Anti-inflammatory activity on mice of extract of Ganoderma lucidum grown on rice via modulation of MAPK and NF-κB pathways

Ganoderma lucidum is a popular medicinal mushroom with anti-inflammatory potential. In the present study, the aim was to determine the anti-inflammatory effect and mode of action of G. lucidum grown on germinated brown rice (GLBR) in a mouse model of colitis. It was shown that GLBR suppressed the production of nitric oxide (NO) and prostaglandin E2 (PGE2) in lipopolysaccharide (LPS)-stimulated macrophages and decreased the expression of COX-2, TNF-α, iNOS, IL-1β, IL-6, and IL-10 mRNAs. GLBR also inhibited activation of p38, ERK, JNK, MAPKs, and nuclear factor kappa-B (NF-κB). In a mouse model of colitis, colonic mucosal injury was evaluated using macroscopic, biochemical, and histopathological testing. Disease activity index (DAI), macroscopic score, and histological score significantly decreased upon GLBR treatment. Moreover, immunofluorescence studies indicated that DSS activates nuclear translocation of NF-κB in colon tissue, which is attenuated by GLBR extract. These findings suggest that GLBR is protective against colitis via inhibition of MAPK phosphorylation and NF-κB activation.

Preventive and therapeutic effects of blueberry (Vaccinium corymbosum) extract against DSS-induced ulcerative colitis by regulation of antioxidant and inflammatory mediators.

Inflammatory bowel disease (IBD) is an inflammatory disorder caused by hyperactivation of effector immune cells that produce high levels of proinflammatory cytokines. The aims of our study were to determine whether orally administered blueberry extract (BE) could attenuate or prevent the development of experimental colitis in mice and to elucidate the mechanism of action. Female Balb/C mice (n=7) were randomized into groups differing in treatment conditions (prevention and treatment) and dose of BE (50 mg/kg body weight). Acute ulcerative colitis was induced by oral administration of 3% dextran sodium sulfate for 7 days in drinking water. Colonic mucosal injury was assessed by clinical, macroscopic, biochemical and histopathological examinations. BE significantly decreased disease activity index and improved the macroscopic and histological score of colons when compared to the colitis group (P<.05). BE markedly attenuated myeloperoxidase accumulation (colitis group 54.97±2.78 nmol/mg, treatment group 30.78±1.33 nmol/mg) and malondialdehyde in colon and prostaglandin E2 level in serum while increasing the levels of superoxide dismutase and catalase (colitis group 11.94±1.16 U/ml, BE treatment group 16.49±0.39 U/ml) compared with the colitis group (P<.05). mRNA levels of the cyclooxygenase (COX)-2, interferon-γ, interleukin (IL)-1β and inducible nitric oxide synthase cytokines were determined by reverse transcriptase polymerase chain reaction. Immunohistochemical analysis showed that BE attenuates the expression of COX-2 and IL-1β in colonic tissue. Moreover, BE reduced the nuclear translocation of nuclear transcription factor kappa B (NF-κB) by immunofluorescence analysis. Thus, the anti-inflammatory effect of BE at colorectal sites is a result of a number of mechanisms: antioxidation, down-regulation of the expression of inflammatory mediators and inhibition of the nuclear translocation of NF-κB.

In vitro antioxidant and anti–inflammatory activities of Korean blueberry (Vaccinium corymbosum L.) extracts

ABSTRACT Objective To investigate in vitro antioxidant and anti-inflammatory activities of Korean blueberry ( Vaccinium corymbosum L.). Methods Total phenolic and flavonoid contents of the Korean blueberry water and ethanol extracts were determined before determining the potential of the extracts as antioxidant. Antioxidant activity of the extracts was determined by following some well established methods for free radical scavenging such as 2,2-diphenyl-picrylhydrazyl hydrate, 1,2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonicacid), free radical induced DNA damage, superoxide dismutase-like and catalase assay etc . Furthermore, 1-(4,5-dimethylthiazol-2-yl)-3,5-diphenylformazan and nitric oxide assay were performed to determine the anti-inflammatory activity of the extracts. Results Total phenolic contents were found (115.0±3.0) and (4.2±3.0) mg GAE/100 g fresh mass for both extracts, respectively and flavonoid contents were (1 942.8±7.0) and (1 292.1±6.0) mg CE/100g fresh mass for water and ethonal extracts, respectively. Both the extracts displayed significant scavenging activity of some radicals such as 2,2-diphenyl-picrylhydrazyl hydrate (IC 50 at 1.8 mg/mL and 2.05 mg/mL, respectively), 1,2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonicacid) (IC 50 at 1.5 mg/mL and 1.6 mg/mL, respectively) and nitrite (IC 50 at 1.7 mg/mL and 1.5 mg/mL, respectively) etc . The extracts were found to prevent inflammation as well by reducing nitric oxide production and cytotoxicity in cell. Conclusions The findings suggest that the fresh Korean blueberry could be used as a source of natural antioxidants and anti-inflammatory agents.

Apigenin Attenuates Melanoma Cell Migration by Inducing Anoikis through Integrin and Focal Adhesion Kinase Inhibition

Apigenin, a nonmutagenic flavonoid, has been found to have antitumor properties and is therefore particularly relevant for the development of chemotherapeutic agents for cancers. In this study, time- and dose-dependent cell viability and cytotoxicity were assessed to determine the effects of apigenin on A2058 and A375 melanoma cells. Melanoma cells were pretreated with different concentrations of apigenin and analyzed for morphological changes, anoikis induction, cell migration, and levels of proteins associated with apoptosis. Apigenin reduced integrin protein levels and inhibited the phosphorylation of focal adhesion kinase (FAK) and extracellular signal-regulated kinase (ERK1/2), which induce anoikis in human cutaneous melanoma cells. Apigenin exhibited dose-dependent inhibition of melanoma cell migration, unlike untreated controls. Furthermore, apigenin treatment increased apoptotic factors such as caspase-3 and cleaved poly(ADP-ribose) polymerase in a dose-dependent manner, demonstrating the metastasis of melanoma cells. Our results provide a new insight into the mechanisms by which apigenin prevents melanoma metastasis by sensitizing anoikis induced by the loss of integrin proteins in the FAK/ERK1/2 signaling pathway. These findings elucidate the related mechanisms and suggest the potential of apigenin in developing clinical treatment strategies against malignant melanoma.

