Effects of yeast culture supplementation on growth performance, intestinal health, and immune response of nursery pigs.

A total of 216 weaning pigs were used in 2 experiments to determine the effects of dietary supplementation of yeast culture (YC) at different dose levels on the growth performance, nutrient digestibility, intestinal morphology, intestinal microflora, and immune response in weanling pigs and to determine whether YC can be a candidate to replace antibiotic growth promoters (AGP). In Exp. 1, 192 pigs (7.5 +/- 0.2 kg of BW) weaned at 28 d of age were randomly allotted to 6 treatments: 1) control (without AGP or YC); 2) AGP (chlortetracycline, 80 mg/kg); 3) 2.5 g/kg of YC (Diamond V XP Yeast Culture); 4) 5 g/kg of YC; 5) 10 g/kg of YC; and 6) 20 g/kg of YC. Each treatment had 8 replicated pens with 4 pigs per pen. Pigs were fed the experimental diets for 21 d. Average daily gain of pigs fed 5 g/kg of YC was greater (P < 0.05) than that of pigs in the control and other YC groups. However, there was no difference between the YC and AGP group. Pigs supplemented with 5 g/kg of YC, 10 g/kg of YC, and AGP had a greater (P < 0.01) ADFI than the control; however, G:F was not affected by treatment. Thus, 5 g/kg of YC supplementation level was chosen for Exp. 2. In Exp. 2, to elucidate the mode of action of YC, 24 nursery pigs (5.8 +/- 0.1 kg of BW; 21 d of age) were randomly allotted into 3 treatments for a 21-d trial. Treatments consisted of 1) control (without AGP or YC), 2) AGP, and 3) 5 g/kg of YC. Blood samples were collected weekly to measure CD4(+), CD8(+) percentage, and blood cytokine content. All pigs were harvested to determine treatment effects on gut microbiota, morphology, and immune function. Dietary supplementation of 5 g/kg of YC improved (P < 0.05) ADG of pigs compared with the control group, but performance of pigs fed YC was similar to those fed AGP. Pigs receiving 5 g/kg of YC had greater (P < 0.05) digestibility of DM, CP, GE, and jejunal villus height and villus height:crypt depth ratio (P < 0.05) compared with pigs fed the control diet. However, no differences in performance, digestibility, or gut morphology were observed between pigs fed YC and AGP. Gut interferon (IFN)-gamma concentrations were greater (P < 0.01) for pigs supplemented with YC compared with control pigs and pigs supplemented with AGP on d 21. However, plasma IFN-gamma concentrations were decreased (P < 0.01) in pigs supplemented with YC and AGP compared with control pigs on d 7, and CD4(+) was decreased (P < 0.01) in pigs supplemented with YC and AGP on d 14. Results indicate that dietary YC supplementation at 5 g/kg had a positive effect on growth performance of nursery pigs by improving jejunal villus height and villus height:crypt depth ratio and by modulating gut immune response. The comparable effect of 5 g/kg of YC supplementation and AGP on the growth performance of nursery pigs indicates that YC may be a good candidate as an antibiotic alternative.

Screening of Bacillus strains as potential probiotics and subsequent confirmation of the in vivo effectiveness of Bacillus subtilis MA139 in pigs.

A total of 124 samples were collected from the intestine of broiler chickens, piglet faeces, fermented foods, soils and Chinese herbs. More than 750 strains of aerobic, spore-forming bacteria were isolated from these samples. The inhibitory activity of these spore-forming strains against Escherichia coli K88, E. coli K99, Salmonella typhimurium and Staphylococcus aureus was assessed using a disc plate diffusion assay. The six bacilli with the largest inhibition zones against the four indicator bacteria were chosen and assessed for their resistance to unfavorable conditions within simulated gut environments. The strain Bacillus subtilis MA139 showed full resistance to pH 2, 0.3% bile salts and exhibited the highest antimicrobial activity. Based on these results, B. subtilis MA139 was selected as a potential probiotic and fed to piglets at concentrations of 2.2 × 105, 2.2 × 106 or 2.2 × 107 CFU/g of feed during a 28-day feeding trial. A negative control consisting of the basal diet with no additives and a positive control consisting of the basal diet supplemented with 16 g/ton flavomycin were also included. Ninety piglets between 35 and 40 days old were used in the in vivo animal trials. B. subtilis MA139 enhanced daily gain (P = 0.10) and feed conversion (P = 0.03) compared with the negative control. The performance of pigs fed B. subtilis MA139 supplemented diets did not differ from that of pigs fed the antibiotic diet. There was a significant increase in Lactobacilli cell counts (P = 0.02) and a numerical decrease in E. coli counts (P = 0.05) in the faecal samples of pigs fed B. subtilis MA139 with 2.2 × 105 CFU/g of feed. The overall results of this study show that the use of initial co-culture with indicator pathogens, a disc plate diffusion assay and simulated gut environment tolerance tests is one of effective methods of screening Bacillus for probiotic use and that B. subtilis MA139 is a promising alternative to antibiotics for use as a feed additive in piglet diets.

