Morphometric characterization of left ventricular myocardial cells of male rats during postnatal development

Abstract Morphometric investigations of the left ventricular myocardial cells of male Wistar rats aged 0, 1, 2, 3, 4, 5, 6, 7, 14, 21 days and 1, 2, 3, 4, 5, 6 months were carried out. The volume density of the sarcoplasm decreased from 0.33 (at birth) to 0.018 (6th month). The volume density of the myofibrils diminished during the first 7 days of life, whereas after the 6th month (0.57) it was nearly the same again as at birth (0.52). The volume density of the mitochondria increased from 0.16 at birth to 0.33 in the 6th month. The surface density of the outer mitochondrial membrane increased at different rates. The number of mitochondria per unit area grew from 0.51 (at birth) to 0.88 (6th month). The distance between the mitochondria decreased from 4.33 μm (at birth) to 1.67 μm (6th month). The mitochondrial/myofibrillar ratio doubled in the course of the period covered by the study (0.32 at birth; 0.64 in the 6th month). The postnatal development of heart muscle cells can be characterized by data obtained on the heart at birth, at the ages of 3, 4, 5, 7, and 14 days and in the 1st, 3rd and 6th month.

Zur Feinstruktur des experimentell erzeugten subakuten und chronischen Hirnödems

ZussamenfassungEs wurden die Veränderungen des Hirngewebes im Verlauf eines experimentell an Kaninchen durch Senkung des Liquordruckes erzeugten subakuten und chronischen Ödems elektronenmikroskopisch untersucht.Die stärksten Grade erreichte die Volumenvermehrung des Hirngewebes im Zeitraum von 1–2 Wochen.Das Ödem ist in Hirnrinde und Marklager gekennzeichnet durch eine hochgradige Anschwellung der Astrocyten und ihrer Fortsätze sowie durch eine deutliche Erweiterung des extracellulären Raumes in der weißen Substanz.Bereits nach 2 Tagen treten im Perikaryon der Astrocyten und in ihren Fortsätzen nach Zunahme der Polysomen Gliafilamente auf als Ausdruck einer nur den Astrocyten eigenen Reaktionsform. Die Gliafaserbildung erreicht ihr stärkstes Ausmaß bei einem 3–4 Wochen bestehenden Ödem.Als zweite reaktive Leistung der Astrocyten kommt es bei einem chronischen Ödem zu einer Anhäufung von Glykogengranula im Perikaryon und in den Zellfortsätzen.SummaryIn rabbits subacute and chronic edema were induced by lowering the pressure of the cerebral fluid. In the course of this experiment the alterations of the cerebral tissue were examinded by electron microscopy.Within a period of 1–2 weeks the volume of the cerebral tissue reached the greatest extent.The edema in cerebral cortex and white matter was characterized by an excessive swelling of astrocytes and their processes as well as by a distinct enlargement of the extracellular space in the white matter.Already two days later after an increase of polysomes, glial filaments appeared in the perikaryon of astrocytes and their processes as a reaction manifested only by astrocytes. The production of filaments of the glia cells reached its peak in edema existing for 3–4 weeks.In chronic edema glycogen granules accumulates in perikaryon and cellular processes as another sign of reactive capacity of astrocytes.

Elektronenmikroskopische Befunde während hypothermer Lagerungskonservierung der Leber

SummaryPorcine livers were preserved by simple hypothermic storage for 6 h using plasma-protein-fraction 5%, albumin 5%, Collins C2, and Ringer-lactate followed by a morphometric evaluation of the electron-microscopic changes. The morphometric parameters reflecting swelling of mitochondriae showed the most pronounced changes and were suited best for reproduction. The best result was achieved by using Collins C2-storage. The endoplasmic reticulum was found to be only slightly swollen in all preservation solutions investigated here, at the same time a degranulation of granular endoplasmic reticulum could be demonstrated. The increase in volume of lysosomes per unit-volume was not significant.ZusammenfassungSchweinelebern wurden durch einfache hypotherme Lagerung mit Plasma-Protein-Fraktion 5 % (PPF), Albumin 5 % (A), Collins C2 (C) und Ringer-Lactat (R) über 6 h konserviert. Es erfolgte eine morphometrische Auswertung der elektronenmikroskopischen Veränderungen. Die morphometrischen Parameter für die Schwellung der Mitochondrien sind am stärksten verändert und am besten reproduzierbar. Die geringsten Schädigungszeichen finden sich nach Collins-Lagerung. Das endoplasmatische Reticulum weist für alle Konservierungsarten eine geringe Schwellung auf, desgleichen ist eine Degranulation des granulären ER nachweisbar. Die Zunahme des Lysosomen-volumens im Einheitsvolumen ist nicht signifikant.

