Technological Potential of Lactobacillus Strains Isolated from Fermented Green Olives: In Vitro Studies with Emphasis on Oleuropein-Degrading Capability

Technological properties of two strains of Lactobacillus plantarum (B3 and B11) and one of Lactobacillus pentosus (B4), previously isolated from natural fermented green olives, have been studied in vitro. Acidifying ability, salt, temperature, and pH tolerances of all strains were found in the range reported for similar strains produced in Italy and optimal growth conditions were found to be 6.0–8.0 pH, 15–30°C temperature, and less than 6% NaCl. Moreover, all strains showed very good tolerance to common olive phenol content (0.3% total phenol) and high oleuropein-degrading capability. It was found that medium composition affected the bacterial oleuropein degradation. B11 strain grown in a nutrient-rich medium showed a lower oleuropein-degrading action than when it was cultivated in nutrient-poor medium. Furthermore, enzymatic activity assays revealed that oleuropein depletion did not correspond to an increase of hydroxytyrosol, evidencing that bacterial strains could efficiently degrade oleuropein via a mechanism different from hydrolysis.

Biodiversity of Lactobacillus plantarum from traditional Italian wines

In this study, 23 samples of traditional wines produced in Southern Italy were subjected to microbiological analyses with the aim to identify and biotype the predominant species of lactic acid bacilli. For this purpose, a multiple approach, consisting in the application of both phenotypic (API 50CHL test) and biomolecular methods (polymerase chain reaction-denaturing gradient gel electrophoresis and 16S rRNA gene sequencing) was used. The results showed that Lactobacillus plantarum was the predominant species, whereas Lb. brevis was detected in lower amount. In detail, out of 80 isolates 58 were ascribable to Lb. plantarum and 22 to Lb. brevis. Randomly amplified polymorphic DNA-polymerase chain reaction was used to highlight intraspecific variability among Lb. plantarum strains. Interestingly, the cluster analysis evidenced a relationship between different biotypes of Lb. plantarum and their origin, in terms of wine variety. Data acquired in this work show the possibility to obtain several malolactic fermentation starter cultures, composed by different Lb. plantarum biotypes, for their proper use in winemaking processes which are distinctive for each wine.

Sub-optimal pH Preadaptation Improves the Survival of Lactobacillus plantarum Strains and the Malic Acid Consumption in Wine-Like Medium

Forty-two oenological strains of Lb. plantarum were assessed for their response to ethanol and pH values generally encountered in wines. Strains showed a higher variability in the survival when exposed to low pH (3.5 or 3.0) than when exposed to ethanol (10 or 14%). The study allowed to individuate the highest ethanol concentration (8%) and the lowest pH value (4.0) for the growth of strains, even if the maximum specific growth rate (μmax) resulted significantly reduced by these conditions. Two strains (GT1 and LT11) preadapted to 2% ethanol and cultured up to 14% of ethanol showed a higher growth than those non-preadapted when they were cultivated at 8% of ethanol. The evaluation of the same strains preadapted to low pH values (5.0 and 4.0) and then grown at pH 3.5 or 3.0 showed only for GT1 a sensitive μmax increment when it was cultivated in MRS at pH 3 after a preadaptation to pH 5.0. The survival of GT1 and LT11 was evaluated in Ringers solution at 14% ethanol after a long-term adaptation in MRS with 2% ethanol or in MRS with 2% ethanol acidified at pH 5.0 (both conditions, BC). Analogously, the survival was evaluated at pH 3.5 after a long-term adaptation in MRS at pH 5.0 or in MRS BC. The impact of the physiologic state (exponential phase vs stationary phase) on the survival was also evaluated. Preadapted cells showed the same behavior of non-preadapted cells only when cultures were recovered in the stationary phase. Mathematical functions were individuated for the description of the survival of GT1 and LT11 in MRS at 14% ethanol or at pH 3.5. Finally, a synthetic wine (SW) was used to assess the behavior of Lb. plantarum GT1 and LT11 preadapted in MRS at 2% ethanol or at pH 5.0 or in BC. Only GT1 preadapted to pH 5.0 and collected in the stationary phase showed constant values of microbial counts after incubation for 15 days at 20°C. In addition, after 15 days the L-malic acid resulted completely degraded and the pH value increased of about 0.3 units.