Essential oil and aromatic plants as feed additives in non-ruminant nutrition: a review.

This paper summarizes the current knowledge regarding the possible modes of action and nutritional factors involved in the use of essential oils (EOs) for swine and poultry. EOs have recently attracted increased interest as feed additives to be fed to swine and poultry, possibly replacing the use of antibiotic growth promoters which have been prohibited in the European Union since 2006. In general, EOs enhance the production of digestive secretions and nutrient absorption, reduce pathogenic stress in the gut, exert antioxidant properties and reinforce the animal’s immune status, which help to explain the enhanced performance observed in swine and poultry. However, the mechanisms involved in causing this growth promotion are far from being elucidated, since data on the complex gut ecosystem, gut function, in vivo oxidative status and immune system are still lacking. In addition, limited information is available regarding the interaction between EOs and feed ingredients or other feed additives (especially pro- or prebiotics and organic acids). This knowledge may help feed formulators to better utilize EOs when they formulate diets for poultry and swine.

Lignans from the fruits of Forsythia suspensa.

Bioactivity-guided fractionation of the methanol extract of the fruits of Forsythia suspensa Vahl has led to the isolation of two new monoepoxylignans, forsythialan A (1) and B (2), together with a known tetrahydrofurofuran lignan, phillyrin (3). The structures of compounds 1 and 2 were elucidated by spectroscopic methods. The antioxidant activities of these lignans have been assessed by evaluating their protective effects against peroxynitrite-induced oxidative stress. The compounds 1 and 2 showed protective effects against renal epithelial cell injury by 3-morpholinosydnonimine (SIN-1), a peroxynitrite generator. The relatively stronger antioxidant activities of compounds 1 and 2 may be associated with the presence of aromatic hydroxy function.

Protective effects of Forsythia suspensa extract against oxidative stress induced by diquat in rats

Forsythia suspensa extract has been proved as a potential antioxidant in the recent years. The present study was undertaken to obtain the optimal antioxidant fraction in vitro and examine its antioxidative potential against diquat-induced oxidative stress in male Sprague Dawley rats in vivo. In vitro, 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging experiment indicated that the CH2Cl2 fraction of F. suspensa (FSC) exerted the strongest scavenging activities; forsythoside A, forythialan A and phillygenin from it might be the major antioxidant constituents. In vivo, pretreatment of rats with different doses of FSC (25, 50 and 100 mg/kg bw) and vitamin C (100 mg/kg bw, positive control) for 15 days significantly lowered the tumor necrosis factor-alpha (TNF-alpha), interleukin-1beta (IL-1beta), and interleukin-6 (IL-6) in plasma compared to the negative control group. Also, FSC significantly increased the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and the levels of glutathione (GSH) in plasma, liver and kidney whereas it decreased the levels of malondialdehyde (MDA) in plasma and kidney. Moreover, the protective effect of FSC (100 mg/kg bw) was better than vitamin C. These results revealed that FSC exerted a protective effect against diquat-induced oxidative stress and is worthy of becoming a potential dietary antioxidant.

Preventive effect of Coptis chinensis and berberine on intestinal injury in rats challenged with lipopolysaccharides

Coptis chinensis has been used in traditional Chinese medicine to treat inflammatory symptoms. Berberine is the main alkaloid compound of C. chinensis. This study utilized a typical lipopolysaccharide (LPS) injured model to investigate the effects of C. chinensis aqueous extract (CCAE) and berberine (major active ingredient in CCAE) in the gut-derived sepsis. In rats, pretreatment with different doses of berberine (30 or 120 mg/kg bw, i.g.; BBR30 or BBR120) or CCAE (containing 9.9% berberine; 300 mg/kg bw, i.g.; CCAE300) prior to the administration of LPS (20 mg/kg bw, i.p.) significantly suppressed the increased tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and nitrite oxide (NO) in plasma as well as the activation of toll-like receptor 4 (TLR4) and nuclear factor-κB (NF-κB) in ileum. In addition, CCAE300 and BBR30 markedly elevated the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px); significantly prevented the increased malondialdehyde (MDA), NO and villi injury in ileum compared with the negative control. Collectively, CCAE300 and BBR30 reduced the LPS-induced intestinal damage by elevating the activities of SOD and GSH-Px and by suppressing the activation of TLR4 and NF-κB in ileum. These results indicate that CCAE and berberine are promising agents for preventing sepsis and its complications.

