Determination of a new cerebral vasodilator 2,6-dimethyl-4-(3-nitrophenyl)-1,4-dihydropyridine-3,5-dicarboxylic acid 3-[2-(N-benzyl-N-methylamino)]-ethyl ester 5-methyl ester hydrochloride (YC-93) in plasma by electron capture gas chromatography

A highly sensitive method for the quantitative determination of 2,6-dimethyl-4-(3-nitrophenyl)-1,4-dihydropyridine-3,5-dicarboxylic acid 3-[2-(N-benzyl-N-methylamino)]-ethyl ester 5-methyl ester hydrochloride (YC-93) in plasma is described. After extraction, YC-93 was oxidized to a pyridine analogue with nitrous acid and detected by electron capture gas chromatography. The sensitivity was 2-3 ng/ml, which is sufficient to determine plasma concentrations of YC-93 after oral administration of clinical doses to humans.

Quantitative determination of the β-adrenoceptor stimulant formoterol in urine by gas chromatography mass spectrometry

Abstract A method for the quantitative determination of the β-stimulant formoterol in urine, using a gas chromatograph—mass spectrometer, is described. Formoterol can be analyzed after the addition of a deuterium-labelled internal standard and conversion to a mixed bispentafluoropropionyl-methyl derivative for selected ion monitoring. The detection limit was 5 ng/ml. Urinalysis after the oral administration of formoterol fumarate, using a combined enzymic hydrolysis method, revealed that the drug was conjugated with glucuronic acid in rats, dogs and humans.

The efficacy of MR imaging in subdural empyema.

MRI findings of a 14-year-old boy with subdural empyema (SE) are reported and compared with those of serial CT-scan. He was admitted with fever, headache, right hemiplegia and facial palsy. Initial enhanced CT-scan revealed a slit left lateral ventricle and a shift in the mid-line structures, but failed to detect any SE. MRI at 10 days after admission clearly demonstrated SE as an area of low intensity on T1-weight (T1WI) and very high intensity on T2-weight (T2WI). Post-contrast enhanced MRI (CE-MRI), using Gd-DTPA, showed a contrast enhancement in the wall of SE. However, no definite parenchymal abnormal intensity areas were detected, suggesting that the diagnosis was made sufficiently early for timely treatment and good neurological outcome. CE-MRI proved to be a more powerful and better diagnostic procedure than enhanced CT-scan, and was very useful in determining the state and development of the disease.

Determination of the α,β-adrenocepor blocker YM-09538 in urine by gas chromatography with a nitrogen-sensitive detector

Abstract A gas chromatographic method for the quantitative determination of the α,β-adrenoceptor blocker YM-09538 in urine is described. YM-09538 was extracted from alkalinized urine with ethyl acetate and converted to its cyclic methylboronate derivative. Analysis by gas chromatography using a nitrogen-sensitive detector allowed quantitation of the drug over a concentration range of 0.2–5.0 μg/ml. Urinary excretion of YM-09538 was determined in humans after oral administration of 50 mg.
