Inhibitory effect of incubation on microbial growth has extensively been studied in wild bird populations using culture-based methods and conflicting results exist on whether incubation selectively affects the growth of microbes on the egg surface. In this study, we employed culture-independent methods, quantitative PCR and 16S rRNA gene pyrosequencing, to elucidate the effect of incubation on the bacterial abundance and bacterial community composition on the eggshells of the Eurasian Magpie (Pica pica). We found that total bacterial abundance increased and diversity decreased on incubated eggs while there were no changes on non-incubated eggs. Interestingly, Gram-positive Bacillus, which include mostly harmless species, became dominant and genus Pseudomonas, which include opportunistic avian egg pathogens, were significantly reduced after incubation. These results suggest that avian incubation in temperate regions may promote the growth of harmless (or benevolent) bacteria and suppress the growth of pathogenic bacterial taxa and consequently reduce the diversity of microbes on the egg surface. We hypothesize that this may occur due to difference in sensitivity to dehydration on the egg surface among microbes, combined with the introduction of Bacillus from bird feathers and due to the presence of antibiotics that certain bacteria produce.

ABSTRACT Plumage coloration has been the subject for a variety of questions that comprise the center of modern evolutionary biology. Unlike carotenoids that the concentration directly influences the intensity of the color, melanin, in addition to produce brown or black colors, is often involved in producing the structural coloration such as glossiness or iridescence. As the melanin granules can be located in the barbs or the barbules, we aim to (i) discern if the colors observed at macro scale comes from the barbs, the barbules or both in a series of related species and (ii) estimate the evolutionary history of the color-producing mechanisms in the family Corvidae that are known to have melanin-based coloration. From a preliminary comparative analysis on eight representative species, we found three coloration schemes in Corvidae; (1) matte colors of brown or black that were produced in barbs and barbules; (2) non-iridescent structural colors such as blue, bluish gray and white, that were produced in the barbs and (3) iridescent structural colors that were produced only in distal barbules. Comparative character analysis of these coloration schemes suggests that the ancestral state among these species were the colors produced in the barbs and that the color produced in the distal barbules is a derived character. The evolution of iridescence seems tightly linked to the evolution of the colors produced in the distal barbules. Data from more species should be incorporated in order to grasp a full picture on the evolutionary history of plumage coloration in this group of birds.

Black-billed magpies (Pica pica) are a highly sedentary species due to their short and round wings, which are not adequate for long distance flights. We investigated geographic variation in the vocal signals of magpies residing in South Korea and Japan (subspecies sericea). Based on the magpie’s limited dispersal ability, we predicted that the variation in vocal signals of black-billed magpies could be explained by geographic barriers such as mountain ranges and straits. We analyzed four-syllable chatter calls of magpies from five localities in South Korea, which are separated by mountain ranges (Seoul, Daejeon, Daegu, Gwangju, and Busan), and two island localities separated from the mainland by straits (Jeju in Korea and Saga in Japan). We found significant differences in the characteristics of magpie chatter calls recorded in the seven localities, and the variation pattern was independent among the variables. Mainland-island differentiation and north-south differentiation were observed. In general, magpies on Saga were the most distinctive. North-south differentiation was observed among the Korean mainland localities. However, the pattern was not related to the presence of putative geographic barriers. We hypothesize that the patterns of geographic variation in the structure of magpie chatter calls residing in South Korea might have been shaped by a sudden expansion of magpies followed by low level of local isolation, which may have led to vocal differentiation. Along with elucidating the vocal environment of Korean magpie populations, more extensive sampling is needed to clarify the functional aspects of geographic variation in the vocal signals of Korean magpies.

Urban environments present novel and challenging habitats to wildlife. In addition to well-known difference in abiotic factors between rural and urban environments, the biotic environment, including microbial fauna, may also differ significantly. In this study, we aimed to compare the change in microbial abundance on eggshells during incubation between urban and rural populations of a passerine bird, the Eurasian Magpie (Pica pica), and examine the consequences of any differences in microbial abundances in terms of hatching success and nestling survival. Using real-time PCR, we quantified the abundances of total bacteria, Escherichia coli/Shigella spp., surfactin-producing Bacillus spp. and Candida albicans on the eggshells of magpies. We found that urban magpie eggs harboured greater abundances of E. coli/Shigella spp. and C. albicans before incubation than rural magpie eggs. During incubation, there was an increase in the total bacterial load, but a decrease in C. albicans on urban eggs relative to rural eggs. Rural eggs showed a greater increase in E. coli/Shigella spp. relative to their urban counterpart. Hatching success of the brood was generally lower in urban than rural population. Nestling survival was differentially related with the eggshell microbial abundance between urban and rural populations, which was speculated to be the result of the difference in the strength of the interaction among the microbes. This is the first demonstration that avian clutches in urban and rural populations differ in eggshell microbial abundance, which can be further related to the difference in hatching success and nestling survival in these two types of environments. We suggest that future studies on the eggshell microbes should investigate the interaction among the microbes, because the incubation and/or environmental factors such as urbanization or climate condition can influence the dynamic interactions among the microbes on the eggshells which can further determine the breeding success of the parents.

Abstract. The role of structural coloration, which is produced by the optical interactions among micro- and nanostructures in the feather barb or barbules, is still unclear in the context of sexual or social signaling, because the mechanism of color production is complex and the factors affecting it are not fully documented. We investigated whether structural colors represent class signals related to age, sex, and territory ownership in a social, sexually monochromatic species, the Eurasian Magpie Pica pica. We examined the reflectance spectra from white scapulars, bluish iridescent secondary and greenish iridescent tail plumage, as well as size of white scapular patch. Significant color differences between age classes were found in all measured plumage parts, with adults having plumage with higher color score, that is brighter, shorter wavelength-directed, and more saturated color, than young magpies. Color differences between males and females and between breeding adults (territorial owners) and non-breeding adults were only detected in the tail plumage. Size of white scapular patch did not differ between age and sex classes. Color differences among individuals belonging to different social classes may lessen agonistic confrontations. Sex differences in coloration may enable prompt sex recognition and thus facilitate pair formation. Higher tail color scores of adults, particularly males, support previous suggestions that the tail characteristics of avian species with relatively long tails represent a visual signal of the bearers quality.
