Presence of Toxoplasma gondii and Neospora caninum DNA in the brain of wild birds

Toxoplasma gondii infections are prevalent in many avian species and can cause mortality in some bird hosts. Although T. gondii has been isolated from various species of birds, the role of many different species of wild birds in the epidemiology of T. gondii remains unknown. Neospora caninum, a closely related parasite to T. gondii, has been recently confirmed to infect domestic chickens and wild birds such as house sparrows (Passer domesticus). The present study reports the presence of T. gondii and N. caninum DNA by PCR in brain tissues of 14 species of wild birds from Spain. From a total of 200 samples analyzed, 12 samples (6%) were positive for T. gondii [5 Eurasian jays (Garrulus glandarius), 5 magpies (Pica pica), 1 black kite (Milvus migrans) and 1 Griffon vulture (Gyps fulvus)], while 3 samples (1.5%) were positive for N. caninum [2 magpies and 1 common buzzard (Buteo buteo)]. This is the first report of detection of T. gondii in magpies, griffon vulture and black kite and of N. caninum in common buzzard and magpies, extending the list of natural intermediate hosts for T. gondii and N. caninum infections to these species.

Prevalence of antibodies against Toxoplasma gondii in roe deer from Spain.

Roe deer (Capreolus capreolus) is an important game animal in Spain. Sera from 278 roe deer from eight areas in mainland Spain were assayed for antibodies to Toxoplasma gondii by modified agglutination test (MAT). Titers of 1:25 or higher were found in 109 (39.2%) of 278 deer. No significant differences in antibody prevalence were found between sex or age categories. In contrast, significant differences in seroprevalence between locations were evident. Roe deer from the Northern coastal habitats (high humidity and roe deer density) had the highest prevalence, compared with low prevalence in Central Spain (arid areas and low roe deer density). There was a positive correlation between antibody prevalence and mean annual rainfall (r(s)=0.85, n=8, P<0.01). These findings have environmental and/or public health implications because venison can be an important meat source of T. gondii infections for humans and feral cats.

Seroprevalence and risk factors associated with Toxoplasma gondii in domestic pigs from Spain.

Serum samples from 2970 (1400 sows, 1570 fattening) pigs, from 100 farms in the 10 main swine production regions in Spain were tested for antibodies against T. gondii by the modified agglutination test (MAT). Antibodies to T. gondii (MAT 1:25 or higher) were detected in 492 pigs (16.6%, 9.7% in fattening pigs and 24.2% in sows). The herd prevalence was 85.0% (95% CI: 78-92) and within-farm prevalence ranged from 2.9% to 92.8% (median=17.6%). Statistically significant differences were observed among sampling regions with seroprevalence significantly higher in pigs from Valencia Community (27.3%), Extremadura (23.3%) and Catalonia (21.2%). A generalized estimating equations model indicated that the risk factors associated with T. gondii seroprevalence were: age, sows compared to fattening pigs (OR=2.9; 95% CI=1.83-4.53), lack of rodent control (OR=1.9; 95% CI=1.04-3.60) and presence of cats (OR=1.6; 95% CI=1.12-2.34). The seroprevalence observed in the present study indicates a widespread, although variable, exposure to T. gondii among domestic pigs in Spain, which might have important implications for public health. Management measures including control of rodents and cats on the farms could help to reduce the observed prevalence levels in Spain.

Border Disease Virus among Chamois, Spain

Approximately 3,000 Pyrenean chamois (Rupicapra pyrenaica pyrenaica) died in northeastern Spain during 2005-2007. Border disease virus infection was identified by reverse transcription-PCR and sequencing analysis. These results implicate this virus as the primary cause of death, similar to findings in the previous epizootic in 2001.

