A 2.99 kb mtDNA fragment containing two variable restriction endonuclease sites (EcoRV and Xbal) was subcloned and sequenced from the Mediterranean fruit fly (Ceratitis capitata). This fragment represents approximately one‐fifth of the entire mitochondrial sequence. The sequence was aligned with the comparable region from Drosophila yakuba and Anopheles gambiae, resulting in 81.8% and 76.7% identity at the nucleotide level, and 77% and 67.7% identity, respectively, at the amino acid level. The sequenced region includes the complete genes for NADH dehydrogenase 4, NADH dehydrogenase 4L, NADH dehydrogenase 6, and transfer RNAs for proline, threonine and histidine, and part of the genes for NADH dehydrogenase 5 and cytochrome b. Oligonucleotide primers were designed to asymmetrically bracket each of two variable restriction endonuclease sites to allow PCR amplification and subsequent restriction endonuclease analysis of individual fly samples.

Molecular genetic research on the Mediterranean fruit fly,Ceratitis capitata, will provide tools to permit determination of source populations for new pest infestations. Restriction fragment length polymorphism (RFLP) of mitochondrial DNA provides some interpopulation discrimination. A restriction map, including the informative variableEcoRV andXbaI restriction sites, is constructed for the Mediterranean fruit fly, and several restriction sites are associated with specific gene regions based on polymerase chain reaction-RFLP and sequence analyses. A partial sequence of the mitochondrial 16S ribosomal RNA gene is reported.

Abstract We analyzed sequence data from 107 base pairs within the nuclear 18S ribosomal RNA gene from 26 tephritid species and four out-group taxa. A total of 45 sites were variable within the Tephritidae, and 28 sites were considered informative for parsimony analysis. Phylogenetic information was extracted from this data set using maximum parsimony and neighbor joining methods and compared to a phylogenetic hypothesis proposed from the morphological literature. Our molecular data suggest: (1) monophyly of the subfamily Tephritinae: (2) membership of Gymnocarena; (3) the possible existence of a large monophyletic group including the subfamily Dacinae and a large part of the Trypetinae. We propose that molecular approaches show great promise for helping to create a phylogenetically-based higher classification for the Tephritidae.

A new genus, Cornutrypeta Han & Wang, is proposed for nine Palaearctic and Oriental tephritid species with enlarged male frontal setae: C. gigantocornuta Han & Wang, sp. n., C. melanonotum (Brunetti), comb. n., C. nigrifemur Han & Wang, sp. n., C. nigritata (Wang), comb. n., C. omeishana Han & Wang, sp. n., C. spinifrons (Schroeder), comb. n., C. superciliata (Frey), comb. n., C. triceratops (Bezzi), comb. n., and C. yushunia Han & Wang, sp. n. All the previously recorded species are transferred from Vidalia Robineau-Desvoidy. A lectotype is designated for C. superciliata. Cornutrypeta is placed in the newly recognized Trypeta group of the tribe Trypetini. A key to the species, descriptions, illustrations, and a cladogram are presented. We also resurrect Stemonocera Rondani, which was considered a synonym of Vidalia.

Abstract The Old World tephritid genus, Pseudina Malloch, is redefined with the descriptions of two new species from China: P. duplicata Han & Wang, and P. eritima Han & Wang. The nomenclatural status of Pseudina is clarified and the nominal genus Vidalia Robineau-Desvoidy is treated as a nomen dubium, and removed from synonymy with Pseudina. An annotated check list of all the known Pseudina species, a key, and illustrations are provided, and the phylogenetic relationships discussed. In addition, a lectotype is designated for Vidalia spadix Chen and Vidalia pulla Wang is transferred to the genus Hoplandromyia Bezzi.

Abstract A new tephritid genus, Paratrypeta, is recognized based on two species from China: P. flavoscutata Han & Wang, sp. nov., and P. appendiculata (Hendel), comb, nov., which was previously placed in Vidalia Robineau-Desvoidy. Their relationship is discussed, and descriptions are provided.
