Qualidade microbiológica de leite cru refrigerado e isolamento de bactérias psicrotróficas proteolíticas

Storing refrigerated raw milk in the source of production reduces economic losses which occur due to the acidifying activity of mesophilic bacteria. Nevertheless, this process selects psychrotrophic bacteria that cause technological and economic problems to the dairy industry. Samples of individual and collective refrigerated tanks and samples from an industrial silo in a dairy industry were collected and analyzed to evaluate the microbiological quality of raw milk. Furthermore, proteolytic psychrotrophic bacteria were isolated from refrigerated raw milk and characterized according to the Gram reaction and glucose fermentation. The refrigerated raw milk from the industrial silo was not compatible with legal microbiological standards, and its microbial count was significantly higher than milk from individual and collective tanks. A significant difference of contamination by mesophiles, proteolytic and non-proteolytic psychrotrophs, and Pseudomonas was observed in samples collected from refrigeration tanks compared to samples collected from the industrial silo. The Gram-negative microbiota was isolated more often, especially the glucose non-fermentative Gram-negative bacteria.

Milk-deteriorating exoenzymes from Pseudomonas fluorescens 041 isolated from refrigerated raw milk.

The practice of refrigerating raw milk at the farm has provided a selective advantage for psychrotrophic bacteria that produce heat-stable proteases and lipases causing severe quality problems to the dairy industry. In this work, a protease (AprX) and a lipase (LipM) produced by Pseudomonas fluorescens 041, a highly proteolytic and lipolytic strain isolated from raw milk obtained from a Brazilian farm, have been purified and characterized. Both enzymes were purified as recombinant proteins from Escherichia coli . The AprX metalloprotease exhibited activity in a broad temperature range, including refrigeration, with a maximum activity at 37 °C. It was active in a pH range of 4.0 to 9.0. This protease had maximum activity with the substrates casein and gelatin in the presence of Ca +2 . The LipM lipase had a maximum activity at 25 °C and a broad pH optimum ranging from 7.0 to 10. It exhibited the highest activity, in the presence of Ca +2 , on substrates with long-chain fatty acid residues. These results confirm the spoilage potential of strain 041 in milk due to, at least in part, these two enzymes. The work highlights the importance of studies of this kind with strains isolated in Brazil, which has a recent history on the implementation of the cold chain at the dairy farm.

Lack of AHL-based quorum sensing in Pseudomonas fluorescens isolated from milk

Numerous bacteria coordinate gene expression in response to small signalling molecules in many cases known as acylhomoserine lactones (AHLs), which accumulate as a function of cell density in a process known as quorum sensing. This work aimed to determine if phenotypes that are important to define microbial activity in foods such as biofilm formation, swarming motility and proteolytic activity of two Pseudomonas fluorescens strains, isolated from refrigerated raw milk, are influenced by AHL molecules. The tested P. fluorescens strains did not produce AHL molecules in none of the evaluated media. We found that biofilm formation was dependent on the culture media, but it was not influenced by AHLs. Our results indicate that biofilm formation, swarming motility and proteolytic activity of the tested P. fluorescens strains are not regulated by acyl-homoserine lactones. It is likely that AHL-dependent quorum sensing system is absent from these strains.

Fruit salad as a new vehicle for probiotic bacteria

This work aimed to study the use of fruit salads as carriers for Lactobacillus rhamnosus HN001. We evaluated the viability of this probiotic in fruit salads and the phsyico-chemical, microbiological and sensory properties of this food. Scanning electron microscopy (SEM) was used to verify microorganism adhesion on the fruit tissues. The viability of L. rhamnosus in fruit salads was 8.49 log CFU.g-1 after 120 hours. SEM images showed that fruit tissue provided protection for probiotic. Adhesion sites were observed in higher quantity in banana, apple and guava. The addition of L. rhamnosus did not alter texture of fruits (p > 0.05). Fruit salads containing probiotic had different values of pH and acidity compared to the control (p 0.05). Fruit salads containing L. rhamnosus showed counts of psychotrophic microorganisms of at least 2.0 log CFU.g-1 lower than control salad after 120 h of refrigerated storage. The fruit salad was well accepted by consumers. Therefore, this product can be used as a carrier for probiotic and an alternative to consuming functional foods.

Fungos Micorrízicos Arbusculares em Euterpe edulis Martius (Palmeira Juçara) no Município de Rio Pomba/MG

This study aimed to evaluate the occurrence of arbuscular mycorrhizal association in Euterpe edulis (Palm Jucara) in the region of Rio Pomba, MG. Initially, soil was collected from the rhizosphere of E. edulis in five distinct areas. Following, there was extraction of spores, and the process of discoloration and staining of roots was established. Observation also showed presence of a characteristic structure of arbuscular mycorrhizal fungi in the roots to be analyzed, and presence of spores of mycorrhizal fungi in all areas. These results indicate the possibility for more comprehensive studies aiming at the efficient selection of fungal strains, and possible forms of this palm in nursery inoculation.

Qualidade do leite cru dos tanques de expansão individuais e coletivos de um laticínio do município de Rio Pomba, MG - um estudo de caso

This study aimed to evaluate the microbiological and physico-chemical quality, and verify the occurrence of microbial growth inhibitors in raw milk maintained in expansion tanks of a dairy industry located in Rio Pomba, Minas Gerais, Brazil.. It was collected 36 samples of raw milk from 12 expansion tanks in three replicates and determined the count of mesophilic aerobic, psychrotrophic and proteolytic psychrotrophic microorganisms. It was also determined the somatic cell count, the occurrence of pus and inhibitors of microbial growth in milk samples. Moreover, it was determined physico-chemical composition of milk. It was found that 80.0% of samples were in disagreement with standards established by Normative Instruction n.o51, once mesophilic microorganisms and somatic cells counts were above 7,5 x 10 5 , in addition to the presence of pus and microbial growth inhibitors substances. Also, the count of psychrotrophic and proteolytic psychrotrophic microorganisms were high, which compromises production of dairy products such as cheese, UHT milk, among others. However, we found no irregularities in levels of protein, fat, lactose, and total solids. Therefore, the implementation of good agricultural practices during handling, storage and transportation of raw milk to the dairy industry is necessary in order to minimize financial losses during production and to obtain better quality and safety of dairy products.