Serosurvey of Dogs for Human, Livestock, and Wildlife Pathogens, Uganda

To the Editor: Domestic dogs live in close association with humans and livestock, participating in the transmission of diseases of zoonotic, veterinary, and conservation interest (1,2). Most households in Uganda have traditionally kept dogs for hunting and for help with herding, security, and guarding livestock. Most dogs receive no prophylactic measures (e.g., vaccinations) and roam freely; this situation exposes them to pathogens from eating garbage, rodents, and stillborn animals and other carcasses and through inhalation during scent communication. Thus, dogs are a reservoir for certain pathogens and a useful sentinel for others (3). In 2011, serum samples were obtained from 116 mixed-breed dogs during a rabies vaccination campaign in and near 3 national parks in southwestern Uganda; the dogs were >4 months of age and were voluntarily brought in by their owners (Figure, Appendix, Table). Two of the parks, Bwindi Impenetrable (BI) and Mgahinga Gorilla (MG), have some of the most biologically diverse tropical forests in eastern Africa and are home to mountain gorillas. The third park, Queen Elizabeth (QE), is home to populations of protected carnivores and ungulates. The parks lie within a densely populated rural landscape; in some areas, the population is as high as 500 persons/km2. Table Methodology and seroprevalence for selected pathogens in rural dogs in 3 national parks, Uganda, 2011* Of the 116 sampled dogs, 4 had been vaccinated against rabies by the authors in 2010 in QE (not included in rabies results), and 11 (all males) had been castrated by local animal healers before serum samples were obtained. The samples were used to test for seroprevalence rates to rabies virus (RABV), canine distemper virus (CDV), canine parvovirus (CPV), Leptospira interrogans, Leishmania sp., Toxoplasma gondii, and Neospora caninum (Table). Seroprevalence rates ranged from 20% to 100% (Table). CPV seroprevalence was higher in BI and QE than in MG (χ2 >12.6, p<0.001); T. gondii seroprevalence was higher in BI than in MG (Fisher p = 0.002); and RABV seroprevalence was higher in castrated than noncastrated dogs (50% vs. 10%; Fisher p = 0.005). For humans, the domestic dog is the main source of exposure to RABV. The possibility that the presence of the rabies titers in the dog serum samples was due to a previous vaccination can be ruled out because the only previous recent campaign in the area was conducted by the authors. Antibodies against RABV in apparently healthy dogs have been reported in Africa (6), and rabies seems to be not invariably fatal in dogs. Dogs that have recovered from a rabies infection are prone to shed RABV in their saliva for long periods (7). Antibodies against RABV were more frequently found in castrated dogs. This finding may be due to an increase in virus-related deaths among noncastrated dogs; such dogs tend to be more aggressive and to roam, so they may come more frequently into contact with pathogenic RABV strains. Results indicate that both CDV and CPV are actively circulating in the studied dog populations. High CDV seroprevalence rates have been reported among other rural dog populations in Africa (8). Sick, debilitated pups are at high risk for predation by wild carnivores, so spillover may take place. A dog population exhibiting similar characteristics to the population we studied was believed to be the origin of the 1994 CDV epidemic among Serengeti wildlife (8). Furthermore, carnivores use feces for scent communication, so the probability of infection by CPV in wild carnivores in the study area may also be high. In developing countries, leptospirosis is emerging as a major public health problem and also causes enormous economic losses because of disease in livestock (9). The most commonly detected serovars in this study were those that have rats and dogs as reservoirs (Table​(Table).). Visceral leishmaniasis in humans is also a major health problem in several areas of eastern Africa, where the number of cases has dramatically increased during the past 20 years. Transmission of Leishmania donovani in eastern Africa may take place through anthroponotic or zoonotic cycles, although, to our knowledge, no reservoir host had been identified (10). The mean T. gondii seroprevalence detected during this survey appears to be the highest reported for dogs worldwide. This protozoon has implications for human and animal health, and dogs, who probably become infected with T. gondii when eating raw meat, are a good sentinel for environmental contamination by this parasite. On the other hand, dogs serve as the definitive host for N. caninum, which is a major cause of abortions in cattle and causes economic losses wherever it is enzootic. Some of these diseases may also have implications for the conservation of endangered mountain gorillas. Diseases such as leptospirosis, toxoplasmosis, and especially, rabies could be fatal for gorillas, and there are unpublished reports of fights between hunting dogs and gorillas. Our work should serve as a first step toward the establishment of preventive strategies for improvements in the health of humans and domestic animals living in rural Uganda and for the health of the country’s unique wildlife. Tracing the role of dogs in the cycle of the studied pathogens is crucial for the design of control programs.

Seroprevalence of Toxoplasma gondii in equids from Southern Spain.

Antibodies to Toxoplasma gondii were determined in serum samples from 616 equids (454 horses, 80 mules and 82 donkeys) in a cross-sectional study of 420 herds in Andalusia (Southern Spain), the region with the highest number of equids in Spain. Antibodies to T. gondii were found in 10.8% horses, 15.0% mules and 25.6% donkeys by using the modified agglutination test (MAT) at a cut-off of 1:25. Herd seroprevalence for horses, mules and donkeys was 14.7% (48/327), 23.9% (11/46) and 34.0% (16/47), respectively, and 75 herds (17.8%) had at least one seropositive animal. Significant differences in T. gondii seroprevalence were observed among species, with donkeys having the highest seroprevalence and horses the lowest (P=0.04). Seroprevalence was significantly higher in herds with presence of domestic ruminants. This study is the first report of the presence of T. gondii antibodies in equine species in Spain and the first reporting T. gondii infection in donkeys in Europe. The presence of antibodies is indication of contact with the parasite and therefore, consumption of equine meat could be a potential source of human infection in Spain.

