Probiotic supplementation protects weaned pigs against enterotoxigenic K88 challenge and improves performance similar to antibiotics

These studies evaluated the effects of probiotics (PB) as a potential substitute for antibiotics (AB) on diarrhea in relation to immune responses and intestinal health in weaned pigs challenged with enterotoxigenic (ETEC) K88 (Exp. 1) and the effects of PB on performance and nutrient digestibility in weaned pigs (Exp. 2). In Exp. 1, 24 weaned barrows (4.9 ± 0.4 kg initial BW) were randomly assigned to 1 of 4 treatments. The treatments consisted of pigs fed an unsupplemented corn-soybean meal basal diet and not challenged (NON-C) or challenged with ETEC K88 (CHA-C) on d 9 and pigs fed the same basal diet supplemented with AB (100 mg/kg zinc bacitracin, 50 mg/kg colistin sulfate, and 100 mg/kg olaquindox; CHA-AB) or 500 mg/kg PB ( and ; CHA-PB) and challenged with ETEC K88 on d 9. In Exp. 2, 108 weaned pigs (7.5 ± 0.9 kg initial BW) not challenged with ETEC K88 were randomly assigned to 1 of 3 treatments, including an AB-free basal diet (CON) and the basal diet with AB (ABD) or 500 mg/kg PB supplementation (PBD). In Exp. 1, after challenge, CHA-C decreased ( < 0.05) ADG and ADFI, whereas CHA-AB and CHA-PB revealed no significant change compared with NON-C. Compared with CHA-C, CHA-AB and CHA-PB improved ( < 0.05) ADG and ADFI and decreased ( < 0.05) the diarrhea incidence in pigs. Mucosal secretory Ig A contents in the jejunum and ileum were greater in CHA-C than in NON-C ( < 0.05) and lower than in CHA-PB ( < 0.05). The diet containing PB alleviated the increase in the endotoxin and diamine oxidase concentration and cecal count ( < 0.05) and the decrease in intestinal villus height, cecal count, and jejunal mucosal occludin protein abundance ( < 0.05). In Exp. 2, dietary supplementation with AB and PB had positive effects on ADG and feed efficiency ( < 0.05). Compared with CON, apparent digestibility of nutrients in PBD was improved ( < 0.05). Collectively, PB supplementation protected the pigs against ETEC K88 infection by enhancing immune responses and attenuating intestinal damage and improved the performance and nutrient digestibility of weaned pigs. Therefore, PB could be a potential effective alternative to AB for ameliorating diarrhea and improving performance in weaned pigs.

Tannin is a key factor in the determination and prediction of energy content in sorghum grains fed to growing pigs.

This study was conducted to determine and compare the DE and ME content as well as the apparent total tract digestibility (ATTD) of GE in 28 samples of sorghum grain fed to growing pigs and to generate prediction equations for DE, ME, and the ATTD of GE based on chemical analysis. The 28 samples included 8 low-tannin sorghum cultivars (LTS; tannin ≤ 0.16%), 10 medium-tannin sorghum cultivars (MTS: 0.67% ≤ tannin ≤ 0.98%), and 10 high-tannin sorghum cultivars (HTS; 1.11% ≤ tannin ≤ 1.51%) obtained from 9 provinces in China and 3 samples of LTS cultivars were obtained internationally. The 28 diets were formulated to contain 96.9% of 1 of the 28 sorghum grains and 3.1% of vitamins and minerals. Eighty-four barrows (36.5 ± 3.4 kg initial BW) were used in a randomized complete block design conducted during 2 consecutive periods ( = 6 per treatment) with the pigs fed different diets during the 2 periods. For each period, pigs were placed in metabolic crates for a 5-d total collection of feces and urine following 7 d of adaptation to the diets. Among the 28 samples, GE, CP, and ADF content (DM basis) ranged from 4,363 to 4,496 kcal/kg, 8.26 to 13.41%, and 3.26 to 5.78%, respectively. The average tannin content was 0.08, 0.84, and 1.29% in the LTS, MTS, and HTS samples, respectively. The DE and ME content (DM basis) ranged from 3,861 to 3,989 and from 3,784 to 3,905 kcal/kg, respectively, in LTS; from 3,646 to 3,740 and from 3,588 to 3,669 kcal/kg, respectively, in MTS; and from 3,481 to 3,617 and from 3,418 to 3,538 kcal/kg, respectively, in HTS. The mean DE, ME, DE/GE, ME/GE, and ATTD of GE in MTS were less than those in LTS ( < 0.05) and greater than those in HTS ( < 0.05). The correlation coefficients between DE, ME, or the ATTD of GE and tannin content were all -0.96 ( < 0.01). The best-fit equations for DE and ME (kcal/kg DM) for the complete set of 28 sorghum grains were 6,974 - (236 × % tannin) - (43.27 × % ADF) + (24.96 × % CP) - (0.71 × % GE) for DE and 3,973 - (262 × % tannin) - (27.24 × % ADF) for ME. Our results strongly indicate that the DE and ME content of sorghum grains are related to their chemical composition, especially their tannin content. The DE and ME content could decrease by more than 200 kcal/kg when the tannin content is increased by 1% in the sorghum grain. Overall, tannin is a key predictor for the DE and ME content as well as the ATTD of GE in sorghum grains, and the potent antinutritive properties from tannin should not be ignored when sorghum is used as an energy source for pigs.

