The Relationship between the Serum Vitellin Reaction and the Disc Electrophoretic Analysis in Estrogen Treated Chickens

エストロゲン投与雛血清の電気泳動分析と血清ビテリン反応との関係を明らかにする為に,60日齢白色レグホーン種雛にエストロゲンを11日間投与し,得られた血清および超遠心分離により分離された血清試料につきディスク電気泳動を実施し,並行して血清ビテリン反応観察を行った.デイスク電気泳動は7.5%,3.75%アクリルアミドゲルを使用し,アミドブラックおよびズダンブラック染色を行い,泳動像と各分画の濃度を記録した.また泳動後のゲルを各分画ごとに切断し,8%食塩水中で溶出させ,その溶液で血清ビテリン反応を観察した.その結果は次の通である.1) 7.5%ゲル泳動アミドブラック染色:I~XIVの分画に分かれ,I,IV,VII,VIII,Xの分画で血清ビテリン反応との間に相関関係が認められた.Iの分画では0.809,Xの分画では0.524の正の相関,IV,VII,VIIIの分画ではそれぞれ-0.553,-0.525,-0.491の負の相関であった.2) 3.75%,7.5%の重層ゲル泳動:1~13の分画に分離し,血清ビテリン反応との相関は1の分画で0.917,9の分画で-0.851,10の分画で-0.884であった.これら1,9,10の分画はズダンブラックでも染色され,分画1は上記の分画Iと対応するものと考えられた.3) 各分画の溶出液による血清ビテリン反応:1の分画で微弱ではあるが反応が観察された.以上の結果および従来の種々の報告の結果から考察して,1の分画および10の分画はそれぞれβ-,α-リポ蛋白と推測された.またエストロゲン投与雛血清の血清ビテン反応の強さはβ-リポ蛋白が量的に増大し,α-リポ蛋白が減少した時に強く現われるものと考えられた.