Yeast Autolysis in Sparkling Wine Aging: Use of Killer and Sensitive Saccharomyces cerevisiae Strains in Co-Culture

BACKGROUND Sparkling wines produced by traditional method owe their characteristics to secondary fermentation and maturation that occur during a slow ageing in bottles. Yeast autolysis plays an important role during the sparkling wine aging. Using a combination of killer and sensitive yeasts is possible to accelerate yeast autolysis and reduce maturing time. METHODS killer and sensitive Saccharomyces cerevisiae strains, separately and in co-cultures, were inoculated in base wine and bottled on pilot-plant scale. Commercial Saccaromyces bayanus strain was also investigated. Protein free amino acid and polysaccharides contents and sensory analysis were determined on the wine samples at 3, 6 and 9 months of aging. Yeast autolysis that occurs during the production of sparkling wines, obtained with co-cultures of killer and sensitive strains, has influenced free amino acids, total protein and polysaccharides content after 3 months aging time: sparkling wines, produced without the use of these yeasts, have reached the same results only after 9 months aging time. RESULTS These results demonstrate that killer and sensitive yeasts in co-culture can accelerate the onset of autolysis in enological conditions, and has a positive effect on the quality of the aroma and flavor of sparkling wine. CONCLUSION This paper offers an interesting biotechnological method to reduce production time of sparkling wine with economical benefits for the producers. We revised all patents relating to sparkling wine considering only those of interest for our study.

Effective assay for olive vinegar production from olive oil mill wastewaters

In this work, an effective and simple approach for vinegar production from olive oil press-mill wastewaters (OMW) is presented. Effects of sterilization and yeast presence on the acetic acid production were investigated. Sugar addition and inoculum of selected yeast starter have been crucial for a satisfactory acidification. In the obtained olive vinegar, the pH and total acidity were 2.92 and 5.6%, respectively. A considerable high level of ash (2%) and total phenols (3600mg/L as GAE) characterized olive vinegar, in comparison with samples of apple, wine and balsamic commercial vinegars. Moreover, a high presence of hydroxytyrosol (1019mg/L) was obtained. This abundant presence of antioxidants makes olive vinegar a promising nutraceutical and environmentally-friendly product, based upon a waste material such as OMW.

Variability in chemical and microbiological profiles of long-ripened Caciocavallo cheeses

Five protected designation of origin (PDO) Caciocavallo Silano and 6 non-PDO Caciocavallo cheeses, ripened for 6mo and collected in the 5 Italian regions of the PDO area (Apulia, Basilicata, Calabria, Campania, and Molise, Italy), were studied to assess their physico-chemical (pH, acidity, moisture, fat, ash, protein, and free amino acid composition) and microbiological profiles. Analyses evidenced a certain fluctuation of previous parameters among samples regardless of the kind of cheese evaluated (PDO and non-PDO). The PCR-denaturing gradient gel electrophoresis analysis performed on the DNA directly extracted from cheeses gave different results, but a low number of bands was always observed. Only one band, corresponding to the species Streptococcus thermophilus, was detectable in 1 PDO and in 2 non-PDO cheese samples, whose free amino acid content was the lowest. Analyses were repeated on experimental Caciocavallo cheeses. Specifically, 2 productions were made, one mimicking the industrial technology (pasteurized milk and selected starter culture) and one the artisanal technology (raw milk and natural whey starter). Results obtained on experimental cheeses at 6mo of ripening showed that industrial samples had lower amounts of total free amino acids then the artisanal ones (1,188.2 vs. 7,523.67mg/100 g of dry matter). Moreover, the PCR-denaturing gradient gel electrophoresis analysis evidenced the sole presence of S. thermophilus in the case of the industrial technology. These data sustain the hypothesis that, out of 11 cheeses analyzed previously, 1 PDO Caciocavallo Silano and 2 non-PDO Caciocavallo cheeses were obtained with the industrial technology. These results could be of help in the discrimination of PDO products, taking into account that the PDO production regulation does not allow the milk pasteurization, nor the use of selected starters.