Forsythia suspensa extract attenuates lipopolysaccharide‐induced inflammatory liver injury in rats via promoting antioxidant defense mechanisms

Reactive oxygen species (ROS) have been shown to have a role in inflammation. We investigated whether Forsythia suspensa extract (FSE) could exert its antioxidant potential against lipopolysaccharide (LPS)-induced inflammatory liver injury in rats. Rats were orally fed FSE once daily for 7 consecutive days prior to LPS (Escherichia coli, serotype O55:B5) injection. LPS treatment caused liver dysfunction as evidenced by massive histopathological changes and increased serum alanine aminotransferase and aspartate aminotransferase activities which were ameliorated by FSE pretreatment. FSE attenuated LPS-induced depletion of cytosolic nuclear factor-erythroid 2-related factor 2 (Nrf2) and suppression of Nrf2 nuclear translocation in liver, and the generation of ROS and malondialdehyde in serum and liver. FSE increased the Nrf2-mediated induction of heme oxygenase-1 in liver, as well as superoxide dismutase and glutathione peroxidase activities in serum and liver. Importantly, FSE attenuated LPS-induced nuclear factor-кB (NF-кB) nuclear translocation in liver, and subsequently decreased tumor necrosis factor-α, interleukin (IL)-1β and IL-6 levels in serum and liver, which were associated with FSE-induced activation of Nrf2 in liver. These results indicate that the protective mechanisms of FSE may be involved in the attenuation of oxidative stress and the inhibition of the NF-кB-mediated inflammatory response by modulating the Nrf2-mediated antioxidant response against LPS-induced inflammatory liver injury.

A new Buttiauxella phytase continuously hydrolyzes phytate and improves amino acid digestibility and mineral balance in growing pigs fed phosphorous-deficient diet

