Influência do desenvolvimento etário e da suplementação com vitamina E (acetato de DL-alfa-tocoferol) no metabolismo oxidativo dos neutrófilos de bovinos da raça Holandesa (Bos taurus)

This study was carried out to evaluate the influence of age and supplementation with acetate of DL-alpha-tocopherol on the neutrophils oxidative metabolism of Holstein calves from birth to 150 days of age. Twenty calves were divided in two groups of ten animals. The animals of group Treatment received 2000UI of DL-alpha-tocopherol acetate, IM, at birth, and at 15, 30, 60, 90 and 120 days of age. The other group (Control), did not receive any supplementation. In both experimental groups the neutrophils oxidative metabolism showed little activity during the first 60 days of life, wich could indicate inefficiency of this important bactericidal mechanism. Neutrophils oxidative metabolism was not significantly affected by treatment with acetate of DL-alpha-tocopherol.

Inquérito soro-epidemiológico de Brucella ovis em rebanhos ovinos no semiárido baiano

In order to analyze the occurrence of antibodies to Brucella ovis in sheep in a semi-arid region, this survey was conducted in the micro-region of Juazeiro, Bahia State, Brazil. The agar gel immunodifusion test (AGID) was used to examine 694 serum samples of 58 flocks. Antibodies to B. ovis were found in 5 (0.72%) of the investigated animals. It is believed that this low number of positive cases is related to the characteristics of the production. The predominant system is the extensive one, with presence of locally adapted breeds and crossbred animals, aiming at the production of meat and skin, with low productivity rates and technical development.

Fatores etários no leucograma de fêmeas zebuínas sadias da raça Nelore (Bos indicus)

In order to evaluate the influence of the age on the white blood cell counts of Nelore (Zebu) cattle, 158 blood samples from seven groups of different ages (group I-up to three months; group II-three to six months; group III-six to 12 months; group IV-12 to 24 months; group V-24 to 48 months; group VI-48 to 72 months and group VII-older than 72 months) were studied. The highest and lowest mean values observed were total white blood cell/mm3-16992±4104 (group III) and 10353±2397 (group VI); neutrophils/mm3-3931±1578 (group I) and 2416 ±1118 (group III); eosinophils/ mm3-999±499 (group V) and 265±276 (group II); basophils/mm3-67±88 (group VII) and 39±78 (group III); typical lymphocytes/ mm3-12758±3608 (group III) and 5906±1883 (group VI); atypical lymphocytes/ mm3-1310±603 (group II) and 760±419 (group VI) ; lymphocytes total/ mm3-14079±4027 (group III) and 6666±2059 (group VI); monocytes/mm3-27±62 (group I) and 0 (group III). Múltiplas estirpes de isolados de Mycobacteriumbovis identificados por tipagem molecular em bovinos abatidos em matadouros-frigoríficos

The aim of this study was to use bacteriological and molecular methods to identify Mycobacteriumbovis in lesions observed in cattle carcasses during routine post-mortem inspection in slaughterhouses with official inspection service. It was accompanied the slaughter and inspection of 825,394 cattle, healthy ante mortem examination by the official inspection service in ten slaughterhouses in the state of Bahia. Carcasses of 180 cattle presented lesions suggestive of tuberculosis and other lymphadenitis. In bacterial isolation, 25 samples showed dysgonic growth of colonies of creamy-yellow in medium-Stonebrink Leslie. From these isolates, 14 were identified as M. bovis and the multiplex PCR technique spoligotyping was discriminated against eight different spoligotypes of M. bovis, seven previously described in the literature and a new spoligotypes without former description. The major spoligotypes was SB0121, with five samples which has been described in Brazil and other countries, followed by two clusters, SB295 and SB1055, with two isolates each. The SB1145 and SB1648 spoligotypes were reported only in Brazil and Denmark, respectively. The spoligotypes SB140 has been found in Brazil, Argentina, Uruguay and Paraguay. These results demonstrate that the spoligotypes obtained are shared, so far, among Brazilian states and among Latin America and Europe. Thus, molecular discrimination of isolates of M. bovis by Spoligotyping constitutes a tool for epidemiological studies of bovine tuberculosis in the state of Bahia.

Identificação de Mycobacterium bovis em carcaças de bovinos abatidos no estado da Bahia, Brasil, por métodos bacteriológico e molecular

The aim of the present study was to perform bacteriological and molecular methods for identification of Mycobacterium bovis in lesions derived from bovine carcasses detected during routine post-mortem examination in officially inspected slaughterhouses. We checked the slaughter and inspection of 825,394 bovines, health upon ante-mortem examination, by the official service in 10 slaughterhouses of Bahia state from April, 2009 to April 2012. Lesions suggestive of tuberculosis were collected from 180 bovines and further evaluated by bacteriology and multiplex PCR. The majority of lesions were located in the respiratory tract lymph nodes and 71% were from male bovines up to 32 months old. 13.9% of samples presented small, granular and creamy-yellowish colonies after being cultured in Stonebrink-Leslie with an average growth time of 34 days. All smears from the isolated samples were Acid Fast Bacilli (AFB) and among them 56% were identified by mPCR as M. bovis. Thus, the association between post-mortem examination, culture and multiplex PCR allowed the bacillus identification in a reduced time and in regions of low prevalence, pointing out its importance for bovine tuberculosis detection and as a supportive tool for the tuberculosis control and eradication program.
