Development of an impedimetric aflatoxin M1 biosensor based on a DNA probe and gold nanoparticles.

The present work describes the construction and application of a new DNA biosensor for detection of aflatoxin M1. In order to immobilize a thiol-modified single stranded DNA (ss-HSDNA) probe that specifically bound aflatoxin M1, a self-assembled monolayer of cysteamine and gold nanoparticles on the SAM were prepared on gold electrodes, layer-by-layer. The assembly processes of cysteamine, gold nanoparticles, and ss-HSDNA were monitored with the help of electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV) techniques. K3[Fe(CN)6]/K4[Fe(CN)6] solution was used as a redox probe for electrochemical measurements. The biosensor provided a linear response to aflatoxin M1 over the concentration range of 1-14 ng/mL with a standard deviation of ±0.36 ng/mL. Finally, the biosensor was applied to a series of real milk samples.

Immobilization of anti-aflatoxin B1 antibody by UV polymerization of aniline and aflatoxin B1 detection via electrochemical impedance spectroscopy

Abstract In the study, we investigated the practicality of the UV polymerization of aniline for anti-aflatoxin B1 antibody immobilization, and utilization of the resulting biosensor in the impedimetric determination of aflatoxin B1. The anti-aflatoxin B 1 antibody was physically immobilized on gold electrodes by UV polymerization of aniline at a fixed wavelength. The biosensor was based on specific interaction anti-aflatoxin B1 – aflatoxin B1 recognition and investigation of this recognition event by electrochemical impedance spectroscopy. A calibration curve was obtained in a linear detection range 1–20 ng/mL aflatoxin B1. Finally, the biosensor was applied to analysis of a real food sample.

Enrichment of probiotic ice cream with different dietary fibers: Structural characteristics and culture viability

This study evaluated the effect of 5 dietary fibers (apple, orange, oat, bamboo, and wheat) on the physicochemical, rheological, and textural characteristics; sensory properties; and culture viability of probiotic ice cream stored at -18°C for 180 d. The presence of orange and apple fibers increased the titratable acidity, decreased the lightness (color) value of the ice creams, and enhanced the red and yellow coloration. Compared with the control sample, the consistency indices and apparent viscosities of the experimental samples increased with the addition of all dietary fibers except oat fiber. The highest viscosity was obtained in the sample fortified with apple fiber, whereas the ice cream containing orange fiber showed the highest hardness after d 60 of storage. The addition of orange and apple fibers significantly increased melting resistance; however, panelists did not generally like these samples in terms of taste-flavor. All ice creams had viable counts of Lactobacillus acidophilus of ≥7 log cfu/g during storage except the samples with orange and bamboo fiber. Bifidobacterium lactis counts were also found to be >6 log cfu/g in those samples until d 150 of storage.

Farklı Süt Çeşitleri ile Üretilen İzmir Tulum Peynirlerinin Bazı Fizikokimyasal Özellikleri ve Aroma Bileşikleri