Ten ileal T-cannulated pigs (19.26 ± 1.06 kg) were used to evaluate the effects of a novel phytase on apparent ileal digestibility (AID) of AA and apparent total tract digestibility (ATTD) and hindgut disappearance of DM, GE, CP, crude fiber, NDF, and ADF as well as minerals balance. Pigs were fed in a duplicated 5 × 4 incomplete Latin square design (5 diets with 4 periods). Each period consisted of a 5-d adjustment period followed by a 3-d total collection of feces and urine and then a 2-d collection of ileal digesta. The 5 diets included a P-deficient basal diet (0.43% Ca and 0.38% total P) that was supplemented with 0 (negative control [NC]), 500, 1,000, or 20,000 phytase units (FTU)/kg phytase and a positive control (PC) diet that was P adequate (0.64% Ca and 0.52% total P). The addition of phytase to the NC diet improved ( < 0.05) AID of phytate from 11.1 to 62.8, 70.6, and 90.5% at the inclusion rates of 500, 1,000, and 20,000 FTU/kg, respectively. In general, phytase supplementation at a dose of 20,000 FTU/kg further increased ( < 0.05) AID of Ca, total P, and phytate and reduced ( < 0.05) the ileal phytate concentration compared with diets with 500 or 1,000 FTU/kg phytase. Pigs fed the diet with 20,000 FTU/kg phytase but not diets with 500 and 1,000 FTU/kg phytase showed improved ( < 0.05) ATTD of CP and AID of DM, GE, CP, Leu, Lys, Thr, Val, Asp, and Ser compared with pigs fed the PC or NC diet. However, hindgut disappearance of crude fiber and NDF ( < 0.05) were reduced in pigs fed the diet with 20,000 FTU/kg phytase compared with pigs fed the PC or NC diet. Pigs fed diets with 500 or 1,000 FTU/kg phytase had greater ATTD and retention of Ca and P than pigs fed the NC diet but less compared with pigs fed the diet with 20,000 FTU/kg phytase. Supplementation of 20,000 FTU/kg phytase to the NC diet improved ( < 0.05) digestibility of Na, Mn, and Zn as well as retention (%) of Zn. Increasing phytase supplementation doses from 0 to 1,000 FTU/kg linearly improved ( < 0.05) retention of Mg; meanwhile, digestibility of Mg and Mn and AID of Thr showed a linear increase trend ( = 0.084). In conclusion, supplementation of the novel phytase at doses up to 20,000 FTU/kg hydrolyzed most of the phytate (90%) and consequently further improved mineral and protein utilization.

Effects of coated compound proteases on apparent total tract digestibility of nutrients and apparent ileal digestibility of amino acids for pigs.

Two experiments were conducted to evaluate effects of coated compound proteases (CC protease) on apparent total tract digestibility (ATTD) of nitrogen (N) and energy, and apparent ileal digestibility (AID) of amino acids (AA) and nutrients in diets for pigs. In Exp. 1, 12 crossbred barrows (initial body weight: 20.14±1.71 kg) were housed in individual metabolism crates and allotted into 2 treatments with 6 piglets per treatment according to weight in a randomized complete block design. The 2 diets were corn-soybean meal basal diets with (0.2 g/kg) or without CC protease supplementation. The CC protease supplementation increased (p<0.05) the digestible and metabolizable N and energy values and the digestibility and retention rate of N in the diet. The ATTD of energy and nutrients had been improved (p<0.05) in the diet supplemented with CC protease. In Exp. 2, 12 crossbred barrows (initial body weight: 20.79±1.94 kg), fitted with T-cannulas at the distal ileum, were blocked by body weight into 2 groups with 6 pigs each. The diets were the same as those in Exp. 1. The CC protease increased (p<0.05) the AID of crude protein and some essential AA including arginine, isoleucine and leucine. The AID and ATTD of energy and nutrients had been improved (p<0.05) by supplemental CC protease, but the hindgut digestibility of nutrients was unaffected. Overall, the CC protease improved the ATTD of N and energy and AID of some indispensible AA and nutrients in the corn-soybean meal diet for pigs. Therefore, the CC protease supplement could improve the utilization of protein in the corn-soybean meal diet and thus contribute to lower N excretion to the environment.

Concentration of digestible and metabolizable energy, standardized ileal digestibility, and growth performance of pigs fed diets containing sorghum produced in the United States or corn produced in China