QUALIDADE DO LEITE PRODUZIDO NO MUNICÍPIO DE RIO POMBA, MG, COM BASE EM ASPECTOS REGULATÓRIOS

Considerando os diversos problemas tecnologicos e economicos que os microrganismos psicrotroficos ocasionam a industria de laticinios, o objetivo deste trabalho foi avaliar a qualidade microbiologica do leite cru granelizado da regiao de Rio Pomba, MG, bem como as condicoes de manutencao e armazenamento deste produto. Um questionario estruturado foi aplicado a fim de promover o levantamento dos dados dos tanques de expansao comunitarios do Municipio de Rio Pomba, Minas Gerais, e amostras de 200 mL de leite cru refrigerado foram coletadas e levadas ao Laboratorio de Microbiologia para determinacao da populacao de microrganismos mesofilos, psicrotroficos e psicrotroficos proteoliticos. Imprudencias foram verificadas durante o processo de recepcao e armazenamento do leite cru refrigerado e durante a higienizacao dos tanques de granelizacao que possibilitam o acesso de microrganismos ao leite. Desta forma, 54% e 59% das amostras, respectivamente, coletadas no periodo de dezembro de 2008 a fevereiro de 2009 e de abril a maio de 2009, nao atenderam a legislacao vigente em relacao ao numero de microrganismos aerobios mesofilos. Constatou-se que 30% das amostras de leite cru coletadas no periodo de abril a maio de 2009 apresentaram acima de 10 4 UFC.mL -1 de microrganismos psicrotroficos proteoliticos. Portanto, os valores de contagem microbiana encontradas neste trabalho sugerem a possibilidade de atividade deterioradora com consequente perda de qualidade do leite cru refrigerado.

Mycorrhizal association in Euterpe edulis Martius (Palmeira Juçara) in the municipality of Rio Pomba, MG

Este trabalho teve como objetivo avaliar a ocorrencia de associacao micorrizica arbuscular em Euterpe edulis (Palmeira Jucara), na regiao de Rio Pomba/MG. Inicialmente, coletou-se solo da rizosfera de E. edulis em cinco areas distintas. Em seguida, realizouse a extracao de esporos e se estabeleceu o processo de descoloracao e coloracao das raizes. Constatou-se a presenca de estruturas tipicas de associacao micorrizica arbuscular nas raizes em estudo e presenca, em todas as areas, de esporos de fungos micorrizicos arbusculares (FMAs). Estes resultados abrem a possibilidade para estudos mais abrangentes visando a selecao de estirpes fungicas eficientes e possiveis formas de inoculacao desta palmeira em viveiro.

Doce em massa misto de jabuticaba e maçã adicionado de albedo de melancia

Este trabalho objetivou elaborar, caracterizar e avaliar a qualidade do doce em massa misto de jabuticaba e maca adicionado de albedo de melancia. Para isso, o doce foi elaborado e avaliado em ate 90 dias de armazenamento, quanto ao pH, solidos soluveis, umidade, acidez titulavel, fibra bruta, acido ascorbico, cinzas, cor, fungos filamentosos e leveduras e aceitacao sensorial. As analises fisico-quimicas e microbiologicas apresentaram congruencia com a literatura e a legislacao ao comparar os diferentes tempos de armazenamento. Na analise sensorial, verificou-se que o doce nao apresentou diferencas estatisticas nos tempos 0 dia e 90 dias de armazenamento, relativas aos atributos cor, sabor, aroma, aparencia, consistencia e impressao global. Porem, houve diferenca significativa no atributo intencao de compra, no qual o doce armazenado no tempo 0 dia apresentou maior escore medio (4,26). O doce foi seguro para consumo humano e apresentou boa aceitacao pelos provadores, sendo tecnologicamente viavel sua producao.

In vitro and in vivo resistance of Lactobacillus rhamnosus GG carried by a mixed pineapple (Ananas comosus L. Merril) and jussara (Euterpe edulis Martius) juice to the gastrointestinal tract

This study evaluated the viability of Lactobacillus rhamnosus GG (LGG) and its in vitro and in vivo resistance to the gastrointestinal tract (GIT) when carried by a mixed fermented pineapple and jussara juice. The effects of product ingestion on the biochemical characteristics of the blood and on the development of aberrant crypt foci (ACF) in Wistar rats were also determined. The LGG viability in probiotic juice was higher than 7.2 log CFU/mL throughout 28 days at 8 °C. The mean count of the probiotic microorganism in the fecal samples of the rats was 5.6 log CFU/g, identical to the count at the end of the in vitro trial (enteric phase II), indicating that the mixed pineapple and jussara juice was an excellent vehicle for LGG. No difference (p > .05) was verified to ACF among the groups. However, the results for the probiotic culture viability and its in vitro and in vivo resistance to the gastrointestinal tract suggest that this juice is an excellent carrier matrix for LGG and contributed to a reduction in the levels of the LDL (low density lipoprotein) fraction of the blood cholesterol, thus being an aid in the control of coronary diseases.