Evaluation of the Antioxidant Activity and Anti-Inflammatory Effect of Hericium erinaceus Water Extracts

This study was conducted to determine the antioxidative activity of Hericium erinaceus (HE) and we investi- gated that HE extract contributes to cell proliferation and to anti-LPS-induced inflammation. HE extract showed high DPPH free radical scavenging activity. However, the reducing power activity slightly increased in compare with control. Nitrite scavenging activity, ABTS radical scavenging activity, SOD-like activity of HE were elevated in dose-dependent. The level of total phenolic and flavonoid showed 30.06 ㎎/100 g and 33.86 ㎎/100 g, respectively. Linoleic acid oxidation inhibi- tion had reached a maximum level on the fourth day and started to drop from the fifth day. HE extract contributes to cell proliferation on the RAW 264.7 cell. Our finding demonstrated that water extracts of HE possess significant antioxidant activities and may be suggested a new potential source of anti-inflammatory medicines.

Chaga mushroom (Inonotus obliquus) grown on germinated brown rice suppresses inflammation associated with colitis in mice

The aim of this study was to evaluate the antiinflammatory activity of ethanol extracts from chaga mushroom (Inonotus obliquus, IOE) grown on germinated brown rice. A total of 35 male BALB/c mice were divided into 5 treatment groups and given a commercial diet (A), IOE administration (B), dextran sodium sulfate (DSS) treatment to induce colitis (C), IOE administration+DSSinduced colitis (D), and sulfasalazine administration+ DSS-induced colitis (E). IOE treatment (D) decreased the expression of tumor necrosis factor (TNF)-α, cyclooxygenase (COX)-2, interleukin (IL)-4, interferon (IFN)-γ, signal transducers and activators of transcription (STAT)1, and STAT6 and showed lower levels of immunoglobulin (Ig)E and IgA in the spleen and mesenteric lymph node (MLN) compared to those of the DSS-induced colitis group (C). In addition, IOE suppressed the DSS-induced colonic tissue destruction. Therefore, our data strongly suggests that IOE could be a potent anti-inflammatory agent.

DNA Protection and Antioxidant and Anti-inflammatory Activities of Water Extract and Fermented Hydrolysate of Abalone (Haliotis discus hannai Ino)

Antioxidant and anti-inflammatory activities of water extract and fermented hydrolysate of abalone (Haliotis discus hannai Ino) were examined. Experimental models were used to characterize the antioxidant activities, which were compared with the standard antioxidants α-tocoferol and EDTA (ethylenediamine tetraacetic acid). Antioxidant activities of water extract and fermented hydrolysate were examined at the cellular level and evaluated for anti-inflammatory activities in LPS (lipopolysaccharides)-stimulated murine RAW 264.7 macrophages. Production of the pro-inflammatory mediators nitric oxide (NO) and prostaglandin (PGE2) was significantly (p<0.05) inhibited by treatments with abalone water extract and fermented hydrolysate, compared with controls. Abalone (Haliotis discus hannai Ino) can be considered as a potential source of natural antioxidants and dietary supplements.

Anti-inflammatory, counteractive, and preventive effects of orally administrated oat extracts on dextran sulfate sodium-induced colitis in mice

The anti-inflammatory effects of oat extracts in vitro and in vivo were determined based on antioxidant functions. To examine the effect of oat extracts in murine macrophage RAW 264.7 cells, MTT assays were used and nitric oxide (NO) production was analyzed based on absorbance and inducible nitric oxide synthase (iNOS) expression levels using Western blot analysis. Immunoglobulin expression levels were determined using an enzyme-linked immunosorbent assay (ELISA). Oat extracts showed significant (p<0.05) antioxidant properties and restored the cell viability of LPS-stimulated RAW 264.7 cells, compared with controls. NO production and iNOS protein levels were also reduced by oat extracts. DSS (3%)-administered mice manifested symptoms similar to human colitis patients, and oat extracts attenuated the symptoms of colitis. Preventive and therapeutic administration of oat extracts used as antioxidants can ameliorate inflammatory conditions in cells and DSS-induced colitis in mice.

Evaluation of Synurus deltoides for antioxidant, antimicrobial, and anti-proliferative activities using In vitro assays

The antioxidant, antimicrobial, and anti-proliferative activities of water and ethanol extracts from leaves of Synurus deltoides (SD) were investigated. Extracts showed strong reductive powers, nitrite-scavenging activities, DPPH, ABTS, hydroxyl, and superoxide radical scavenging activities, and DNA damage prevention activities. SD extracts also showed strong cellular antioxidant activities in a dose-dependent manner with IC50 values of 0.93 and 0.65 mg/mL for water and ethanol extracts, respectively. Antimicrobial activities were evaluated against 4 microorganisms. SD extracts showed high inhibitory activities against Pseudomonas aeruginosa and Staphylococcus aureus. The anti-proliferative activity was dose-dependent against human colon adenocarcinoma (HT-29) and mouse rectum carcinoma cells (CMT-93). In addition, HPLC analyses of SD leaf extracts revealed the presence of different phenolic compounds. SD can be considered as a source of functional foods and pharmaceuticals.