Chito-oligosaccharide reduces diarrhea incidence and attenuates the immune response of weaned pigs challenged with Escherichia coli K88.

Seventy-two barrows (Landrace × Large White, initial BW of 4.9 ± 0.3 kg and 17 ± 3 d old) were used to determine if dietary chito-oligosaccharides can replace antibiotics as a means to reduce signs associated with infection in weaned pigs challenged with Escherichia coli. Pigs were assigned to 1 of 4 treatments in a randomized complete block design using 6 pens per treatment with 3 pigs per pen. The treatments consisted of pigs fed the unsupplemented corn-soybean meal diet challenged or unchallenged with E. coli K88 and pigs fed the same diet supplemented with 160 mg of chito-oligosaccharides or 100 mg of cyadox/kg and challenged with E. coli K88. On d 7, 1 group of pigs fed the unsupplemented diet, as well as all pigs fed diets containing chito-oligosaccharides or cyadox, were orally dosed with 30 mL of an alkaline broth containing E. coli K88. Another group of pigs fed the unsupplemented diet was orally dosed with 30 mL of sterilized alkaline broth. Fecal consistency was visually assessed each morning from d 7 to 14. Blood samples were collected at 0, 24, 48, and 168 h postinfection. On d 14 postchallenge, all pigs were killed to evaluate intestinal morphology and determine E. coli concentrations in the intestine. During the postchallenge period (wk 2), unsupplemented pigs challenged with E. coli had decreased (P < 0.05) BW gain, feed intake, fecal consistency, villus height, villus height:crypt depth ratio, and plasma IGF-1, and increased (P < 0.05) diarrhea incidence, E. coli counts in the intestine, plasma interleukin-1β, plasma IL-10, and IGA-positive cells in the jejunal and ileal lamina propria, compared with unchallenged pigs. Supplementation with cyadox largely mitigated these effects. Although chito-oligosaccharide reduced the incidence of diarrhea, the growth performance of E. coli-challenged pigs supplemented with chito-oligosaccharide was not better than that of unsupplemented pigs challenged with E. coli. Therefore, chito-oligosaccharide, at the amount used in this experiment, does not seem to be an effective substitute for antibiotics as a growth promoter for newly weaned pigs challenged with E. coli.

Effects of Forsythia suspensa Extract on Growth Performance, Nutrient Digestibility, and Antioxidant Activities in Broiler Chickens Under High Ambient Temperature

A 42-d trial was conducted with 252 broiler chickens (d 1 of age, 38.8 +/- 1.3 g of BW) to determine the effects of Forsythia suspensa extract on growth performance, nutrient digestibility, and antioxidant activities under high ambient temperature (32 +/- 1 degrees C). The feeding program consisted of a starter diet from d 1 to 21 of age and a finisher diet from d 22 to 42 of age. Dietary treatments included a negative control group (NC) fed a cornsoybean meal based diet without vitamin C or Forsythia suspensa extract, a positive control group fed a diet with 200 mg of vitamin C/kg, and a test group (FS) fed with 100 mg of Forsythia suspensa extract/kg. There were 14 cages per treatment and 6 birds per cage during the study. Birds had free access to diets and water during the entire period. Body weight and feed intake were measured at d 21 and 42. Blood and tissue samples were collected at d 21 and 42 for assay of antioxidant indices. Growth performance did not differ among treatment groups during the starter period. In the finisher phase, birds in FS had greater (P < 0.05) average daily gain, feed conversion, and apparent digestibility of energy, CP, calcium, and phosphorus than birds in NC. Furthermore, birds in FS had greater (P < 0.05) total antioxidant capacity and superoxide dismutase activity and lower (P < 0.05) malondialdehyde activity in the serum than birds in NC. The FS birds had greater (P < 0.05) muscle superoxide dismutase activity and lower (P < 0.05) malondialdehyde than NC birds. During the entire period, hepatic superoxide dismutase activity of FS birds was greater (P < 0.05) than that of NC birds. Dietary supplementation with Forsythia suspensa extract can enhance nutrient digestibility and growth performance possibly by reducing oxidative stress of broiler chickens under high ambient temperatures.

