Development of Microphalloides japonicus (Osborn, 1919) metacercariae in vitro (Trematoda: Microphallidae).

Metacercariae of the trematode Microphalloides japonicus (Osborn, 1919) were cultivated in various media to gravid adults, and the results were compared with those in vivo. A few eggs were produced in the uterus even in balanced salt solution, although cultivation in Eagles MEM and NCTC 109 media was much better. Addition of heat-inactivated serum resulted in even better development. Encysted metacercariae also produced eggs in some media, but most of them were incomplete and without shells.

Studies on metabolism of lung fluke genus Paragonimus. VII. Action of bithionol on glycolytic and oxidative metabolism of adult worms.

Abstract The present study was carried out in order to obtain information on the mechanism of action of bithionol on Paragonimus westermani (Kerbert 1878). Bithionol stimulated lactic acid production of intact adult worms above the level of the control worms, while it inhibited oxygen consumption of intact adult worms in vitro. Bithionol treatment of adult worms in vivo decreased glycolytic and oxidative metabolism of homogenates of uterine eggs and adult worms. Bithionol inhibited lactic acid formation except when fructose 1, 6-diphosphate (FDP) was used as a substrate in the homogenates of adult worms, and it also inhibited oxygen consumption of homogenates of eggs and adult worms. Bithionol inhibited reduction of methylene blue when succinate was used as a substrate. Bithionol inhibited oxidation of reduced cytochrome c in the 1000g supernatant of homogenates of adult worms. Bithionol inhibited activity of the succinate oxidation in homogenates of adult worms.

The cell content of peritoneal exudates following the injection of neutral thiol protease into guinea pigs

The cell content of the peritoneal exudate was examined 4 days after an intraperitoneal injection of glycogen in uninfected and Paragonimus westermani-infected guinea pigs. In uninfected animals a reduction in macrophage count and an accumulation of granulocytes in the exudate were observed at 3 and 6 hr after an intraperitoneal injection of purified neutral thiol protease from P. westermani metacercariae. No such effect occurred after the enzyme was injected into infected animals. At 9 hr after enzyme injection, vacuoles were found in the cytoplasm of macrophages in uninfected animals.

Studies on host specificity in Paragonimus westermani. II: Histochemical and cytochemical characterization of metacercariae and worms from rats and dogs

Histochemical tests were done on newly excysted metacercariae and worms recovered from an abnormal host (rat) and the definitive host (dog) for some oxidoreductases, phosphatases and glycosidases. The results demonstrate that rat worms have enzymatic distribution and intensities more similar to those of metacercariae than to adult worms from dogs. Ultracytochemical examination of acid and alkaline phosphatase and Mg-ATPase activity was also carried out. Acid phosphatase activity occurred exceptionally in the excretory bladder and caeca of dog worms. No activity was observed in rat worms except for lysosomal granules in the tegument. Alkaline phosphatase activity was exhibited in the excretory bladder in both dog and rat worms. Mg-ATPase activity occurred in the tegument and parenchymal cells in dog worms and in the excretory bladder in rat worms. In metacercariae, little or no reaction for these enzymes was present except for Mg-ATPase activity on the excretory ducts. These observations, together with the histochemical results, indicate that metabolic activity in rat worms is higher than in metacercariae although it is strongly reduced compared with dog worms.

Studies on metabolism of lung flukes of the genus Paragonimus. VI. Succinoxidase system in homogenates of eggs, larvae, and adults

Abstract The present study was concerned with the succinoxidase system in Paragonimus westermani, Paragonimus ohirai, and Paragonimus miyazakii. Potassium cyanide inhibited the motility of larval and adult forms. Succinate stimulated the reduction of methylene blue by homogenates of embryonated eggs, larvae, and adults, while malonate inhibited the reduction. Reduced cytochrome c was oxidized by the 1,000g supernatant from homogenates of embryonated eggs, larvae, and adults. The supernatant prepared from unembryonated eggs did not oxidize reduced cytochrome c. Succinate stimulated oxygen consumption by the homogenate of adult worms. Oxygen consumption markedly increased in the homogenate of adults when both succinate and cytochrome c were added as substrate to the reaction mixture, while malonate and cyanide inhibited oxygen consumption.