Sequential inoculum of Hanseniaspora guilliermondii and Saccharomyces cerevisiae for winemaking Campanino on an industrial scale

In this study, the effect of sequential inoculation with non-Saccharomyces (Hanseniaspora guilliermondii) and Saccharomyces cerevisiae yeast on the distinctive characteristics of the Campanino white wine was investigated. For this purpose, three independent winemaking experiments were carried out on an industrial scale (batches A, B and C). In detail, the first one was carried out using the sequential inoculation technique while the other two, using a S. cerevisiae single-strain starter or no inoculation representing the control batches. Microbiological and chemical parameters and sensorial profiles of the wines were defined. Interestingly, the results showed that when sequential cultures (H. guilliermondii in a sequential mixture with S. cerevisiae) were used, a better wine aroma and quality was observed. More specifically, the wine obtained by sequential inoculation showed lower acetic acid values and enhanced volatile profiles than the wine from the control batches. Finally, sensorial analysis confirmed that the sequential cultures led to an improvement in wine flavour. Therefore, results suggest that the sequential inoculation using non-Saccharomyces and Saccharomyces yeast represents a biotechnological practice that can improve the quality features of traditional white wine. It has been shown for the first time that on an industrial scale H. guilliermondii could be used in sequential inoculum with S. cerevisiae in making white Campanino wine.Graphical abstract

EFFECT OF DIFFERENT STORAGE CONDITIONS ON THE SHELF LIFE OF NATURAL GREEN TABLE OLIVES

The aim of this research is to study the effects of different storage conditions on Spanish (alkaline debittering) and natural (directly brined) green olives. Laboratory-processed olives were stored in 6% brine or in a vacuum bag without brine, at 6 or 20°C. After 18 months, natural olives showed higher microbial and olive oil stability than NaOH-treated olives. The lower pH (<4.80) and higher total phenol content (0.2 g/100 g wet pulp) influenced positively the long shelf life of natural olives. The packaging in 6% NaCl brine and in a vacuum bag stored at 20°C gave better performance, while growth of psychrophilic spoilage bacteria occurred at cold temperature.

Detection of Antilisterial Activity of 3-Phenyllactic Acid Using Listeria innocua as a Model

The 3-Phenyllactic acid (PLA) produced by various lactic acid bacteria (LAB) possesses a broad spectrum of antimicrobial activity. In this study, the effect of PLA against Listeria innocua was studied with the aim to obtain additional information about its mechanism of action. The effect of pH on the antilisterial activity of PLA was investigated and a pH-dependent behavior, typical of weak acid, was detected. The antilisterial effect of PLA was firstly compared to that produced by lactic acid (LA) and than to that expressed by phenolic acids (gallic, caffeic, and ferulic acids) evaluating minimum inhibitory concentration (MIC), MBC, and survival kinetic parameters. PLA showed MIC values and death kinetic parameters significantly different from those exhibited by LA and by tested phenolic acids. In particular, the MIC value observed for PLA vs L. innocua resulted lower than that of the other preservative compounds studied herein, and consistent with the quantity generally produced by LAB. Moreover, the effect of PLA and phenolic acids on bacterial surface charge and loss of cellular content resulted different. The overall results highlighted strong differences in the antilisterial mechanism of action among PLA and other compounds such as LA and phenols. Specifically, it is possible to hypothesize that the antilisterial mechanism of action due to PLA is associated with the affinity to cell surface, which contributes to the cellular damage.