Seroprevalence of Toxoplasma gondii and Neospora caninum in feral cats (Felis silvestris catus) in Majorca, Balearic Islands, Spain

Felids are important in the epidemiology of Toxoplasma gondii infection because they are the only hosts that can excrete environmentally resistant oocysts. Antibodies to T. gondii and Neospora caninum were determined in serum samples from 59 feral cats (Felis silvestris catus) captured in baited traps during authorized predator control campaigns in Majorca, Balearic Islands, Spain. Antibodies to T. gondii assayed by the modified agglutination test (MAT) were found in 50 (84.7%) of 59 cats with MAT titers of 1:25 in one cat, 1:200 in four cats, 1:500 in seven cats, 1:1000 in nine cats, and >or=1:2000 in 29 cats. Seroprevalence (MAT, 1:25 or more) was significantly higher in adults (94.6%) than in juveniles (<6 months old; 40.0%). Seroprevalence to N. caninum, assayed by cELISA (VMRD) and confirmed by an indirect fluorescent antibody test, was low (6.8%, 4 of 59). The prevalence of T. gondii observed in wild cats in Majorca is one of the highest reported worldwide in this species and the highest observed in Europe to date. The results suggest that feral cats in Majorca have a high rate of T. gondii infection with important implications for public health on the island since the seropositive cats are likely to have already shed T. gondii oocysts in the environment.

Toxoplasma gondii in Ruminant Species (Cattle, Sheep, and Goats) from Southern Spain

Abstract: Seroprevalences of Toxoplasma gondii antibodies were assessed with the use of a commercial indirect ELISA in 1,501 domestic ruminants in southern Spain. Antibodies against T. gondii were detected in 420 (83.3%) of 504 cattle, 248 (49.3%) of 503 sheep, and 124 (25.1%) of 494 goats. The herd seroprevalence was 100% (72/72), 84.7% (61/72), and 72.2% (52/72) for cattle, sheep, and goats, respectively. Seropositivity was significantly higher in herds with a low density of animals (P < 0.001). Significant differences (P < 0.05) among municipalities were also found. The seroprevalence observed in the present study indicates a widespread exposure to T. gondii in livestock in southern Spain.

Seropositivity and Risk Factors Associated with Toxoplasma gondii Infection in Wild Birds from Spain

Toxoplasma gondii is a zoonotic intracellular protozoan parasite of worldwide distribution that infects many species of warm-blooded animals, including birds. To date, there is scant information about the seropositivity of T. gondii and the risk factors associated with T. gondii infection in wild bird populations. In the present study, T. gondii infection was evaluated on sera obtained from 1079 wild birds belonging to 56 species (including Falconiformes (n = 610), Strigiformes (n = 260), Ciconiiformes (n = 156), Gruiformes (n = 21), and other orders (n = 32), from different areas of Spain. Antibodies to T. gondii (modified agglutination test, MAT titer ≥1∶25) were found in 282 (26.1%, IC95%:23.5–28.7) of the 1079 birds. This study constitute the first extensive survey in wild birds species in Spain and reports for the first time T. gondii antibodies in the griffon vulture (Gyps fulvus), short-toed snake-eagle (Circaetus gallicus), Bonellis eagle (Aquila fasciata), golden eagle (Aquila chrysaetos), bearded vulture (Gypaetus barbatus), osprey (Pandion haliaetus), Montagus harrier (Circus pygargus), Western marsh-harrier (Circus aeruginosus), peregrine falcon (Falco peregrinus), long-eared owl (Asio otus), common scops owl (Otus scops), Eurasian spoonbill (Platalea leucorodia), white stork (Ciconia ciconia), grey heron (Ardea cinerea), common moorhen (Gallinula chloropus); in the International Union for Conservation of Nature (IUCN) “vulnerable” Spanish imperial eagle (Aquila adalberti), lesser kestrel (Falco naumanni) and great bustard (Otis tarda); and in the IUCN “near threatened” red kite (Milvus milvus). The highest seropositivity by species was observed in the Eurasian eagle owl (Bubo bubo) (68.1%, 98 of 144). The main risk factors associated with T. gondii seropositivity in wild birds were age and diet, with the highest exposure in older animals and in carnivorous wild birds. The results showed that T. gondii infection is widespread and can be at a high level in many wild birds in Spain, most likely related to their feeding behaviour.

Seroprevalence and risk factors associated with Toxoplasma gondii infection in pig farms from Catalonia, north-eastern Spain

Seroprevalence and associated risk factors for Toxoplasma gondii infection in pigs were analyzed in 1202 sera samples, including sows and pigs of three, seven, 11, 15 and 20 weeks of age, from 23 farms in Catalonia, north-eastern Spain. Antibodies were tested by the modified agglutination test (MAT) at titers > or =1:25. Antibodies to T. gondii were found in 228 samples (19.0%; 95% CI: 16.8-21.2). The individual prevalence in animals higher than 7 weeks of age was 22.8% (174/762; 95% CI: 16.6-29.0) and the within-farm prevalence ranged from 7.1% to 36.4%. Statistically significant differences were found among age classes. The risk factors significantly associated with T. gondii seroprevalence were the presence of cats, percentage of mortality at weaning and the presence of outdoor facilities in the farms. The seroprevalence observed in the present study indicates widespread exposure to T. gondii among domestic pigs in Catalonia, which may have important implications for public health.
