Facile synthesis of α-aminonitriles

Abstract α-Aminonitriles were prepared by reacting α-trimethylsilyloxynitriles with various amines in methanol. The silyloxynitriles were obtained by condensing a wide range of aldehydes (aliphatic, aromatic and heteroaromatic) with trimethylsilyl cyanide (TMSCN) in the presence of a catalytic amount of zinc iodide.

A Fast N-Substituted α-Aminonitrile Synthesis

Abstract A series of α-aminonitriles bearing various functionalities was prepared by reacting a neat mixture of an aldehyde or ketone, an amine and trimethylsilyl cyanide. The reaction was essentially complete in 5 minutes.

Facile conversion of carboxamides to nitriles

Abstract Alkyl, aralkyl, aryl, heteroaryl carboxamides bearing various functionalities are readily converted to the corresponding nitriles in good yields using the liquid “diphosgene”, trichloromethyl chloroformate, as dehydrating agent. In many cases, the procedure does not require extraction, and hence offers a very simple work-up.

Asymmetric Synthesis of α-Aminonitriles

Abstract The asymmetric synthesis of α-aminonitriles was carried out by the reaction of α-trimethylsilyloxynitriles with optically active α-methylbenzylamine in methanol. The reactions provided moderate yields and good optical purities.
