentrations also significantly correlated with body length, an indicator of dolphin age. PCA analysis results showed no significant difference in PBDE congener pattern distributions in blubber tissues, indicating that blubber may be the final storage of contaminants in cetaceans, and that bioaccumulation of PBDEs may be dependent on chemical properties. BDE-154 and BDE-47 were the predominant PBDE congeners in stranded dolphins, and their correlation with body length suggests the significant metabolic depletion of BDE-154 in this species and possible exposure to both penta-BDE and octa-BDE mixtures.

Kinetics of polychlorinated biphenyl partitioning to marine Chrysophyte Isochrysis galbana.

This study focused on the uptake kinetics of polychlorinated biphenyl (PCB) congeners by the Chrysophyte, Isochrysis galbana. A gas-purging experimental system was used to maintain constant dissolved PCB concentrations. Three phases of absorption were observed: first, a rapid absorption phase within the first 15min, second, a first order process reaching the maximum concentration within 48h of exposure, and third, a plateau phase as yet to be determined with very slight increases in concentration. In this study, the percentage of the maximum concentration reached within the first phase varied from 10% to 67%, depending on the size of the PCB (as determined by molecular weight and total surface area), whereas the uptake rate (k(u)) during the second phase was more comparable across different PCBs. In addition, for the first phase, the bioconcentration factor (BCF) of PCBs deviated from its expected relationship with hydrophobicity, as determined by K(ow), and was instead related to the molecular structure of the compound.

Assessing the effects of polychlorinated biphenyls (Aroclor 1254) on a scleractinian coral (Stylophora pistillata) at organism, physiological, and molecular levels.

Polychlorinated biphenyls (PCBs) are a group of widespread contaminants, and accumulation of PCBs has been observed in corals in the field. However, the toxic effects of PCBs on corals have not been investigated. In this study, we tested short and long term toxicity of Aroclor 1254, a commercial PCB mixture, on the scleractinian coral Stylophora pistillata. Coral nubbins were incubated in either control seawater or seawater dosed with PCBs (approximately 300ng/L) for 96h. The effect of PCB exposure on coral gene expression at 4h post exposure was tested with the suppression subtractive hybridization (SSH) and quantitative PCR methods. Photosystem II activity of the zooxanthellae was measured at 96h. After the exposure, nubbins were moved into clean seawater and their survival and growth were observed for another 50 days. All nubbins survived during the exposure and the following 50-d recovery period. Photosystem II activity and coral growth were not affected by PCB exposure in this study. Fifty-four clones were sequenced for gene expression analysis, and 15% of these sequences were identified, including genes involved in general stress response, peptide metabolism, cellular receptor, cytoskeleton organization, membrane trafficking, and oxidative stress response. However, the quantitative PCR did not show significant difference in the five selected genes. In conclusion, acute exposure of S. pistillata to Aroclor 1254 at 300ng/L did not affect coral survival, photosynthesis or growth but may alter the expression of certain genes involved in various important cellular functions. The nubbin technique proved to be an efficient approach to simultaneously characterize the impact of PCBs on the corals at multiple biological levels.