Vergleichende Untersuchung über die Abhängigkeit quantitativer Ergebnisse in der elektronenmikroskopischen Autoradiographie von der Art des Entwicklers (Microdol-X, Phenidon)

It was the aim of the investigation to compare quantitative results of electron microscopic autoradiographs after the use of different developers (Microdol X, Phenidon) adapting morphometrical methods. Erythroblasts of rabbit were tested after incorporation of 3H-uridine in vitro. Both developers showed the same tendency at higher rates of incorporation (basophilic erythroblasts and polychromatic erythroblasts). Using Phenidon, however, we could obtain only half the values for the individual cell components. At high rates of incorporation Microdol X results in an overlapping of the silver filaments. Thus, more distinct grains can be reached by Phenidon. At smaller rates of incorporation (orthochromatic erythroblasts) after using Phenidon a quantitative evaluation is however influenced by too many factors of uncertainty so that according to our investigations Microdol X is better suited for a quantitative evaluation of electron microscopic autoradiographs.

ZusammenfassungBei der elektronenmikroskopischen Untersuchung der Hirnrinde des Hundes nach experimenteller Sog- und Schlageinwirkung finden sich schon nach wenigen Minuten die charakteristischen Zeichen des Hirnödems. Die perivasculären Gliafortsätze sind geschwollen und können eine Ruptur der Zellmembran zeigen. Auch in capillarfernen Gebieten ist die Glia herdförmig ödematös geschwollen. Zusätzlich kommt es zum Austritt von geformten Blutbestandteilen insbesondere von Erythrocyten. Die Extravasation von Erythrocyten scheint bei der Schlagwirkung im Vordergrund zu stehen, während das Ödem nur zweitrangige Bedeutung hat.Die erhobenen Befunde werden hinsichtlich der Pathogenese der Contrecoup-Herde diskutiert.

Quantitative ultrastructural characterisation of rabbit erythroblasts and of 3H uridine incorporation into their nuclear components.

Electron microscopic-autoradiographic and morphometric investigations were carried out in erythroblasts from rabbit bone marrow. 1. to obtain quantitative data for the erythroblast classes. 2. to determine quantitative-morphometrically the incorporation rate of 3H uridine into the cell components of the different erythroblast classes. 3. to evaluate the influence of UV irradiation on the incorporation of 3H uridine. According to ultrastructural criteria erythroblasts are divided into 3 classes: basophilic erythroblasts (BE), polychromatic erythroblasts (PE) and orthochromatic erythroblasts (OE). The areas of the 3 classes of erythroblasts are in a ratio of 1:0.5:0.43. In all classes the portion of the nucleus is between 56 and 58.8%. The portion of mitochondria decreases from 13.3% (BE) to 9.3% (PE), and to 6.3% (OE) respectively. Simultaneously the portion of heterochromatin in the nuclear area rises from 24.2% (BE) to 49.8% (PE), and to 63.2% (OE) whereas the percentage of euchromatin decreased correspondingly. Nucleoli were found only in 78% of the BE and in 3.4% of the PE. For the quantitative determination of the silver grains and silver filaments, respectively, the morphometric procedures have been adapted to the particular conditions. So it was necessary to estimate the areas covered by silver grains of filaments. The silver grains or filaments localised in the nuclear area are in the range from 84.6 to 93.4%. The percentage of silver grains of the total cellular area is 14.4% in BE, 6.8% in PE and 4% in OE. In a similar way the relative values of the nucleolus (total) and the euchromatin and heterochromatin are reduced. UV irradiation of bone marrow in vitro for 10 to 20 min does not cause any significant changes in the incorporation of 3H uridine. This result is in contrast to other published experiments on the influence of UV on the RNA synthesis. It is assumed that the irradiation dose was too low to induce a measurable effect on the RNA synthesis or the point of investigation was too early.

Bei der elektronenmikroskopischen Untersuchung der Hirnrinde des Hundes nach experimenteller Sog- und Schlageinwirkung finden sich schon nach wenigen Minuten die charakteristischen Zeichen des Hirnodems. Die perivascularen Gliafortsatze sind geschwollen und konnen eine Ruptur der Zellmembran zeigen. Auch in capillarfernen Gebieten ist die Glia herdformig odematos geschwollen. Zusatzlich kommt es zum Austritt von geformten Blutbestandteilen insbesondere von Erythrocyten. Die Extravasation von Erythrocyten scheint bei der Schlagwirkung im Vordergrund zu stehen, wahrend das Odem nur zweitrangige Bedeutung hat. Die erhobenen Befunde werden hinsichtlich der Pathogenese der Contrecoup-Herde diskutiert.