Bu calismada geleneksel bir turk peynir cesidi olan Izmir Tulum Peyniri’nin fiziksel vexa0 kimyasal gibi bazi karakteristik ozellikleri ile aroma maddeleri calisilmistir. 21 adet inek sutunden ve 16 adet ise inek, koyun ve keci sutu karisimindan uretilen olmak uzere toplam 37 peynir ornegi analiz edilmistir. Yalnizca inek sutu kullanilarak uretilen 21 adet pH, titrasyon asitligi, kuru madde icerigi, yag ve kurumaddede yag, tuz ve kurumaddede tuz, toplam azot, protein, suda cozunen azot, trikloroasetik asitte cozunen azot, proteoz-pepton azotu degerleri sirasiyla 4.11-4.92, %0.522-1.408, %49.920-68.648, %17.50-30.50, %35.06-51.98, %2.340-4.563, %3.75-5.26, %3.29-4.29, %20.99-27.37, %0.47-1.21, %0.32-0.91, %0.12-0.46 arasinda degisiklik gostermistir. Buna ek olarak, inek, koyun ve keci sutu karisimlarindan uretilen toplam 16 peynir orneginde bu degerler sirasiya 4.01-4.75, %0.86-1.447, %54.587-71.854, %24.00-32.00, %33.59-52.71, %1.872-5.265, %3.02-9.65, %3.37-4.31, %21.50-27.50, %0.56-1.29, %0.34-1.03, %0.12-0.60 ve 14.51-33.78 arasinda degisiklik gostermistir. Izmir Tulum Peynirlerinin aroma maddesi profilleri kati faz mikroekstraksiyon teknigi ile gaz kromotografisinde belirlenmistir. Gaz kromotografisinden elde edilen veriler ucucu profillerine dayali temel bilesenler analizi ile analiz edilmistir. Tulum peyniri orneklerinde serbest yag asidi, ester, aldehit ve ketonlar temel aroma fraksiyonlari olarak one cikmis ve ayni zamanda aroma maddesi kompozisyon ozelliklerinin sut cesidi ve uretim yontemlerine gore etkilendigini gostermistir.

Calismanin amaci farkli modifiye atmosfer kosullarinda paketlenen geleneksel Sepet peynirlerinin fiziksel, kimyasal ve duyusal ozelliklerini incelemektir.xa0xa0xa0xa0 xa0+4 °C’ de depolanan Sepet peynirlerinin tum ozellikleri depolamanin 1., 45., 90. xa0xa0xa0xa0xa0xa0xa0xa0ve 180. gunlerinde incelenmistir. Peynirlerin paketlenmesinde 3 farkli modifiye atmosfer kosulu denenmistir. Uretilen peynirlerxa0 % 100 N 2 (N), % 80xa0 N 2 + % 20xa0 xa0xa0CO 2 (NC), % 100 CO 2 (C) iceren atmosfer kosullarda polistiren ambalajlarda paketlenmistir. Istatistiksel analiz sonucunda modifiye atmosfer kosullarinda (MAP) paketlenen Sepet peyniri orneklerinin kurumadde, kurumadde de yag % ve kurumadde de tuz % iceriklerinde meydana gelen degisimlerin onemli oldugu tespit edilmistir. Kitle, tekstur, gorunus, lezzet ve aromayi iceren duyusal ozellikler istatistiksel olarak incelendiginde, sadece CO 2 iceren atmosferik kosullarda paketlenen peynirlerde depolama suresince farklilik oldugu gozlenmistir. Genel olarak bakildiginda % 100xa0 N 2 (N), % 80xa0 N 2 + % 20xa0 CO 2 (NC) gazlarini iceren farkli modifiye kosullarda paketlenmis olan Sepet peynirlerinin fizikokimyasal ve duyusal ozelliklerini daha iyi muhafaza ettigi gorulmustur.

Yeşil ve Siyah Çay İlavesinin İçilebilir Yoğurdun Viskozitesi ve Duyusal Özellikleri Üzerine Etkisi

Siyah ve yesil cayin icilebilir yogurdun viskozitesi ve duyusal ozellikleri uzerine etkisi arastirilmistir. Calismada iki cay infuzyon konsantrasyonu (%2 ve %4) kullanilirken herhangi bir ilave yapilmayan icilebilir yogurt kontrol ornegi olarak yer almistir. En yuksek viskozite degerleri kontrol orneginde tespit edilirken cay ekstrakti ilave edilen icilebilir yogurtlar arasinda 14.gun disinda onemli bir fark bulunamamistir. %4 oraninda cay ilave edilmesi %2 orani ile kiyaslandiginda tat puanlarini olumsuz yonde etkilemistir. %2 yesil cay ile muamele edilen icilebilir yogurt kontrol ornegine benzer renk ve tekstur degerleri gostermistir. %2 siyah veya yasil cay ilave edilen orneklerde toplam kabul edilebilirlik acisindan istatistiksel olarak herhangi bir fark belirlenememistir.