The DE and ME content (Exp. 1) as well as the apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of essential AA (EAA; Exp. 2) were compared between Chinese corn and U.S. sorghum. The effects of U.S. sorghum as a potential substitute for Chinese corn on growth performance of 114 weaned pigs (8.8 ± 1.0 kg BW; Exp. 3) and 60 growing pigs (23.4 ± 1.6 kg BW; Exp. 4) were evaluated, and the effect of protease supplementation on N utilization was determined in sorghum-based diets fed to growing pigs (Exp. 4). In Exp. 1, there was no difference in DE and ME content between corn and sorghum. In Exp. 2, the AID and SID of most EAA and the concentrations of standardized ileal digestible Lys, Met, Thr, and His were less in sorghum than in corn ( < 0.05). In Exp. 3, there was no difference in ADG and ADFI among treatments during the experimental period. The G:F and apparent total tract digestibility (ATTD) of CP was decreased for pigs fed diets with sorghum in the first 2 wk ( < 0.05) and for pigs fed diets containing 60% sorghum in the following 2 wk ( < 0.05). The fecal score for pigs fed diets with sorghum, regardless of the substitute level, was less ( < 0.05) or tended to be less ( = 0.086) than that for pigs fed diets containing 60% corn. In Exp. 4, no differences were observed in ADG and ADFI overall among pigs fed diets based on corn and soybean meal (CSBM) or sorghum and soybean meal (SSBM). Pigs fed CSBM or SSBM with protease supplementation had greater ( < 0.05) or tended to have greater ( = 0.062) G:F than pigs fed SSBM. Compared with CSBM, SSBM increased fecal N excretion by more than 25% and decreased the ATTD of CP by more than 7% during the whole experiment ( < 0.05). Protease supplementation reduced fecal N excretion by more than 12% and increased ATTD of CP by more than 6% ( < 0.05). In conclusion, based on optimal G:F and CP digestibility, diets for weaned pigs should contain less than 20% sorghum during the first 2 wk and no more than 40% during the subsequent 2 wk after weaning. Sorghum used as an alternative energy source for corn in diets fed to growing pigs decreases CP utilization by increasing manure N output, which might be partially offset by protease supplementation.

Determination and prediction of the digestible and metabolisable energy content of barley for growing pigs based on chemical composition

ABSTRACT This experiment was conducted to define the sources of variation determining the energy content of barley and to develop a practical method to predict the digestible energy (DE) and metabolisable energy (ME) content of individual barley samples. The 19 barley samples used in this study were diverse varieties grown in different regions. The feeding experiment used 57 barrows (initial body weight 31.5 ± 3.2 kg) and was conducted over two consecutive periods (n = 6 per treatment) using a completely randomised design. During each period, the pigs were placed in metabolic crates for a 5-d total collection of faeces and urine following a 10-d adaptation to the diets. Among the barley samples, on dry matter (DM) basis the levels of neutral detergent fibre, acid detergent fibre (ADF), crude protein and starch ranged from 16.1% to 38.9%, 3.9% to 9.6%, 10.1% to 16.8% and 43.5% to 57.9%, respectively. The mean determined DE and ME contents amounted to 14.7 and 14.4 MJ/kg DM and varied among the samples by 1.85 MJ (13.6%) and 1.78 MJ (13.3%), respectively. The ADF fraction accounted for 73% and 76% of the total variation in the DE and ME content, respectively. It revealed that for prediction of the DE and ME contents in barley, equations had the best fit when the analysed contents of ADF, neutral detergent fibre and gross energy were used for calculation (R2 = 0.92). On the basis of the developed equations, the DE and ME contents of barley of different origin can be predicted with an acceptable accuracy when used as feed for growing pigs.

Dietary supplementation with DHA-rich microalgae improves performance, serum composition, carcass trait, antioxidant status, and fatty acid profile of broilers

ABSTRACT This experiment was conducted with 126 as‐hatched male Arbor Acres chicks (1‐d‐old, weighing 45.3 ± 0.72 g) to determine the effects of microalgae [MA, containing 29% docosahexaenoic acid (DHA)] on performance, serum composition, carcass trait, antioxidant status, and fatty acid deposition of birds. The birds were allocated randomly to 1 of 3 treatments with 7 replicate pens per treatment (6 birds per pen). The dietary treatments included a control diet [corn‐soybean basal diet supplemented with 3% soybean oil (SO), CON], 1% MA diet (basal diet supplemented with 1% MA and 2% SO, 1MA), and 2% MA diet (basal diet supplemented with 2% MA and 1% SO, 2MA). All birds were raised in wire‐floored cages. The trial consists of a starter phase from d 1 to 21 and a grower phase from d 22 to 42. Compared with CON, birds supplemented with MA (1MA or 2MA) had greater (P < 0.05) average daily gain, liver percentage (liver weight/body weight), and serum glucose, as well as lower (P < 0.05) feed conversation ratio, abdominal fat percentage (abdominal fat weight/body weight), and total serum cholesterol. Moreover, due to the high concentration of DHA in MA, birds fed MA showed increased (P < 0.05) concentration of eicosapentaenoic acid, DHA, superoxide dismutase, and total antioxidant capacity, as well as decreased (P < 0.05) n‐6/n‐3 polyunsaturated fatty acid ratio, polyunsaturated fatty acid/saturated fatty acid ratio, and malondialdehyde in the breast and thigh muscle compared with CON. In conclusion, dietary supplementation with 1% or 2% DHA‐rich microalgae had positive effects on performance, serum composition, carcass trait, antioxidant status, and fatty acid deposition in birds.