Effects of Adding Essential Oil to the Diet of Weaned Pigs on Performance, Nutrient Utilization, Immune Response and Intestinal Health

The objective of this study was to evaluate the effects of adding essential oils to the diet of weaned pigs on performance, nutrient utilization, immune response and intestinal health. A total of 96 weaning pigs (8.37±1.58 kg) were allotted to one of three dietary treatments. The treatments consisted of an unsupplemented basal diet (negative control, NC) or similar diets supplemented with 0.01% of an essential oil product which contained 18% thymol and cinnamaldehyde (EOD) as well as a diet supplemented with 0.19% of an antibiotic mixture which provided 150 ppm chlortetracycline, 80 ppm colistin sulfate and 50 ppm kitasamycin (positive control, PC). Each treatment was provided to eight pens of pigs with four pigs per pen. Over the entire 35 d experiment, ADG and fecal score were improved (p<0.05) for pigs fed the PC and EOD compared with the NC. Dry matter and crude protein digestibility as well as lymphocyte proliferation for pigs fed the PC and EOD diets were increased significantly compared with NC (p<0.05). IGF-I levels in plasma were significantly increased (p<0.05) in pigs fed the PC diet compared with pigs fed the NC diet. Interleukin-6 concentration was lower (p<0.05) and the tumor necrosis factor-α level was higher (p<0.05) in the plasma of pigs fed the EOD diet than the NC diet. Plasma total antioxidant capacity level increased (p<0.05) in pigs fed the EOD diet compared with pigs fed the NC. Villus height to crypt depth ratio in the jejunum was greater (p<0.05) in pigs fed the PC and EOD diets than the NC. The numbers of E. coli in the cecum, colon and rectum were reduced (p<0.05) in pigs fed the PC and EOD diets compared with the control. In the colon, the ratio of Lactobacilli to E. coli was increased (p<0.05) in pigs fed the EOD diet compared with NC diet. Total aerobe numbers in the rectum were decreased (p<0.05) in pigs fed the PC and EOD diets compared with the control. Collectively, these results indicate that blends of essential oils could be a candidate for use as an alternative to traditional antibiotics in weaning pig diets.

Effects of dietary Astragalus polysaccharide on growth performance and immune function in weaned pigs

An experiment was conducted to evaluate the effects of dietary supplementation of a polysaccharide isolated from Astragalus membranaceus (APS) on performance and immune responses in weaned pigs. A total of 144 crossbred pigs weaned at 26 to 30 days of age with an average initial live weight (LW) of 7·64 (s.d. 0·290) kg were randomly allotted to six diets supplemented with APS at 0, 100, 250, 500, 750, and 1000 mg/kg. There were six replicates (three barrow pens and three gilt pens) per diet treatment with four pigs per pen. Pigs were given food ad libitum for 21 days and the LW and food intake were measured on days 14 and 21. Pigs were intramuscularly injected with 1 mg/kg LW ovalbumin (OVA) on day 14 to evaluate humoral immune response. Blood samples were collected on day 21 to measure leukocyte differential counts, percentage of blood CD4 þ and CD8 þ lymphocyte subsets, lymphocyte proliferation response to Concanavalin A, serum concentration of immunoglobulin G (Ig G), interleukin-2 (IL-2), interleukin-4 (IL-4), interleukin-10 (IL-10), interferon-g (IFN-g) and specific OVA antibody. The results showed that the average daily gain, the numbers of WBC and lymphocytes, the proportion of CD4 þ lymphocyte subset, and the contents of IL-2 and IFN-g increased ( P , 0·05) as pigs were fed increased supplemental level of APS during the 21 d period. However, the contents of specific OVA antibody, Ig G, IL-4, and IL-10 were not affected ( P . 0·05) by dietary levels of APS. The broken line analysis and quadratic regression analysis indicate that the optimal APS supplemental level would be between 381 mg/kg and 568 mg/kg for the maximal ADG and from 324 to 563 mg/kg for immune responses. Collectively, this study suggests that dietary APS can be used as a potential immuno-modulating agent by affecting cellular immunity of weaned pigs.
