Effects of hydrodynamics on the distribution of trace persistent organic pollutants and macrobenthic communities in Bohai Bay

In recent years, the rapid economic development along Bohai Bay, has brought out continuous increasing of the pollution loads in the Bohai Sea, especially by the large coastal reclamation project, Tianjin Binhai New Area. In the period of 2007-2009, we collected the sediments of the main rivers, estuaries, intertidal zone, and near-shore area of Bohai Bay, and Macrobenthos associated with the marine sediments to assess the influence of hydrodynamics in the coastal environment on the pattern of trace contaminants and the macrobenthic community. Based on data derived from these samples, the levels of polycyclic aromatic hydrocarbons (PAHs), organochlorine pesticides (OCPs), polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs) in sediments followed the order PAHs>OCPs>PCBs>PBDEs. The higher concentrations of PCBs, OCPs, PAHs and PBDEs were found in the estuarine and near-shore environment of the Dagu Drainage River. The spatial distribution of OCPs was different to that of PCBs due to the direction of the velocity field of Bohai Bay in its old and new topography, and the higher water-solubility of OCPs than that of PCBs. The results of the Pearson correlation and the PCA indicate that the medium diameter (MD) of sediments was the predominant factor influencing the distribution of PCBs and OCPs, most sampling sites were characterized mainly by TOC of sediments and biomass of macrobenthos. The results indicate that the distribution of trace contaminants and macrobenthic community in Bohai Bay are mainly affected by the hydrodynamic conditions.

Use of heat shock protein mRNA expressions as biomarkers in wild crucian carp for monitoring water quality.

In organisms, the production of heat shock proteins (HSPs) can be induced after exposure to various stressors. In the present study, partial cDNA sequences encoding of HSP30, HSP60, HSP70, and HSP90 genes were isolated and sequenced in the crucian carp (Carassius carassius). Next, quantitative real-time reverse transcription PCR (qRT-PCR) assays normalized to beta-actin were developed to quantify HSP mRNA expression. The same methods were used to detect HSP mRNA expression in the wild crucian carp that were collected from the Hun River. In fish located downstream of the river, we found that the levels of HSP70 and HSP30 in the liver and kidney were higher than fish located upstream, and these differences coincided with changes of the water quality. These results suggest that kidney HSP30 and liver HSP70 expression can serve as sensitive biomarkers for the presence of field environmental stressors in wild crucian carp.

Biomarker responses and genotoxicity in the mud snail (Bullacta exarata) as indicators of coastal contamination.

In the present study different biomarker responses and genotoxicity were determined in wild mud snails (Bullacta exarata) collected from 3 sampling sites in Bohai Bay in northeastern China, which is a region that is under considerable anthropogenic influence. Significant spatial variability of superoxide dismutase (SOD) and metallothionein (MT)-like proteins were recorded, while glutathione transferase (GST), catalase (CAT), and vitellin-like proteins (Vns) were not observed. Furthermore, genomic DNA that was amplified with 4 fluorescence-labeled primer pairs showed variable genetic distances among the 3 wild mud snail populations found in Bohai Bay, which may be correlated with differences in the types of environmental genotoxicants, such as heavy metals and polycyclic aromatic hydrocarbons. This multi-biomarker approach provided an improved understanding of the potential toxicological impact of contaminated sediments on benthic organisms.

Ribosomal protein L7 as a suitable reference gene for quantifying gene expression in gastropod Bellamya aeruginosa

Expression levels of eight candidate reference genes were quantified in tissues of gastropod Bellamya aeruginosa exposed for 10 d to various stressors, including fasting, 17β-estradiol, 17α-methyltestosterone, and Cd(2+). The results showed that 18s rRNA was the most highly expressed of the candidate reference genes, while H2A was the least expressed. There were no significant changes (p>0.05) in the expression of the eight genes in tissues among the different treatments. Using RefFinder to evaluate the expression stabilities of the eight candidate reference genes, ribosomal protein was shown to be the most stable reference gene, and no effects were observed among the different stressor treatments. These results indicate that RPL 7 is the most suitable reference gene for quantifying gene expression in B. aeruginosa under environmental stress, which was verified in B. aeruginosa exposed to high doses of E2 for 24 and 72h.

Integrated biomarkers in wild crucian carp for early warning of water quality in Hun River, North China

Metabolizing enzymes play important roles in the detoxification of various pollutants in aquatic organisms, thereby they can also be used to provide early-warning signals of environmental risks. Real-time quantitative reverse-transcription polymerase chain reaction assays were developed to quantify cytochrome P450 1A (CYP1A), superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT), and glutathione-S-transferase (GST) in crucian carp (Carassius auratus). The methods were then used to detect the respective mRNA expression levels in liver tissue in wild crucian carp from the Hun River, North China. CYP1A mRNA expression was significantly up-regulated in fish from stations S5, S6, and S8 (p < 0.05). SOD mRNA expression was significantly down-regulated in downstream areas relative to fish from upstream sites (p < 0.05); GPx and CAT mRNA expression levels were also down-regulated at S9 (p < 0.05). In contrast, GST mRNA expression showed no obvious change between fish collected from up- or downstream areas of the river. Finally, an integrated biomarker response was used to evaluate the integrated impact of pollutants in the Hun River and allow better comprehension of the real toxicological risk of these investigated sites.

Preliminary evidence for snail deformation from a Eutrophic lake

The incidence of deformities in snails Bellamya aeruginosa was investigated in a typical eutrophicated lake - Taihu Lake. A total of 15 105 specimens were collected, and 0.18-0.93% of the snails exhibited abnormal tentacle bifurcations. Abnormally developed snails were all female and were found in regions with relatively high Chlorophyll a levels (12.40±7.23μg/L). As tentacles are sexually dimorphic in B. aeruginosa, we postulated that factors associated with eutrophication might be responsible for the partial masculinization of tentacles in females. Differential gene expression analyses revealed that a number of unigenes were significantly up-regulated or down-regulated in snails sampled from three locations having high Chlorophyll a levels compared with snails sampled from the region with lower Chlorophyll a level (2.95μg/L). Thus, transcriptomic profiling revealed potential molecular signal of eutrophication that can lead to developmental abnormalities in this species.

No impacts of microcystins on wild freshwater snail Bellamya Aeruginosa fecundity from a eutrophic lake

The preliminary investigation at shoreline along Taihu lake with different degrees of eutrophication status found no significant relationship between the microcystin-LR concentrations and the freshwater snail Bellamya aeruginosa fecundity or the abundance of wild freshwater snails. To further confirm the impact of eutrophication on the reproductive ability of snails, ecological mesocosm experiments were employed at four sites in Taihu lake during the algal blooming period, and no significant relationship was also found between MC-LR concentrations and snail fecundity. These results implied that eutrophication does not negatively or positive affect snail fecundity in Taihu Lake, a typical eutrophication lake in China.