Effects of Flavomycin, Bacillus licheniformis and Enramycin on Performance, Nutrient Digestibility, Gut Morphology and the Intestinal Microflora of Broilers

The effects of Flavomycin, Bacillus licheniformis and Enramycin on broiler performance, nutrient digestibility, gut morphology and the intestinal microflora were studied in a 42-d experiment. A total of 288, one-day-old, male, Arbor Acres broilers were randomly assigned to 1 of 4 dietary treatments with 12 pens per treatment and 6 birds per pen. The treatments were comprised of a control diet without supplementation, a diet supplemented with 5 ppm Flavomycin, a diet supplemented with the combination of 5 ppm Flavomycin and 1.35 × 109 CFU/kg Bacillus licheniformis, as well as a diet supplemented with 5 ppm Enramycin. The average daily gain (ADG) and feed conversion ratio (FCR) of birds fed the diet with Flavomycin combined with Bacillus licheniformis and the Enramycin diet were improved (P<0.05) compared with the control diet. The digestibility of dry matter, energy, and calcium for birds fed the combination of Flavomycin and Bacillus licheniformis and the Enramycin diet were also enhanced compared with the control diet. All additives improved the villus height and crypt depth in the duodenum, jejunum and ileum on d 21. In addition, reduced numbers of cecal E. coli (P<0.01) were found in birds fed all three supplemented diets on d 42. In conclusion, supplementation with Flavomycin and Bacillus licheniformis in combination or Enramycin would appear to be superior to supplementation with Flavomycin alone. All three supplemented diets were superior to the control.

Partial dehulling increases the energy content and nutrient digestibility of barley in growing pigs

Objective The hull attached to the barley kernel can be mechanically removed thus reducing the fiber content of the barley. This experiment was carried out to evaluate the effects of partial dehulling on the nutrient digestibility as well as the digestible energy (DE) and metabolizable energy (ME) content of barley in pigs. Methods Two hulled barley samples (high fiber barley [HF] and low fiber barley [LF]) with either high or low fiber contents were obtained from the Hubei and Jiangsu Provinces of China. A portion of the two barleys was mechanically dehulled (dehulled high fiber barley [DHF] and dehulled low fiber barley [DLF]). Thirty barrows (initial BW = 31.5±3.2 kg) were assigned to one of five diets in a randomized complete block design. The five diets consisted of 96.9% corn, HF, LF, DHF, or DLF supplemented with 3.1% minerals and vitamins. Each diet was fed to six barrows housed in individual metabolism crates for a 10-d acclimation period followed by a 5-day total but separate collection of feces and urine. Results The daily loss of gross energy (GE) in feces was lower (p<0.01) for pigs fed DHF than for those fed HF. The daily N intake and fecal N loss were lowest (p<0.01) for pigs fed the corn diet. The DE and ME as well as the apparent total tract digestibility (ATTD) of dry matter, GE, organic matter, neutral detergent fiber (NDF) and acid detergent fiber (ADF) of DHF or DLF were higher (p<0.01) than the values in HF and LF, respectively while the values except the ATTD of NDF and ADF in DLF were higher (p<0.01) than the values in DHF and were comparable to corn. Conclusion The DE and ME contents as well as the ATTD of nutrients in both DHF and DLF barley were improved compared with the HF and LF barley. Moreover, the nutritive value of DLF barley was comparable to the yellow-dent corn used in the study.