Our model develops the Data Specification on Protected Sites , part of the INSPIRE Annex I. Hence its orientation towards georeferenced heritage data. The data model has been developed by an interdisciplinary group made up of specialists in the field of Geomatics and Cultural Heritage. It is our aim to achieve a generic, extendable and interoperable schema. It should be generic enough to embrace all kind of protected heritage data, from an ancient pilgrim’s way to the last artifact found in an archaeological excavation, extendable to allow any kind of data producer to adapt the model to the nature of their own information and interoperable to combine spatial data sets from different sources through network services, via Internet. The achievement of these three characteristics features international norms and standards referred to our kind of data. This implies adaptation to INSPIRE as well as to several ISO norms: ISO 19100 series regarding geographical information, ISO 21127 (CIDOC-CRM Model) for heritage thematic data and ISO 15836 (Dublin Core) for document resources. The data model comprehends two main dimensions: cultural entities in a strict sense, and the legislative figures created to protect them. This allows for the representation of cultural objects (i.e. historical buildings or archaeological sites) and their link to their legislative protection, keeping them as separate realities. In order to describe the data model, an international common language has been used: UML (Unified Modeling Language), a standard itself. Su transformacion en glucosa se considera la etapa mas importante en la produccion de biocombustibles a partir de biomasa lignocelulosica. Para esta transformacion es necesaria la accion sinergica de tres tipos de enzimas que hidrolizan los enlaces s-1,4 de la celulosa: (i) las endoglucanasas, que actuan al azar sobre enlaces internos, (ii) las celobiohidrolasas, que operan progresivamente por los extremos reductores y no reductores de la cadena, y (iii) las s-glucosidasas (BGL), que hidrolizan celobiosa y los celooligosacaridos mas pequenos hasta glucosa. Los hongos filamentosos son la principal fuente de celulasas, siendo los crudos del genero Trichoderma los mas estudiados y comercializados historicamente. Sin embargo, estos hongos secretan niveles insuficientes de BGL para una conversion efectiva de la celulosa, por lo que con frecuencia requieren ser suplementados con preparaciones ricas en esta enzima procedentes de otros hongos. Recientemente, se han descrito cepas de Penicillium con gran capacidad para secretar altos niveles de BGL al medio extracelular. Ademas de su papel en la hidrolisis de la celulosa las BGL tambien pueden emplearse para sintetizar compuestos de interes industrial mediante reacciones de transglicosilacion. En este trabajo se ha estudiado una nueva cepa fungica productora de celulasas, identificada como Talaromyces amestolkiae, en base a un analisis molecular y morfologico. Se han purificado a homogeneidad electroforetica tres BGL (BGL-1, BGL-2 y BGL-3) secretadas por el hongo y se han caracterizado bioquimicamente. BGL-1 y BGL-2 son proteinas monomericas, mientras que BGL-3 es un dimero funcional. Los valores de actividad maxima de estas enzimas se obtuvieron a pH 4,0 y entre 50-60 oC, siendo estables en un rango de pH de 4-7 y a 50 oC. Las tres mostraron distinto comportamiento en funcion del sustrato (pNPG o celobiosa), ensayandose tambien el efecto de determinados compuestos quimicos e inhibidores en su actividad. Las tres pueden hidrolizar celooligosacaridos de diferente longitud, disminuyendo la eficacia de hidrolisis con el aumento de la polimerizacion, y no son activas frente a polisacaridos. Ademas, mostraron actividad de transglicosilacion, formando alquilglicosidos y celooligosacaridos de mayor longitud que los usados como sustratos. Las tres BGL se identificaron mediante huella peptidica y en base la alta homologia con otras BGL fungicas relacionadas filogeneticamente, se disenaron cebadores especificos que permitieron la secuenciacion de los genes que codifican cada una de ellas. El analisis de las secuencias aminoacidicas mostro que BGL-1 es miembro de la familia 1 de las glicosil hidrolasas, mientras que BGL-2 y BGL-3 pertenecen a la familia 3. Teniendo en cuenta que existen pocas BGL fungicas cristalizadas, se construyeron modelos moleculares de estas ultimas en base a las estructuras con las que presentaron mayor identidad. El crudo enzimatico de T. amestolkiae, rico en BGL, se uso de forma individual y como suplemento de otros cocteles comerciales en experimentos de sacarificacion a partir de slurry acido de paja de trigo. Los resultados obtenidos indican que el hongo secreta, ademas de celulasas, otras enzimas complementarias que potencian la liberacion de azucares fermentables. Se analizo el secretoma de T. amestolkiae usando Avicel o slurry acido de paja de trigo como fuente de carbono. Las celulasas fueron las enzimas mas abundantes en ambas condiciones, sin embargo, en el crudo obtenido a partir de los cultivos en slurry aumento la proporcion de BGL y otras enzimas distintas a las celulasas, indicando que para la degradacion de un sustrato complejo es necesaria mayor diversidad enzimatica. El olivo (Olea europaea L.) es el cultivo lenoso no tropical que ocupa mayor superficie en todo el mundo, con el 95% de la produccion mundial localizada en la cuenca mediterranea. Espana es el pais con mayor superficie de olivar del mundo con 2,6 MHa y aproximadamente el 39% de la produccion mundial. La Verticilosis, causada por el hongo de suelo Verticillium dahliae Kleb, constituye la principal amenaza para el olivar. Esta enfermedad afecta al olivo en todos los paises de tradicion olivarera y causa importantes perdidas de rendimiento y muerte de los arboles (Jimenez-Diaz y col., 2012). Este patogeno coloniza el sistema vascular de la planta, bloqueando el flujo del agua y finalmente induciendo estres hidrico (Van Alfen, 1989).Espana es el mayor productor de citricos de la Union Europea y el quinto a nivel mundial, con una produccion anual superior a cinco millones de toneladas (MAGRAMA, 2016). Este sector genera una gran cantidad de subproductos, con un elevado aporte energetico y alto contenido en pectinas y azucares, por lo que podrian usarse como materias primas en dietas para rumiantes. Sin embargo, los subproductos de citricos son muy diversos, por lo que su composicion quimica y valor nutritivo tambien puede ser variable (FEDNA, 2010; Feedipedia, 2016). Estos efectos se producen fundamentalmente a traves de su relacion, directa o indirecta, con la arquitectura del racimo, el comportamiento reproductivo y el grado o modo de exposicion de las bayas al ambiente. Respecto a los dos primeros, en un estudio previo, determinamos que tanto la longitud del raquis y sus primeras ramas como el numero de bayas por racimo son los caracteres con mayor incidencia en la compacidad entre los analizados. Ahora bien, el estudio de la base genetica responsable del numero de bayas requiere de su diseccion previa, dado que es una variable secundaria, resultado del numero de flores por inflorescencia y de la tasa de cuajado o conversion de flor en fruto. En este trabajo se presenta un estudio preliminar en el que se han caracterizado fenotipicamente 104 variedades de vid para estudiar su comportamiento reproductivo. Asi, sobre una base amplia de diversidad varietal, se han estimado variables como el numero de flores, la tasa de cuajado, corrimiento, millerandage, etc., evaluando su posible relacion con la compacidad del racimo. Este trabajo, una vez completado durante varios anos, permitira conocer la diversidad existente para el comportamiento reproductivo en la vid, asi como, de forma mas concreta, abordar la caracterizacion de los procesos geneticos responsables de las diferencias fenotipicas detectadas para el comportamiento reproductivo y sus elementos individuales y para la compacidad del racimo, identificando los posibles genes involucrados en dichos procesos.Trabajo presentado en la 11th International Conference on Grapevine Breeding and Genetics, celebrada en Pekin del 29 de julio al 2 de agosto de 2014.The Polarimetric and Helioseismic Imager (PHI) is the first deep-space solar spectropolarimeter, on-board the Solar Orbiter (SO) space mission. It faces: stringent requirements on science data accuracy, a dynamic environment, and severe limitations on telemetry volume. SO/PHI overcomes these restrictions through on-board instrument calibration and science data reduction, using dedicated firmware in FPGAs. This contribution analyses the accuracy of a data processing pipeline by comparing the results obtained with SO/PHI hardware to a reference from a ground computer. La mayor parte de los yacimientos se concentran en las regiones de Amêndoa-Mação y Buçaco, aunque también existen citas de equinodermos en Arouca, Dornes, Moncorvo y Valongo. Por último se comentan futuras líneas de trabajo que ayudarán a comprender mejor estas faunas, perfeccionar y completar el conocimiento del registro fósil del grupo en el Ordovícico centroibérico.
